SummaryThe observed anomalies in high density lipoproteins in Friedreich's ataxia led us to investigate the state of cellular membranes in this illness. As a preliminary screening program, we studied the shape of erythrocytes; the phospho-lipid content of platelets and the transport properties of these membranes as indirectly reflected in the absorption of Vit E and the renal handling of orally injected taurine. All these investigations were normal, except for a tendency towards more echinocytes in Friedreich's ataxia and the significant increase in taurine urinary excretion after an oral load. We concluded that the possible membrane abnormalities are not major and will have to be searched for with more subtle and specific tests.

The concentrations of lipids in the plasma and adrenal cortical tissue were compared in British (Bos taurus) and Zebu (Brahman cross and Africander cross) cattle. Plasma cholesterol and phospho-lipid concentrations were higher (P < 0'001) in Zebu than in British animals. In grazing steers the adrenal cholesterol concentrations were higher (P < 0'01) in Zebu than in British steers but phospholipid concentrations were lower (P < 0'01) in Brahman cross than in Africander cross and British animals. The correlations between the concentrations of cholesterol in plasma and adrenal glands were significant. In Zebu steers on different dietary treatments the concentrations of choles-terol and phospholipid were higher (P < 0'01) and triglyceride lower (P < 001) in the adrenal tissue of steers growing normally than of steers losing weight. The adrenal to body weight ratios were lower (P < 0'01) in Brahman cross than in Africander cross and British grazing animals.

Abstract Aminopropyl solid-phase columns can be used for phospholipid fractionation. A method has been developed to elute the acidic phospholipid phosphatidylglycerol (PG) separately, and it is applicable to both standard (phospho)lipid mixtures and pulmonary surfactants. The simplicity, rapidity, and high recovery make this method of isolating PG superior to other chromatographic procedures.
