: The content and the application of pharmaceutical dosage forms must meet several basic requirements to ensure and maintain efficiency, safety and quality. A large number of active substances have limited ability to direct administration. Excipients are generally used to overcome the limitation of direct administration of these active substances. However, the function, behavior and composition of the excipients need to be well known in the design, development and production of pharmaceutical dosage forms. In this review, excipients used to assist in any pharmaceutical dosage form production processes of drugs, to preserve, promote or increase stability, bioavailability and patient compliance, to assist in product identification / separation, or to enhance overall safety and effectiveness of the drug delivery system during storage or use are explained. Moreover, the use of these excipients in drug delivery systems are identified. Excipient toxicity, which is an issue discussed in the light of current studies, also discussed in this review.

Polystyrene-polyethylene glycol (PS-PEG) tentacle polymers with loadings of up to 60/<μmol/g were used for standard oligonucleotide synthesis. As these resins are easy to handle and stable under reaction and cleavage conditions they may be used alternatively to controlled pore glass (CPG) as the most commonly used solid support for oligonucleotide synthesis. However, structural and chemical properties of the PS-PEG resins require modified conditions to guarantee syntheses with high coupling efficiencies. Oligonucleotides (ODN ) of various sequences and lengths have successfully been synthesized using HPLC and capillary electrophoresis (CE) for purity control. Additionally, electrospray mass spectrometry (ES-MS) was used for product identification.

The sulfurylation of estrone-6,7-3H, estradiol-17β-6,7-3H, and dehydroisoandrosterone-4-14C by laying hen liver, oviduct, and vaginal preparations was investigated. Purification and product identification included ether and ethyl acetate extraction, paper chromatography, and isotope dilution, before and after hydrolysis with a sulfatase preparation. The esterifying enzymes were found in the 105 000 × g supernatants of the three tissues. The liver preparation was many times more active in steroid sulfate synthesis than the corresponding oviduct or vaginal fractions. Sulfurylation of dehydroisoandrosterone displayed the same cofactor, pH, and temperature requirements as did that of estrone and estradiol-17β. The degree of dehydroisoandrosterone sulfate synthesis was considerably lower than that of the estrogens, however. It is suggested that the laying hen possesses an enzyme system which is more efficient for the sulfurylation of estrogens than of other steroids such as dehydroisoandrosterone.
