The present investigation was undertaken in Black Night Shade (Solanum nigrum L.) which is an important medicinal plant. Direct multiple shoots proliferation was achieved from shoot tip. The shoot tips were cultured on MS medium fortified with Thidiazuron (TDZ) (1.0-7.0 mg/L) for multiple shoot induction. Multiple shoots proliferation was best observed at 3.0 mg/L TDZ from the shoot tip explants within three weeks of culture. Shoot number per explant ranged between 2 and 10. Individual shoots were aseptically excised and sub cultured in the same media for shoot elongation. The elongated shoots were transferred to Indole Acetic Acid/Indole Butyric Acid (IAA/IBA) (0.5mg/L–2.0mg/L) for root induction. Rooting was observed within two weeks of culture. Rooted plantlets were successfully hardened under culture conditions and subsequently established in the field conditions. The recorded survival rate of the plants was 86%. Plants looked healthy with no visually detectable phenotypic variations.

A rapid, simple and efficient protocol for <i>in vitro</i> multiple shoot induction and plantlet regeneration was achieved from three different explants of <i>Cicer arietinum</i>. The explants viz shoot tip, cotyledonary node and node were cultured on MS medium fortified with Benzyl Adenine (BA) (0.44&minus;8.88&mu;M) for multiple shoot induction. Multiple shoots proliferation was best observed at 4.44&mu;M BA from all the three explants within two weeks of culture. Of the three different explants tested, cotyledonary nodes produced the maximum number of shoots. Shoot number per explant ranged between 7 and 15. Individual shoots were aseptically excised and subcultured in the same media for shoot elongation. The elongated shoots were transferred to Indole Butyric Acid (IBA) (2.46&minus;12.30&mu;M) for root induction. Rooting was observed within two weeks of culture. Rooted plantlets were successfully hardened under culture conditions and subsequently established in the field conditions. The recorded survival rate of the plants was 76.3%. Plants looked healthy with no visually detectable phenotypic variations.

An efficient protocol for plant regeneration through multiple shoots induction from shoot tips of Polygonum hydropiper (L.) was established. The highest percentage (96.6) of multiple shoot induction and number of shoots (9.0) per culture were found on MS supplemented with 2.0 mg/l Kn. The induced shoots were excised and inoculated on to MS contains different concentrations of IBA or NAA for rooting. The highest percentage (90.0) of root induction and the highest number of roots per shoot (12.0) was found on MS having 1.0 mg/l IBA. Context: Orchid produces a huge number of minute seeds but the seeds can not germinate easily in nature due to the lack of endosperm in the seeds is an incompatibility barrier that limits its propagation in nature. Objectives: To develop in vitro culture techniques for quick propagation of Vanda tessellate, a commercially important orchid species. Materials and Methods: Shoot tips were used as experimental materials. The explants were surface sterilized and the shoot tips were excised. The isolated shoot tips were cultured in MS medium supplemented with different concentration and combinations of auxin and cytokinin. Results: The combination of 1.5 mgl-1 NAA and 1.0 mgl-1 BAP was proved to be the best medium formulation for multiple shoot formation as well as maximum shoot elongation. The single shoots were isolated from the multiple shoots and subcultured in MS medium having NAA and IBA individually and in combinations for root induction. Maximum root induction was obtained in MS agarified medium having 0.5 mgl-1NAA and 1.0 mgl-1IBA. The well rooted plantlets were hardened successfully in the potting mixture containing coconut husk, perlite, charcoal, brick pieces in the ratio of 2:1:1:1 and eventually established under natural condition.Conclusion: An efficient regeneration protocol for micropropagation in V. tessellata through shoot tip culture has been established.Key words: Shoot tip; micropropagation; orchid.DOI: 10.3329/jbs.v17i0.7122J. bio-sci. 17: 139-144, 2009

Nepenthes albomarginata Lobb ex Lindl. is a carnivorous plant, distributes in several regions in Indonesia. The plant population decreases drastically because of over exploitation and ruining nature habitat. Plant propagation by nature and cutting are not enough to rehabilitation its population. In vitro culture of N. albomarginata was carried out using plantlets grown from the seeds in vitro. Plantlets were cut to became two part explants, consisted of shoot tip and under-shoot tip cuttings. These cutting explants were grown on Murashige & Skoog (MS) media with addition of plant growth regulators of 6-benzylaminopurine (BA), combined with or without-naphthalene acetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D) at 1 mg/l. Shoot tip cuttings of N. albomarginata formed double multiple shoot 25,00% on control; formed triple multiple shoots 25,00% onBA 1 mg/l treatment; formed callus 37,50%, triple or quartet shoots 25,00% and rooted plantlets 25,00% on BA 1 mg/l + NAA 1 mg/l treatment. The under-shoot tip cuttings ofN. albomarginata formed double – triple shoots 25,00% and rooted plantlets 37,50% on control; formed double – triple shoots 25,00% and rooted plantlets 12,50% on BA 1 mg/ltreatment; formed callus 12,50%, double - pentacle shoots 37,50% and rooted plantlets 25,00% on BA 1 mg/l + NAA 1 mg/l treatment. 2,4-D 1 mg/l or its combined with BA 1mg/l treatment caused deadly shoot tip or under-shoot tip explants. The combination of BA 1 mg/l + NAA 1 mg/l was the best treatment for producing callus, multiple shootsand rooted plantlets of N. albomarginata.

An experiment was carried out to optimize the doses and combinations of growth regulators for in vitro propagation of Dendrobium Red Bull through shoot tip culture. The experiment was laid out according to completely randomized design with 15 treatments and three replications. For multiple shoot induction, newly grown shoot buds (keikies) were cultured on MS supplemented with various combinations of NAA and BAP. The regenerated plants were transferred to ex vitro in different substrates and acclimatized. Best response for the shoot length (21.19 mm) was obtained from the medium supplemented with 3.0 mg/l BAP with 1.5 mg/l NAA. Similarly maximum number of leaves was observed from the same combination at all the recorded days except 60 days, whereas the maximum shoot number (7.66) was obtained from the medium containing 3.0 mg/l BAP + 1.0 mg/l NAA. The shoots were then transferred to root inducing medium and well rooted plants were transferred to different substrates. Survivability of the regenerated plants varied from 60 to 92%. The substrate containing cocodust showed maximum survivability (92%) whereas minimum (60%) was observed from the substrate containing course sand. BAP and NAA proved to be good growth regulators for in vitro multiplication of Dendrobium. Plant Tissue Cult. & Biotech. 28(2): 161-169, 2018 (December)

A simple micropropagation method from nodal and shoot tip explants is reported here for Mentha viridis, an economically important medicinal plant. High frequency of microshoots was obtained from these two explants on MS supplemented with various concentrations of BAP (1.0 - 4.0 mg/l) and Kn (1.0 -4.0 mg/l). Maximum number of shoots was obtained from nodal explants in the medium containing 3.0 mg/l BAP. The root induction was carried out by using IBA and IAA (0.5 - 2.0 mg/l). Among these, high frequency of root proliferation was achieved in the medium containing 1.5 mg/l of IBA. The rooted plantlets were hardened and transferred to the field.  Explants of Solanum nigrum L., collected from Pachamalai hills callused successfully on MS basal medium supplemented with IAA and BAP. The highest frequency of green compact callus and multiple shoots were obtained on MS containing 2.0 mg/l IAA and 0.5 mg/l BAP. The callus when cultured on MS basal medium fortified with different concentrations of BAP (3.0 - 8.0 mg/l) and IAA (0.5 mg/l) showed multiple shoot formation. The highest frequency of multiple shoots was obtained on MS containing 6.0 mg/l BAP and 0.5 mg/l IAA. For in vitro flowering, the node explants were cultured on MS fortified with different concentrations of BAP (2.0 - 7.0 mg/l) and NAA (0.5 mg/l). The highest number of multiple shoots were obtained in MS supplemented with 6.0 mg/l BAP and 0.5 mg/l NAA. The in vitro flowering was observed on MS containing 2,4-D and BAP 1.5 mg/l, respectively. The best rooting was obtained on MS containing 0.5 mg/l IBA. Efficientin Vitro direct multiple shoot regeneration from Solanum pubescens was achieved from leaf explants on MS medium Sublimated with B5 vitamins and different concentrations and different combinations of PGRs like BAP, NAA and GA3. The maximum numbers of multiple shoots were achieved from leaf explants on 3.0 mg/l BAP + 1.0mg/l GA3. The regenerated shoots were transferred in to half strength MS medium fortified with IBA for root induction. Rooted plantlets were successfully acclimatized. This new and transfer into the field Conditions. Standardized and reproducible protocol useful the mass propagation of Solanum pubescens.

The shoot-tip explant harvested from ethyl methane sulphonate (EMS) and gamma ray (GR) mutagenized seedling was cultured over MS medium fortified with NAA and BAP for five generations to amplify the mutated sector. Mutagens reduced the regeneration efficiency of the explant and affected its plant growth regulator-dependence for multiple shoot induction. While the 12d-old shoot-tip from GR-treated seedling induced shoots with 0.5µM NAA+6.6µM BAP; that from EMS-treated seedling induced shoots with 8.8µM BAP. The present study establishes that the mutagens affect the regeneration process in the explant.
