The moisture status of seeds is usually high during the period of harvest and deterioration (loss of viability) starts to occur when seeds are stored for longer periods. In the present study, soybean seeds were evaluated using a standard germination test, in vitro germination, and for efficient multiple shoot induction, following storage under ambient conditions for 0, 3, 6 and 9 months. Results showed that seeds stored for more than 3 months had reduced moisture content and decreased germination percentages in LS677, LS678, Dundee, Peking, TGx1740-2F and TGx1835-10E of the tested genotypes. In particular, seeds stored for 9 months showed significantly poor seed viability and less than 50% overall seed germination (Dundee—42%, LS678—49%, TGx 1740-2F—44%, TGx 1835-10E—48%), except for LS677 and Peking, with 52 and 55%, respectively. The efficiency of multiple shoot induction also decreased with prolonged seed storage, with all genotypes recording an overall decline from about 96% to 40% regeneration efficiency within 9 months. The results obtained clearly indicated that high germination rates and efficient in vitro shoot induction depended largely on seed viability and storage duration, and significantly differed according to genotypes.

High frequencies of multiple shoot regeneration were achieved from shoot tip explants cultured on MS media fortified with 4.0 mgl-1 BAP + 0.1 mgl-1 NAA. On an average 10.2?0.38 shoots per explant were obtained. The regenerated shoots were rooted in MS medium supplemented with 1.0 mgl-1 IBA. The in vitro grown plantlets were acclimatized and successfully transferred to natural condition with 80% survival. An efficient protocol for plant regeneration through multiple shoots induction from shoot tips of Polygonum hydropiper (L.) was established. The highest percentage (96.6) of multiple shoot induction and number of shoots (9.0) per culture were found on MS supplemented with 2.0 mg/l Kn. The induced shoots were excised and inoculated on to MS contains different concentrations of IBA or NAA for rooting. The highest percentage (90.0) of root induction and the highest number of roots per shoot (12.0) was found on MS having 1.0 mg/l IBA. ‘Hom Rangsi’ was the non-photoperiod aromatic mutant rice which derived from fast neutron radiation KDML 105. ‘Hom Rangsi’ seeds were cultured on MS solid medium without any supplemented for a week. And then, all explants were placed on MS (Murashige and Skoog, 1962) medium supplemented with 0, 5, 10, 15, 20, 25 mg/L BA (benzyladenine) for multiple shoot induction. The optimal concentration of BA for induced multiple shoot induction of ‘Hom Rangsi’ line was MS + BA 25 mg/L, the highest number of shoots were 5.38 shoot/seed. The following experiment was done, irradiated ‘Hom Rangsi’ seeds with 0, 100, 200, 300, 400 Gy gamma ray which cultured on MS solid medium supplemented with 400 mg/L Al3+ pH 2.9 were selected for acid tolerance lines. After six weeks cultured, the survivals of irradiated plantlets were 86.32, 77.78, 58.95, 58.95, 21.87% and the height of irradiated plantlets were 8.4, 8.3, 6.7, 6.6, 6.1 cm respectively without any shoot budding. All survival plantlets were transferred to suitable MS + BA 25 mg/L medium which discovered from the first experiment for multiple shoot budding. After six weeks cultured, the maximum of 5.24 shoots/plantlet were found from 300 Gy irradiation significantly and followed by 400, 200, 0 and 100 Gy irradiation treatments which gave 4.55 and 4.41, 4.37 and 4.31 shoots/planlet respectively.

An efficient protocol for the rapid micropropagation of medicinally important Elephantopus scaber has been standardized using cotyledonary node explants. Direct multiple shoot induction was observed when the cotyledonary node explants at various age groups were cultured on MS medium supplemented with various plant growth regulators. The highest shoot induction was obtained when the cotyledonary node explants from 20-day-old seedlings were cultured on MS medium supplemented with 1.5 mg L-1 TDZ and 0.5 mg L-1 NAA. On this medium, 98% of the cultures responded, with an average number of 33.7 shoots per explant. The highest frequency of rooting (100%) and mean number of roots (3.3 per shoot) were observed when the shoots were transferred to MS medium supplemented with 1.0 mg L-1 IBA. The plantlets raised in vitro were acclimatized and transferred to soil with a 92% success rate. The protocol described here may be utilized for multiplication and conservation of elite clones of E. scaber.
