The results of selection studies conducted at the Central Experimental Base of the Federal State Budgetary Scientific Institution Federal Williams Research Center of Forage Production and Agroecology in the Non-Chernozem Zone of the Russian Federation are presented. Brief data on promising samples and hybrids of alfalfa created in the department of alfalfa breeding for seed productivity are presented. For a long time, alfalfa breeding was mainly in the direction of obtaining new varieties with high yields of fodder mass and little attention was paid to seed yields, which led to the creation of good varieties for collecting green mass and medium or even poor seed productivity. Therefore, the creation of varieties with high feed and seed productivity is the primary task of alfalfa breeding. For this purpose, the main methods of selection are used — polycross, interspecific and intervariety hybridization and selection. In the obtained populations, a careful selection for early ripeness, pollen fertility, plant flowering friendliness is carried out, the power of hybrids, their foliage and other useful indicators are taken into account. By the nature of flowering and pollination, alfalfa refers to cross-pollinating plants. For agricultural production, it is necessary to have alfalfa varieties not only with high, but also stable seed productivity, which does not depend on the conditions of the growing environment and the presence of pollinators.

Due to its fast deterioration, soybean (Glycine max L.) has an inherently poor seed vigor. Vigor loss occurring during storage is one of the main obstacles to soybean production in the tropics. To analyze the genetic background of seed vigor, soybean seeds of a recombinant inbred line (RIL) population derived from the cross between Zhonghuang24 (ZH24, low vigor cultivar) and Huaxia3hao (HX3, vigorous cultivar) were utilized to identify the quantitative trait loci (QTLs) underlying the seed vigor under −20 °C conservation and accelerated aging conditions. According to the linkage analysis, multiple seed vigor-related QTLs were identified under both −20 °C and accelerated aging storage. Two major QTLs and eight QTL hotspots localized on chromosomes 3, 6, 9, 11, 15, 16, 17, and 19 were detected that were associated with seed vigor across two storage conditions. The indicators of seed vigor did not correlate well between the two aging treatments, and no common QTLs were detected in RIL populations stored in two conditions. These results indicated that deterioration under accelerated aging conditions was not reflective of natural aging at −20 °C. Additionally, we suggest 15 promising candidate genes that could possibly determine the seed vigor in soybeans, which would help explore the mechanisms responsible for maintaining high seed vigor.

The treatment modality for recurrent cervical cancer (rCC) is limited, and the prognosis of these patients is poor. Seed implantation could be an important component of rCC management in the context of dose boost or salvage therapy after surgery or radiotherapy, which is characterized by a minimally invasive, high local dose, and rapidly does fall, sparing normal tissue. For patients with good performance status and lateral pelvic wall recurrence with an available puncture path, seed implantation was recommended, as well as for selected central pelvic recurrence and extra-pelvic recurrence. The combination of brachytherapy treatment planning system and CT guidance was needed, and three-dimensional printing templates could greatly improve the accuracy, efficiency, and quality of seed implantation to achieve a potential ablative effect and provide an efficient treatment for rCC. However, the recommendations of seed implantation were mainly based on retrospective articles and lack high-quality evidence, and multicenter prospective randomized studies are needed. In this consensus on iodine125 seed implantation for rCC, indication selection, technical process and requirements, dosimetry criteria, radiation protection, combined systemic therapy, and outcomes of seed implantation for rCC are discussed.

Diospyros mespiliformis Hochst. is a multipurpose tree species from tropical dry forests that is still exploited in the wild and whose silvicultural practices are not documented. The aim of this study was to understand the peasants’ perceptions on the socio-economic importance, the level of vulnerability and the cultivation practice of this species in the Sudanese zone of Chad. The methodological approach consisted of surveys carried out among 350 people in 10 villages, as well as field observations. The results showed that D. mespiliformis is well known to local populations (100% of respondents) who use it in six categories of use, with a broad consensus for use in medicine (Cs = 0.96), food (Cs = 0.33), constructions (Cs = 0.26) and as fuelwood (Cs = 0.24). On the other hand, the consensus remained weak for use in crafts (Cs = - 0.76) and as lumber (Cs = - 0.83). According to 80.2 % of respondents, income from the exploitation of this resource significantly contributes to improving household well-being. Apart from picking and collecting fruit, all other harvesting methods cited by respondents (debarking, root picking, leaf stripping and felling) are destructive, but only 17.7 % of respondents are aware of this. A regressive trend in the population dynamics of the species is perceived by the majority (52 %) of respondents. According to 83.5 % of respondents, the cultivation habits of this species are rare or even non-existent, because of poor seed germination, the scarcity of seeds and climatic conditions unfavorable to the success of suckering. Almost all respondents (99 %) declared themselves ready to adopt the practice of growing the plant if the regeneration constraints are lifted. The results of this study clearly indicate that D. mespiliformis would be an interesting species for participatory domestication for the intensification of the agro-forestry system in the Sudanese zone of Chad.

The low phytic acid (lpa) trait in soybeans can be conferred by loss-of-function mutations in genes encoding myo-inositol phosphate synthase and two epistatically interacting genes encoding multidrug-resistance protein ATP-binding cassette (ABC) transporters. However, perturbations in phytic acid biosynthesis are associated with poor seed vigor. Since the benefits of the lpa trait, in terms of end-use quality and sustainability, far outweigh the negatives associated with poor seed performance, a fuller understanding of the molecular basis behind the negatives will assist crop breeders and engineers in producing variates with lpa and better germination rate. The gene regulatory network (GRN) for developing low and normal phytic acid soybean seeds was previously constructed, with genes modulating a variety of processes pertinent to phytic acid metabolism and seed viability being identified. In this study, a comparative time series analysis of low and normal phytic acid soybeans was carried out to investigate the transcriptional regulatory elements governing the transitional dynamics from dry seed to germinated seed. GRNs were reverse engineered from time series transcriptomic data of three distinct genotypic subsets composed of lpa soybean lines and their normal phytic acid sibling lines. Using a robust unsupervised network inference scheme, putative regulatory interactions were inferred for each subset of genotypes. These interactions were further validated by published regulatory interactions found in Arabidopsis thaliana and motif sequence analysis. Results indicate that lpa seeds have increased sensitivity to stress, which could be due to changes in phytic acid levels, disrupted inositol phosphate signaling, disrupted phosphate ion (Pi) homeostasis, and altered myo-inositol metabolism. Putative regulatory interactions were identified for the latter two processes. Changes in abscisic acid (ABA) signaling candidate transcription factors (TFs) putatively regulating genes in this process were identified as well. Analysis of the GRNs reveal altered regulation in processes that may be affecting the germination of lpa soybean seeds. Therefore, this work contributes to the ongoing effort to elucidate molecular mechanisms underlying altered seed viability, germination and field emergence of lpa crops, understanding of which is necessary in order to mitigate these problems.

Abstract Background Direct seeding is an efficient rice cultivation practice. However, its application is often limited due to O2 deficiency following submergence, leading to poor seed germination, seedling establishment, and consequently yield loss. Identification of genes associated with tolerance to submergence and understanding their regulatory mechanisms is the fundamental way to address this problem. Unfortunately, the molecular mechanism of rice response to submergence stress is still not well understood. Results Here, we have performed a genome-wide identification of FCS-like zinc finger (FLZ) proteins and assessed their involvement in submergence response in rice. We identified 29 FLZ genes in rice, and the expression analysis revealed that several genes actively responded to submergence stress. Eight OsFLZ proteins interact with SnRK1A. As a case study, we demonstrated that OsFLZ18 interacted with SnRK1A and inhibited the transcriptional activation activity of SnRK1A in modulating the expression of its target gene αAmy3, a positive regulator in rice flooding tolerance. In line with this, OsFLZ18-overexpression lines displayed retarded early seedling growth and shorter coleoptile following submergence. Conclusions These data provide the most comprehensive information of OsFLZ genes in rice, and highlight their roles in rice submergence response.

Soapnut (Sapinduas emarginatus) is one of the most primitive precious useful plants since ancient times. This plant was domesticated due to its multifarious usefulness. The study was conducted in College of Forestry, Sirsi during 2019-20 to know the influence of post scarification storage treatments on seed germination and quality in Sapinduas emarginatus. The seeds were collected from in and around Sirsi area. In soap nut, the basic problem is poor seed germination due to hard seed coat. The seeds were treated with concentrated H2So4 for 12min and stored under laboratory condition for six months. At monthly interval, seed samples were drawn and evaluated for post scarification germination. The germination behaviour remarkably influenced due to scarification and without scarification during experimentation. The untreated seeds recorded the maximum germination (68%) compared to scarified seeds (22.85%).Speed of germination was highest in seed without scarification treatment (2.7) compared to scarified seeds. Mean daily germination was highest in without scarified seeds (1.04) and lowest in the scarified seeds (0.32). Peak value was maximum in without scarified seeds (0.50) compared to scarified seeds (0.22). Seedling vigour index was significantly highest in without scarified seeds (1290) compared to scarified seeds (455). In general, scarified seeds with conc. H2SO4 for 12 min affected seed germination due high conc. H2SO4 because of chemical residues remaining in seed that affects the embryo. As the advancement in storage period the seed coat may lose the integrity so, germination was improved in without scarified seeds.

Background: Soybean is an important oilseed crop but it has poor seed longevity. Studies on seed longevity are very complex and hence, an indirect selection criterion should be identified. In this study, seed longevity and its association with agronomic traits of seven soybean varieties was evaluated in the year 2019-2020.Methods: Seed were kept for storage under ambient conditions for a period of 16 months (January 2019 to May 2020). Their germination was recorded at bimonthly intervals and seed longevity was inferred as the time period in months up to which the seed maintains its germination percentage above IMSCS.Result: Accordingly, Palam Early Soya 1 and Him Palam Hara Soya were classified as having short seed longevity (12 months). Hara Soya, Shivalik, Him Soya and Palam Soya were classified as having medium seed longevity (14 months). Whereas, PS 1556 was classified as having long seed longevity ( greater than 16 months). Further, it was found that seed longevity was positively associated with number of pods per plant and seed yield, and negatively associated with number of branches per plant and 100-seed weight. This study presents an indirect selection criterion based on some easily accessible parameters which have strong association with seed longevity.

The low reproductive success of Arctic plants is predicted to increase as the climate warms. However, climate extremes add complexity to these predictions. In the extremely cold year of 2018, multiple Arctic trophic levels experienced reproductive failure. We analysed a unique long-term record of seed viability from experimentally warmed and ambient plots at Alexandra Fiord, Ellesmere Island which has been running since 1992 and included 2018 and the extremely warm year of 2019. Positive and negative July temperature anomalies and summer temperatures have increased significantly by 0.5, -0.3 and 0.4°C/decade since 1977, resulting in greater extremes and increased inter-annual variation. Seed viability of some species has increased with climate warming. Across years, seed viability of woody species was consistently higher in warmed than ambient plots while the opposite was true for forbs. In 2018, seed viability of woody species in ambient plots was significantly lower than normal but comparable with past years for forb species. Not all species returned to normal seed viability levels in 2019. Our study highlights the potential for greater sexual reproductive failure in tundra plants with increasing climate extremes. We suggest that poor seed viability of woody species in cold years could constrain shrub recruitment and may aid forb species establishment on bare tundra.

In view of the problems of a poor seed-filling state and poor seed-cleaning effect of a precision metering device under high-speed working conditions, an inside-filling air-assisted high-speed precision maize seed-metering device was designed, fabricated, and evaluated. The overall structure and working principles of the seed-metering device are explained. Theoretical analysis and parametric design of the key parts, such as the horizontal stirring notch seed-metering plate and the wheel seed-cleaning mechanism, were carried out. The main factors that produce multiple seeding problems were studied. The forward speed (FS) and working pressure (WP) of the seed-metering device are determined as the main factors affecting the seeding performance. A quadratic orthogonal rotation combination experiment was designed. The relationship between influencing factors and performance indexes was constructed using the Design-Expert 10.0.7 software. The response surface method was used to optimize the experimental results. The results of the bench validation experiments showed that the seeding performance of the seed-metering device was excellent when the FS of the seed-metering device was 13.10 km/h and the WP of the seed-metering device was 4.75 kPa. The results of the bench comparison showed that the seeding performance of the designed seed-metering device was better than that of the conventional air-suction precision seed-metering device. This study provides theoretical and practical references for the optimized design of a pneumatic precision seed-metering device and the sustainable development of grain.
