Functional state of the organism of the hybrid Russian sturgeon × Siberian sturgeon (Acipenser gueldenstaedtii × Acipenser baerii) after winter in cages in Volga Delta

Two-year-olds (15 individuals) and five-year-olds (15 individuals) of the Russian sturgeon × Siberian sturgeon (Acipenser gueldenstaedtii × Acipenser baerii) hybrid, kept in net cages in the Volga delta, were studied after wintering in order to establish the functional state of the organism according to the complex of indicators of the erythron system and metabolism substances. The results of the performed study found that the erythron system in five-year-olds is characterized by a higher content of erythrocytes and blood hemoglobin (0.58 ± 0.01×10 12 l –1 and 64.98 ± 1.29 g/l, respectively), compared with two-year-olds (0.49 ± 0.02×10 12 l –1 and 64.98 ± 1.29 g/l, respectively).The serum iron content in both groups was the same. A higher content of total protein and albumin in the blood, as well as a greater activity of ALT were found in five-year-olds, compared with two-year-olds. Thus, the content of total protein and albumin in blood serum in five-year-olds was at the level of 48.23 ± 1.54 and 21.58 ± 2.34 g/l, respectively, and in two-year-olds — 32.42 ± 0.92 and 13,84 ± 0.61 g/l, respectively (p < 0.05). The activity of serum ALT in five-year-olds was 1.07 ± 0.06 μkat/l, and in two-year-olds — 0.85 ± 0.03 μkat/l (p < 0.01). Serum cholesterol in five-year-olds (3.94 ± 0.04 mmol/l) and two-year-olds (4.4 ± 0.08 mmol/l) did not differ significantly (p > 0.05). The content of serum triglycerides in five-year-olds (4.50 ± 0.49 mmol/l) and in two-year-olds (5.36 ± 0.45 mmol/l) did not differ significantly (p > 0.05). The study of carbohydrate metabolism revealed a pronounced variability in the glycemic index in the younger age group (coefficient of variation 44.7 %) with an average blood glucose value of 2.69 ± 0.31 mmol/l. The blood glucose content of five-year-olds was higher than that of two-year-olds — 3.84 ± 0.24 mmol/l (p < 0.05). The established parameters of variation and the age specificity of the indicators of the erythron system, protein, lipid and carbohydrate metabolism can be used to assess the state of physiological adaptation of the hybrid after wintering in cages in the Volga delta.

Oxidant and antioxidant activity in seminal plasma was compared with that in spermatozoa of teleost (common carp Cyprinus carpio and brook trout Salvelinus fontinalis) and chondrostean (Russian sturgeon Acipenser gueldenstaedtii, Siberian sturgeon Acipenser baerii, and sterlet Acipenser ruthenus) fishes. No differences were found between seminal plasma and spermatozoa in the level of thiobarbituric-acid-reactive substance (0.24 &plusmn; 0.08 to 0.33 &plusmn; 0.04 nmol/mg proteins) in Russian sturgeon, Siberian sturgeon, and sterlet. Carbonyl protein concentration was significantly higher in spermatozoa than in seminal plasma of all studied species. Analyzed antioxidants included superoxide dismutase, glutathione reductase, and glutathione peroxidase activity. Significant differences (P &lt; 0.05) were detected between seminal plasma and spermatozoa in total superoxide dismutase (SOD) and glutathione reductase (GR). Total glutathione peroxidase (GPx) activity was significantly higher in brook trout (12.56 &plusmn; 3.23 mU/mg proteins) and Russian sturgeon (11.56 &plusmn; 3.12 mU/mg proteins) spermatozoa compared to seminal plasma (6.81 &plusmn; 1.56 mU/mg proteins in brook trout and 9.56 &plusmn; 3.12 mU/mg proteins in Russian sturgeon). This study provides new data on oxidant and antioxidant balance between spermatozoa and seminal plasma that may be of value in the development of methods for artificial reproduction of teleost and chondrostean species. &nbsp;

The aim of this study was to evaluate and compare the haematological parameters of cultured Russian sturgeon (Acipenser gueldenstaedtii), stellate sturgeon (Acipenser stellatus) and Siberian sturgeon (Acipenser baerii) in recirculating aquaculture system. Total erythrocytes, thrombocytes, red blood cell distribution, monocytes, basophils as well as haemoglobin levels of stellate sturgeon differed significantly from that of the other two species (p<0.05). Leukocyte and lymphocyte levels of Siberian sturgeon were significantly higher than in stellate sturgeon and Russian sturgeon (p<0.05). Eosinophil percentages of Siberian sturgeon were significantly lower than stellate sturgeon and Russian sturgeon (p<0.05). Haematocrit levels of stellate sturgeon differed from Siberian sturgeon (p<0.05), but no differences between Siberian and Russian sturgeons were found. Neutrophil, mean cellular haemoglobin and mean cellular volume and mean cellular haemoglobin concentration levels did not differ significantly between the three sturgeon species (p>0.05). The information generated may help in monitoring health status and to adopt better management strategies in culture systems.

In the paper, rearing results and comparative assessment of interspesific hybrids and pure species of the Siberian sturgeon Acipenser baerii (Brandt, 1869)and the Russian sturgeon Acipenser gueldenstaedti (Вrandt, 1833) have been provided at various stages of ontogenesis by combinated cultivation under RAS conditions and in an industrial uniflow tank farm. Investigations were carried out at the “Konakovski” department of the Branch on Freshwater Fisheries FSBSI “VNIRO” (VNIIPRKh). To obtain the experimental material breeders were used of the Siberian sturgeon from the sixth domestication generation and of the Russian sturgeon from the third one. Fish fry, switched to mixed feeding ongrowing and further rearing, were cultivated under production conditions in normal technological mode and separately from each female using different granulated mixed feeds (BioMar firm) of different recipes. Up to 41 day-age fish were reared in RAS, further in race-ways and tanks of the shop with uniflow water-supply. The middle water temperature was 16ºC during the all period of experimental works (557 days). Best results of the mass accumulation showed interspecific LS×RS hybrids, at the second position seemed to be RS×LS hybrids. Pure species of the Siberian and Russian sturgeons showed practically similar middle weights to the end of the experimental stage. As a result of the histological analysis, it has been ascertained that gonads of pure species of one-year-old Siberian and Russian sturgeons and their hybrids had comparatively the same picture of sex cells development, but some advantage possessed the Siberian sturgeon and LS×RS hybrid. Thus, malesʼ cysts and femalesʼ nests of one-year-old fish show multiple mitotic divisions of sex cells (spermatogonia and oogonia) what corresponds with the first stage of gonades maturity in males and the first one and the beginning of the second ones in females.

The study was conducted on 12 healthy male buffalo calves weighing between 50 to 60 Kg to evaluate haemato-biochemical alterations after epidural injection of dexmedetomidine alone and in combination with bupivacaine. Haematological examination showed nonsignificant alterations. Biochemical examination showed significant increase in blood glucose, blood urea nitrogen and serum creatinine while there was nonsignificant alterations in total protein and alkaline phosphatase. All haemato-biochemical changes were transient and compensatory.
