ASSESSMENT OF SERUM SCLEROSTIN LEVEL AS A BIOMARKER ASSOCIATED WITH BONE DISORDERS IN Β-THALASSEMIA PATIENTS IN AL- NAJAF CITY, IRAQ

Aim of the study: To assess serum sclerostin in female patients with beta-thalassemia and compare with the healthy controls and to predict its complication associated with the bone pathophysiology, for designed improvement the lifestyle goodliness for these patients. Material and methods: Sixty-nine female beta-thalassemia (βT) patients (54 βT major and 15 βT Intermedia), aged 8-40 years who dependent on transfused blood, and 20 healthy controls were evaluated serum sclerostin, and was examined the relationship with hematological parameters RBC, Hb, PCV, WBC, PLT, BMI, splenic status, iron, and ferritin levels. The information of beta-thalassemia patients was collected and recorded by the questioner. Results: A significantly increased serum sclerostin level (mean 26.80±0.91) pg/ml was shown in βT patients compared with the healthy controls (10.03±0.68, p < 0.001) pg/ml. Furthermore, a significant decrease (p<0.05) of the sclerostin level was observed in β-thalassemia major compared to intermedia β-thalassemia patients. Serum sclerostin level revealed a significant increase in progress age; it is highest in the age group (30-40) year as compared with age group (8-18) and (19-29) year respectively. Sclerostin showed no associations with the RBC, Hb, PCV, and significantly positively correlated (p<0.05) with serum iron, ferritin levels, WBC, and PLT count. Significantly higher sclerostin levels in splenectomized and underweight groups were observed compared to unsplenectomized and normal-weight groups (p<0.05) of βT patients. Conclusion: Sclerostin plays an important role in beta-thalassemia patients and can serve as a biomarker associated with the bone pathophysiology and indicator to prevent the continuation of such serious diseases caused by iron overload in these patients.

Effects of Se nanoparticles supplementation on growth performance, hematological parameters and nutrient digestibility of Labeo rohita fingerling fed sunflower meal based diet

The aim of the present study is to assess the effects of selenium nanoparticles on the growth, hematology and nutrients digestibility of Labeorohita fingerlings. Fingerlings were fed with seven isocaloric sunflower meal-based diet supplemented with different concentrations of nanoparticles naming T1 to T7 (0, 0.5, 1, 1.5, 2, 2.5, and 3 mg/kg), with 5% wet body weight while chromic oxide was used as an indigestible marker. After experimentation for 90 days T3 treated group (1mg/kg -1Se-nano level) showed the best result in hematological parameters (WBC’s 7.97 ×103mm-3, RBC’s 2.98 ×106 mm-3 and Platelet count 67), nutrient digestibility (crude protein: 74%, ether extract: 76%, gross energy: 70%) and growth performance (weight gain 13.24 g, weight gain% 198, feed conversion ratio 1.5, survival rate 100%) as compared to the other treatment groups. Specific growth rates were found significantly higher in T5 than in other groups. The present study indicated positive effect of 1 mg/kg Se-nanoparticles on growth advancement, hematological parameters, and nutrients digestibility of L. rohita fingerlings.

Impact of dietary leaf extracts of Black pepper Piper nigrum L. on the growth, hematological and immunological parameters of Labeo rohita (Hamilton, 1822) cultured in glass aquaria

A study was conducted to evaluate the effect of Piper nigrum (black pepper) leaf extract on on the growth performance, proximate composition, hematological parameters, and immune response of Labeo rohita fingerlings with an average weight of 22.14 ± 0.98g. Aftrer acclimation for two weeks, fish (n=25) were randomly selected and placed in four glass aquaria (T0, T1, T2 and T3) at constant water temperature (30.0 ± 1.0 °C), pH (7.50 ± 0.5) and total hardness (200 ± 2.0 mgL,-1) for a period of 12 weeks, with three replicates each. Fish were fed with P. nigrum leaf extract supplemented feed @ 0.0%, 1.0%, 2.0% and 3.0% in T0, T1,T2 and T3, respectively. At the end of experiment, five fish were randomly selected from each aquaria for proximate composition, gut and skin microbial load, hematological parameters. Total proteins, albumins, and globulins were also recorded to evaluate immunological memory. The result revealed that fish in T2 showed better growth performance with an average weight gain of 56.11 ± 0.51 g. Thus, it had been concluded that Piper nigrum, a medicinal plant, can also be used to enhance the growth performance and immune response of Labeo rohita as attractive alternatives against antibiotics and vaccines and has shown no negative side effects on fish health as well as on its environment.

Chromium supplemented carbohydrate diets for Cirrhinus mrigala: effects on body composition, gut enzyme activity and hematological parameters

A ninety days nutritional trial was directed to explore the effects of dietary chromium on body composition, gut enzyme activity and physiological status of Cirrhinus mrigala by using G & NG corn. Six experimental diets were prepared by using different levels of chromium chloride hexahydrate (0, 0.2, 0.4 mg/kg, each with G & NG corn). For this experimental trial, 480 fingerlings, irrespective of sex were distributed in six aquariums each with replicate. Results revealed that gelatinized corn along with increasing level of Cr2Cl3.6H2O have a positive impact upon body composition of fish. Hematology was positively correlated with chromium chloride hexahydrate supplementation in gelatinized corn. Amylase gut enzyme also showed significant (P<0.05) increase in group fed with chromium chloride hexahydrate supplemented diet (G corn). However, corn with chromium chloride hexahydrate supplementation did not revealed any significant impact on gut protease enzyme activity. From these results it can be concluded that both chromium chloride hexahydrate and gelatinized corn in fish feed are very beneficial to improve body composition, enzymes activity and physiological health status of fish.

Use of multi-strain probiotics in linseed meal based diet for Labeo rohita fingerlings

Unavailability of probiotics in fish digestive system fingerlings is unable to digest and absorb their food properly. The current research was conducted to investigate the influence of probiotics added Linseed meal based (LMB) diet on hematology and carcass composition of Labeo rohita juveniles. Hematological parameters are essential diagnostics used to estimate the health status of fish. The usage of probiotics for fish health improvement is becoming common due to the higher demand for environment-friendly culture system in water. Linseed meal was used as a test ingredient to prepare six experimental test diets by adding probiotics (0, 1, 2, 3, 4 and 5 g/kg) and 1% indigestible chromic oxide for seventy days. According to their live wet weight, five percent feed was given to fingerlings twice a day. Fish blood and carcass samples (Whole body) were taken for hematological and carcass analysis at the end of the experiment. The highest carcass composition (crude protein; 18.72%, crude fat; 8.80% and gross energy; 2.31 kcal/g) was observed in fish fed with test diet II supplemented with probiotics (2 g/kg). Moreover, maximum RBCs number (2.62× 106mm-3), WBCs (7.84×103mm-3), PCV (24.61), platelets (63.85) and hemoglobin (7.87) had also been reported in the fish fingerlings fed on 2 g/kg of probiotics supplemented diet. Results indicated that probiotics supplementation has a critical role in improvement of fingerlings’ body composition and hematological indices. Present findings showed that probiotics supplementation at 2 g/kg level in linseed by-product-based diet was very useful for enhancing the overall performance of L. rohita fingerlings.

Effect of replacing maize with plantain peels on the performance traits and haematological characteristics of African giant land snail

The study was carried out at the Niger Delta University, Faculty of Agriculture Teaching and Research Farm, Bayelsa State. A total of Ninety six (96) growing snails (Archachatina marginata). Using Plantain peel as an additive, four diets were formulated at the levels of 0%, 3%, 6%, and 9% to form the treatment diets. The experiment lasted for eight (8) weeks during which the weight gain, feed intake, shell width, shell length, shell thickness and hematological parameters were obtained. The weight gain, shell width, shell length and shell thickness are not significantly different (p<0.05) whereas, the feed intake differed significantly (p<0.05) for all the treatment diets in the experiment. From the results, it was concluded that concentrate with 0% inclusion of Plantain peel meal was suitable for snail diets as it gave the best performance. The hematological parameters measured in the African giant land snails are White blood cell, Neutrophile and Lymphocyte differed significantly (p<0.05). Plantain peel can serve as source of potassium for other animals, as the highest percentage of potassium was found in the hemolymph of snails fed with 9% inclusion of plantain peel. It is recommended that; there should be no inclusion of plantain peels in the diet of the African giant land snails in order to achieve higher weight gain and feed intake.

Positive Associations between Body Mass Index and Hematological Parameters, Including RBCs, WBCs, and Platelet Counts, in Korean Children and Adolescents

We conducted this study to investigate the associations between hematological parameters and obesity in children and adolescents. The levels of hematological parameters (including white blood cells [WBCs], red blood cells [RBCs], hemoglobin [Hb], hematocrit [Hct], and platelets) of 7997 participants (4259 boys and 3738 girls) aged 10–18 years were recorded. The parameters were compared among participants with normal weight, overweight, and obesity. Significantly higher mean levels of WBCs (7.16 vs. 6.16 × 103/mm3, p < 0.001), RBCs (4.90 vs. 4.82 × 106/mm3, p < 0.001), Hb (14.07 vs. 13.99 g/dL, p < 0.05), Hct (42.31 vs. 41.91%, p < 0.001), and platelets (311.87 vs. 282.66 × 103/mm3, p < 0.001) were found in the obese than normal weight group, respectively, after adjusting for body mass index (BMI) and sex. BMI SDS had significant positive associations with the levels of WBCs (β = 0.275, p < 0.001), RBCs (β = 0.028, p < 0.001), Hb (β = 0.034, p < 0.001), Hct (β = 0.152, p < 0.001), and platelets (β = 8.372, p < 0.001) after adjusting for age, sex, and socioeconomic factors in a multiple linear regression analysis. A higher BMI was associated with elevated WBC, RBC, Hb, Hct, and platelet counts in children and adolescents. Because higher levels of hematological parameters are potential risk factors for obesity-related diseases, hematological parameters should be evaluated in obese children and adolescents.

Impact of The Ageing on Oxidant/Antioxidant Balance And Hematology of Indigenous Cow and Its Amelioration by Ascorbic Acid

Ageing is a complex biological process of all flora and fauna. It leads to a gradual reduction in the ability to maintain homeostasis under internal physiological and external environmental stresses, hence reducing the viability of individual's and increasing their vulnerability to diseases. The objective of this study was to investigate the alteration in the oxidant /antioxidant balance and hematology in four different age group of indigenous Tharparkar cattle and its amelioration by ascorbic acid. A total 24 female Tharparkar cattle were included and were categorized into 4 groups of six cows in each group. Animals below 1 year-age were kept in group I, between 1 to 8 year-age in group II, between 8-10 year-age in group III and above 10 year-age in group IV. The oxidant/antioxidant markers (LPO, SOD, GSH, GPX and catalase activity) and haematological panels (Hb, TEC, TLC, Platelet and DLC) were determined on day 0, before ascorbic acid supplementation, and on day 6, 12, 18 and 24 post ascorbic acid supplementation. In the present study LPO and SOD levels were significantly (P < 0.05) increased with the ageing. GSH and GPX activities significantly (P < 0.05) decreased with ageing in different age groups. Variations in hematological parameters were also observed with ageing in different age groups. The geriatric cattle (> 10 years) exhibited remarkable alteration in oxidative stress indices, and haematological panels when compared to other groups. Supplementation of ascorbic acid reduced the oxidative stress and improved hemoglobin, PCV, TEC and TLC levels. From the findings of this study, it can be inferred that administration of ascorbic acid is helpful in ameliorating altered oxidant/antioxidant balance and hematological parameters with ageing in various age groups of cattle.

Analysis of CircRNAs and CircRNA-Associated Competing Endogenous RNA Networks in β-Thalassemia

The involvement of circRNAs in β-thalassemia and their actions on fetal hemoglobin (HbF) is unclear. Here, the circRNAs in β-thalassemia carriers with high HbF levels were comprehensively analyzed in comparison with healthy individuals. Differential expression of 2183 circRNAs was observed and their correlations with hematological parameters were investigated. Down-regulated has-circRNA-100466 had a strong negative correlation with HbF and HbA2. Bioinformatics was employed to construct a has-circRNA-100466‑associated competing endogenous RNA (ceRNA) network with the determination of hub genes and associated miRNAs. In combination with previous reports, the has-circRNA-100466▁miR-19b-3p▁SOX6 pathway was identified. The ceRNA network was verified by qRT-PCR on β-thalassemia samples and RNA immunoprecipitation of K562 cell lysates. Has-circRNA-100466, miR-19b-3p, and SOX6 were present together in anti-argonaute 2 immunoprecipitates, indicating involvement with HbF induction. Furthermore, spearman correlation coefficients revealed their significant correlations with HbF. In conclusion, a novel has-circRNA-100466▁miR-19b-3p▁SOX6 pathway was identified, providing insight into HbF induction and suggesting targets β-thalassemia treatment.

Dietary supplements based on modified and enriched with amino acids zeolite in the cultivation of young turkeys

Relevance. Modern turkey crosses have a genetically determined high growth rate. These farm birds are very sensitive to even minor fluctuations in their diet, are susceptible to a deficiency of amino acids, protein, mineral elements and vitamins, which can contribute to a decrease in their productivity, the development of pathological processes and diseases. The use of modern technologies and scientific achievements has made it possible to prepare a new product — modified zeolite. This natural mineral is already free of impurities and microbes, with open pore windows, has a high ability to selectively exchange, and is obtained by various methods of activating zeolite in industrial conditions. At the same time, Ulyanovsk specialists have mastered the technology of enriching modified zeolite with useful substances.Methods. The aim of the work was to study the effect on the body of turkeys and the level of their productivity of additives based on modified zeolite enriched with amino acids. The experiment was organized for 60 days on 250 turkeys of the breed Hybrid Cread-mayker aged 55–60 days in the conditions of the Ulyanovsk region. The control group received the main ration (MR), the experimental group was given once a day in addition to the MR in the morning feeding in a mixture with mixed feed 5% additives of modified zeolite enriched with amino acids.Results. The inclusion of a supplement based on modified zeolite enriched with amino acids in the diet of turkeys does not have a negative effect on the hematological parameters of their blood, all indicators corresponded to the physiological norm. During the experiment in the 2nd group, the live weight of poultry was obtained larger by 8.70%, which amounted to 12,296 kg, respectively, more profit was obtained from the sale of turkey meat, total revenue was 133,210, and additional profit was 27,960 rubles, 2.41 rubles of profit was received for 1 ruble of costs. Тhe data indicate the normalization of mineral homeostasis in the body of turkeys and better absorption of nutrients in the feed diet.
