Role of Seed Mycoflora on Seed Germination of Oroxylum indicum (L.) Vent. in Kumaun region of Indian Central Himalaya

A number of fungus was isolated from the seeds of Oroxylum indicum (L.) Vent. The mycoflora associated with the seeds of this tree have not been previously reported from Kumaun region of Indian Central Himalaya. During the course of study mycoflora such as Fusarium solani, Aspergillus niger, Aspergillus nidulens, Penicillium sp., Trichoderma harzianum, Alternaria solani, Alternaria alternata, Curvolaria lunata., Stachybotryis chartarum, Acremonium sp., Rhizoctonia solani, Chaetomium globosum, Cladosporium cladoporoides and Torula allii were isolated from the seeds.

El chile es la segunda hortaliza de mayor producción en México. El objetivo de la investigación fue evaluar la actividad antagónica <em>in vitro</em> e <em>in vivo</em> de <em>Trichoderma harzianum</em> (T-H4) y <em>T. koningiopsis</em> (T-K11) <em>versus Fusarium solani</em> (MX-MIC 798) en la germinación y establecimiento de plántula de chile Miahuateco. Se utilizó la técnica de cultivo dual para determinar el porcentaje de inhibición de crecimiento radial (PICR) de la cepa MX-MIC 798. Además, se analizó el porcentaje de germinación en semillas de chile Miahuateco en vivero, así como la mortalidad de plántulas y grado de severidad a los 40 días después del trasplante (ddt) en Santa María Tecomavaca, Oaxaca, a través de biocontrol y control químico (Mancozeb 80®). La cepa T-H4 presentó el nivel antagónico PICR más alto (53.3%) <em>in vitro</em> y clase II en la escala de Bell, asimismo obtuvo 82% de germinación en semillas de chile Miahuateco en vivero y 48% de mortalidad en campo; de manera que igualó al control químico y superó a <em>T. koningiopsis</em> T-K11. La actividad antifúngica de <em>Trichoderma</em> spp., ofrecen una alternativa para el biocontrol de la marchitez y necrosis en raíz del cultivo de chile Miahuateco causada por <em>F. solani</em> MX-MIC 798.

Abstract. Soesanto L, Hartono ARR, Mugiastuti E, Widarta H. 2020. Seed-borne pathogenic fungi on some soybean varieties. Biodiversitas 21: 4010-4015. The present study was conducted to detect and identify seed-borne pathogenic fungi in some soybean varieties and their effect on seed germination. Experiment was performed in a completely random design with eight treatments and four replicates. Eight soybean varieties i.e., Malabar, Kaba, Dering, Detam I, Sinabung, Dena, Gepak Kuning, and Slamet were selected for the investigation. Seed borne fungi were isolated using blotter test and agar plate techniques. The variables observed were morphological/cultural characteristics, microscopic features of fungi, and percentage of seed germination. A total of eight fungi namely Aspergillus flavus Link, Aspergillus niger van Tieghem, Cladosporium oxysporum Berk. & M.A. Curtis, Colletotrichum dematium (Pers. et Fr.) Grove f.sp. truncate (Schw.) Arx, Curvularia pallescens Boedijn, Fusarium solani (Mart.) Sacc., Melanospora zamiae Corda, and Nigrospora sp. Mason were isolated from different varieties of soybean seeds. The highest seed germination was found to be 80.5% in Daring and Detam I varieties, respectively, and the lowest 53% was recorded in Dena variety.

Onion production in Burkina Faso faces to several constraints including diseases. Symptoms such as seedling damping-off are currently been observed. Most of the fungi responsible for onion diseases are seed-transmitted but in Burkina Faso, little information on onion seed quality were available. In a previous study, Aspergillus niger, Fusarium oxysporum and Fusarium solani were found to be the main fungi present on onion seeds but their responsibility in the development of seedlings damping-off has not been established. The overall aim of this study is to evaluate in vitro, the effect of these fungi on onion seed germination and coleoptile growth in order to establish this responsibility. Onion seeds were inoculated with mycelium suspension of the fungi concentrated at 13.3 g/ml and incubated for 6 days under appropriates conditions. All the tested fungal isolates had an inhibitory effect on onion seed germination at rates ranging from 31 to 60%. The Fusarium species were the most virulent. Coleoptile length was also reduced by the fungi, leading to significant growth time-lag at rates ranging from 34 to 100%. Aspergillus niger was the most virulent. In conclusion, Aspergillus niger, Fusarium oxysporum and Fusarium solani are responsible of seedling damping-off of onion in Burkina Faso.

SUMMARYAll four seed diffusates used for treatment of sunflower seeds, Azadirachtaindica, Capsicum annuum, Coriandrum sativum and Eugenia jambulana, reduced the populations of seed-borne fungi: Alternaria alternata, Drechslera tetramera, Emericellopsis terricola, Fusarium moniliforme, F.semitectum, Macrophomina phaseolina and Phoma oleracea. Of four seed diffusates, those from A.indica and C.sativum controlled the fungal populations almost 100%. Seed germination was increased in seed samples of both sunflower cultivars under study, HO-1 and NK-212. The obtained results indicate that seed diffusates could substitute costly chemicals for safe control of seed-borne diseases, protecting at the same time the environment from chemical pollution.
