Clinical populations have memory deficits linked to sleep oscillations that can potentially be treated with sleep medications. Eszopiclone and zolpidem (two non-benzodiazepine hypnotics) both enhance sleep spindles. Zolpidem improved sleep-dependent memory consolidation in humans, but eszopiclone did not. These divergent results may reflect that the two drugs have different effects on hippocampal ripple oscillations, which correspond to the reactivation of neuronal ensembles that represent previous waking activity and contribute to memory consolidation. We used extracellular recordings in the CA1 region of rats and systemic dosing of eszopiclone and zolpidem to test the hypothesis that these two drugs differentially affect hippocampal ripples and spike activity. We report evidence that eszopiclone makes ripples sparser, while zolpidem increases ripple density. In addition, eszopiclone led to a drastic decrease in spike firing, both in putative pyramidal cells and interneurons, while zolpidem did not substantially alter spiking. These results provide an explanation of the different effects of eszopiclone and zolpidem on memory in human studies and suggest that sleep medications can be used to regulate hippocampal ripple oscillations, which are causally linked to sleep-dependent memory consolidation.

Gamma rhythms in many brain regions, including the primary visual cortex (V1), are thought to play a role in information processing. Here, we report a surprising finding of 3 narrowband gamma rhythms in V1 that processed distinct spatial frequency (SF) signals and had different neural origins. The low gamma (LG; 25 to 40 Hz) rhythm was generated at the V1 superficial layer and preferred a higher SF compared with spike activity, whereas both the medium gamma (MG; 40 to 65 Hz), generated at the cortical level, and the high gamma HG; (65 to 85 Hz), originated precortically, preferred lower SF information. Furthermore, compared with the rates of spike activity, the powers of the 3 gammas had better performance in discriminating the edge and surface of simple objects. These findings suggest that gamma rhythms reflect the neural dynamics of neural circuitries that process different SF information in the visual system, which may be crucial for multiplexing SF information and synchronizing different features of an object.

The SPADE (spatio-temporal Spike PAttern Detection and Evaluation) method was developed to find reoccurring spatio-temporal patterns in neuronal spike activity (parallel spike trains). However, depending on the number of spike trains and the length of recording, this method can exhibit long runtimes. Based on a realistic benchmark data set, we identified that the combination of pattern mining (using the FP-Growth algorithm) and the result filtering account for 85–90% of the method's total runtime. Therefore, in this paper, we propose a customized FP-Growth implementation tailored to the requirements of SPADE, which significantly accelerates pattern mining and result filtering. Our version allows for parallel and distributed execution, and due to the improvements made, an execution on heterogeneous and low-power embedded devices is now also possible. The implementation has been evaluated using a traditional workstation based on an Intel Broadwell Xeon E5-1650 v4 as a baseline. Furthermore, the heterogeneous microserver platform RECS|Box has been used for evaluating the implementation on two HiSilicon Hi1616 (Kunpeng 916), an Intel Coffee Lake-ER Xeon E-2276ME, an Intel Broadwell Xeon D-D1577, and three NVIDIA Tegra devices (Jetson AGX Xavier, Jetson Xavier NX, and Jetson TX2). Depending on the platform, our implementation is between 27 and 200 times faster than the original implementation. At the same time, the energy consumption was reduced by up to two orders of magnitude.

Although the cerebellum has been traditionally considered to be exclusively involved in motor control, recent anatomical and clinical studies show that it also has a role in reward processing. However, the way in which the movement related and the reward related neural activity interact at the level of the cerebellar cortex and contribute towards learning is still unclear. Here, we studied the simple spike activity of Purkinje cells in the mid-lateral cerebellum when monkeys learned to associate a right or left hand movement with one of two visual symbolic cues. These cells had distinctly different discharge patterns between an overtrained symbol-hand association and a novel symbol hand association, responding in association with the movement of both hands, although the kinematics of the movement did not change between the two conditions. The activity change was not related to the pattern of the visual symbols, the movement kinematics, the monkeys' reaction times or the novelty of the visual symbols. The simple spike activity changed with throughout the learning process, but the concurrent complex spikes did not instruct that change. Although these neurons also have reward related activity, the reward-related and movement related signals were independent. We suggest that this mixed selectivity may facilitate the flexible learning of difficult reinforcement learning problems.
