AbstractThe purpose of this paper is to propose a new inertial self-adaptive algorithm for generalized split system of common fixed point problems of finite family of averaged mappings in the framework of Hilbert spaces. The weak convergence theorem of the proposed method is given and its theoretical application for solving several generalized problems is presented. The behavior and efficiency of the proposed algorithm is illustrated by some numerical tests.

The use of unnatural fluorogenic molecules widely expands the pallet of available genetically encoded fluorescent imaging tools through the design of fluorogen activating proteins (FAPs). While there is already a handful of such probes available, each of them went through laborious cycles of in vitro screening and selection. Computational modeling approaches are evolving incredibly fast right now and are demonstrating great results in many applications, including de novo protein design. It suggests that the easier task of fine-tuning the fluorogen-binding properties of an already functional protein in silico should be readily achievable. To test this hypothesis, we used Rosetta for computational ligand docking followed by protein binding pocket redesign to further improve the previously described FAP DiB1 that is capable of binding to a BODIPY-like dye M739. Despite an inaccurate initial docking of the chromophore, the incorporated mutations nevertheless improved multiple photophysical parameters as well as the overall performance of the tag. The designed protein, DiB-RM, shows higher brightness, localization precision, and apparent photostability in protein-PAINT super-resolution imaging compared to its parental variant DiB1. Moreover, DiB-RM can be cleaved to obtain an efficient split system with enhanced performance compared to a parental DiB-split system. The possible reasons for the inaccurate ligand binding pose prediction and its consequence on the outcome of the design experiment are further discussed.

We report on the experimental implementation of the transgenic phiC31 recombinase C. elegans intron-split system. The three-component plasmid-based phiC31 recombinase system consists of i) two intron-split segments of the C. briggsae-unc-119 gene, and ii) the plasmid that provides phiC31 recombinase activity. Described results constitute the proof-of-concept assay for the implementation of bacteriophage phiC31 integrase in C. elegans.

The coordination control of a transmission system has gradually attracted more attention with the development of hybrid electric vehicles. However, nonlinear coupling of multiple power sources, superposition of different dynamic characteristics in multiple components, and withdrawal and intervention for a power-split powertrain with a two-speed automated manual transmission (AMT) gearbox can cause jerk and vibration of the transmission system during the shift, which has higher requirements and challenges for the overall performance improvement of the system. This paper designs a novel, robust, augmented H∞ shift control strategy for a power-split system with a two-speed AMT gearbox of a heavy commercial vehicle and verifies the strategy’s effectiveness with simulations and experiments. First, the dynamic plant model and kinetic equations are established, and the shift is divided into five stages to clearly reveal the jerk and vibration problem. Based on augmented theory, a robust H∞ shift control strategy is proposed. Shift coordination is transformed into a speed tracking problem, and state variable and disturbance are reconstructed to obtain a new augmented system. Simulation and hardware-in-the-loop test are carried out to verify the effectiveness of the strategy, which mainly includes simulation of pneumatic actuator and H∞ control strategy. Results show that the proposed H∞ control strategy can greatly reduce the jerk of the transmission system. The jerk produced by the proposed strategy is decreased from 20.4 to 4.07 m/s3, leading to a substantial improvement of 80%. Therefore, the proposed strategy may offer a theoretical reference for the actual vehicle controller during the shift.

The flexibility and versatility of self-complementing split fluorescent proteins (FPs) have enabled a wide range of applications. In particular, the FP1-10/11 split system contains a small fragment that facilitates efficient generation of endogenous-tagged cell lines and animals as well as signal amplification using tandem FP11 tags. To improve the FP1-10/11 toolbox we previously developed, here we used a combination of directed evolution and rational design approaches, resulting in two mNeonGreen (mNG)-based split FPs (mNG3A1-10/11 and mNG3K1-10/11) and one mClover-based split FP (CloGFP1-10/11). mNG3A1-10/11 and mNG3K1-10/11 not only enhanced the complementation efficiency at low expression levels, but also allowed us to demonstrate signal amplification using tandem mNG211 fragments in mammalian cells.
