The enormous increase in world population has resulted in generation of million tons of agricultural wastes. Biotechnological process for production of green chemicals, namely, enzymes, provides the best utilization of these otherwise unutilized wastes. The present study elaborates concomitant production of protease and amylase in solid state fermentation (SSF) by a newly isolated Bacillus megaterium B69, using agroindustrial wastes. Two-level statistical model employing Plackett-Burman and response surface methodology was designed for optimization of various physicochemical conditions affecting the production of two enzymes concomitantly. The studies revealed that the new strain concomitantly produced 1242 U/g of protease and 1666.6 U/g of amylase by best utilizing mustard oilseed cake as the substrate at 20% substrate concentration and 45% moisture content after 84 h of incubation. An increase of 2.95- and 2.04-fold from basal media was observed in protease and amylase production, respectively. ANOVA of both the design models showed high accuracy of the polynomial model with significant similarities between the predicted and the observed results. The model stood accurate at the bench level validation, suggesting that the design model could be used for multienzyme production at mass scale.

Background: Rhizopus arrhizus is a potential microorganism for lipase production. Solid-state fermentation is used for microbial biosynthesis of enzymes, due to advantages, such as high productivity, utilization of abundant and low-cost raw materials, and production of enzymes with different catalytic properties. Objective: The objective of the research is optimization of the conditions for lipase production in solid-state fermentation by Rhizopus arrhizus in a nutrient medium, containing agroindustrial wastes. Method: Biosynthesis of lipase in solid-state fermentation by Rhizopus arrhizus was investigated. The effect of different solid substrates, additional carbon and nitrogen source, particles size and moisture content of the medium on enzyme production was studied. Response surface methodology was applied for determination of the optimal values of moisture content and tryptone concentration. A procedure for efficient lipase extraction from the fermented solids was developed. Results: Highest lipase activity was achieved when wheat bran was used as a solid substrate. The addition of 1% (w/w) glucose and 5% (w/w) tryptone to the solid medium significantly increased lipase activity. The structure of the solid medium including particles size and moisture content significantly influenced lipase production. A mathematical model for the effect of moisture content and tryptone concentration on lipase activity was developed. Highest enzyme activity was achieved at 66% moisture and 5% (w/w) tryptone. The addition of the non-ionic surfactant Disponyl NP 3070 in the eluent for enzyme extraction from the fermented solids increased lipase activity about three folds. Conclusion: After optimization of the solid-state fermentation the achieved 1021.80 U/g lipase activity from Rhizopus arrhizus was higher and comparable with the activity of lipases, produced by other fungal strains. The optimization of the conditions and the use of low cost components in solid-state fermentation makes the process economicaly effective for production of lipase from the investigated strain Rhizopus arrhizus.

Abstract El objetivo de este estudio fue evaluar el posible efecto probiótico del sustrato de residuos agroindustriales fermentado con BAL y levaduras sobre los indicadores productivos, reproductivos y de salud de cobayas primíparas y su descendencia. Se utilizaron 80 cuyes nulíparas distribuidas en 4 grupos de 20 hembras nulíparas cada uno, de 120 ± 5 días de edad, 1450 ± 25 g de peso vivo, todas sometidas al mismo sistema de manejo y alimentación actual de la granja. Se aplicaron con una dosis de 1 mL del bioaditivo / animal. Los tratamientos suministrados fueron las siguientes variantes: T1. control. T2, Sustrato (residuos agroindustriales) fermentado con L. acidophilus ATCC® 4356 ™. T3, sustrato más Kluyveromyces fragilis L4 (UCLV). T4, sustrato fermentado con microorganismos T2 y T3.En el experimento se utilizó un diseño completamente al azar, donde se evaluaron los indicadores reproductivos, productivos y de salud. Weight gain in pregnancy was greater (P < 0.05) in T4 in this same group, weight loss was less (P < 0.05) in lactation; the age at first conception and at delivery decreased (P < 0.05) in T4; fertility, the number of live-born animals and the weight at birth and at weaning were higher (P < 0.05) in T4 compared to the other groups; Likewise, blood parameters notably improved in the animals of treatment 4. The treaties with bioaditive were the ones whit the best behavior as reflected in table 1. In the experiment, the beneficial effect as a probiotic was confirmed by the treatments (T2, T3 and T4) of these with the best performance was T4. The inclusion of substrates fermented with LAB and yeasts significantly improves the productive, reproductive and health indicators of primiparous guinea pigs and their offspring.
