ABSTRACT Retrotransposon Ty1 faces a formidable cell barrier during transposition—the yeast nuclear membrane which remains intact throughout the cell cycle. We investigated the mechanism by which transposition intermediates are transported from the cytoplasm (the presumed site of Ty1 DNA synthesis) to the nucleus, where they are integrated into the genome. Ty1 integrase has a nuclear localization signal (NLS) at its C terminus. Both full-length integrase and a C-terminal fragment localize to the nucleus. C-terminal deletion mutants in Ty1 integrase were used to map the putative NLS to the last 74 amino acid residues of integrase. Mutations in basic segments within this region decreased retrotransposition at least 50-fold in vivo. Furthermore, these mutant integrase proteins failed to localize to the nucleus. Production of virus-like particles, reverse transcriptase activity, and complete in vitro Ty1 integration resembled wild-type levels, consistent with failure of the mutant integrases to enter the nucleus.

Η µιτωτική άτρακτος είναι µία µοριακή µηχανή δοµικά βασισµένη σε µικροσωληνίσκους, που παρουσιάζει ιδιαίτερη δυναµικότητα και διενεργεί την ίση κατανοµή των διπλασιασµένων χρωµοσωµάτων στα δύο θυγατρικά κύτταρα κατά την κυτταρική διαίρεση.Ο σχηµατισµός της ατράκτου σε σωµατικά κύτταρα θηλαστικών βασίζεται στις συντονισµένες δραστηριότητες δύο βιοχηµικών µονοπατιών: το µονοπάτι εξαρτώµενο από τα κεντροσώµατα και το µονοπάτι εξαρτώµενο από τη χρωµατίνη, τα οποία ικανοποιούν δύο ευρέως αποδεκτά µοντέλα για τη συναρµολόγηση της ατράκτου, το µοντέλο “search–and-capture” και το µοντέλο του “self-organization”,αντίστοιχα. Γνωρίζουµε ότι η χρωµατίνη παράγει ενδοκυτταρικά σήµατα χωρικής οργάνωσης που ευνοούν την εµπυρήνωση ή/και τη σταθεροποίηση των µικροσωληνίσκων. H κύρια ενδοκυττάρια διαβάθµιση συγκέντρωσης που δηµιουργείται από τα χρωµοσώµατα και συµβάλλει στην αυτο-οργάνωση της µιτωτικής ατράκτου είναι η διαβάθµιση συγκέντρωσης του RanGTP. Η µικρή GTPάσηRan όταν είναι συνδεδεµένη µε GTP, παρουσιάζει µια ενδογενή φθίνουσα διαβάθµιση συγκέντρωσης γύρω από την επιφάνεια των χρωµοσωµάτων λόγω παρουσίας εκεί του παράγοντα ανταλλαγής GTP,RCC1. Το RanGTP που επάγεται από τη χρωµατίνη προωθεί τη συναρµολόγηση της ατράκτου µέσω απελευθέρωσης παραγόντων που ευνοούν την συναρµολόγηση της ατράκτου (Spindle AssemblyFactors, SAFs) και περιέχουν σήµα πυρηνικού εντοπισµού (Nuclear Localization Signal, NLS) από ανασταλτικά σύµπλοκα µε ιµπορτίνες α ή / και β. Το ρυθµιστικό δίκτυο της συναρµολόγησης της ατράκτου ελέγχεται επιπροσθέτως από τη δράση των µιτωτικών κινασών που οδηγούν σε γεγονότα αναστρέψιµης φωσφορυλίωσης, ρυθµίζοντας έτσι τη λειτουργία των πρωτεϊνών που προσδένονται σε µικροσωληνίσκους (Microtubule Associated Proteins, MAPs) και των κινεσινών.Η σωστή διαµόρφωση και λειτουργία της µιτωτικής ατράκτου απαιτούνται για τη διενέργεια της κυτταρική διαίρεση χωρίς σφάλµατα. Παρ 'όλα αυτά, ακόµη και µια µικρή απορρύθµιση αυτών των µιτωτικών πρωτεϊνών, που δεν θέτει σε κίνδυνο τη βιωσιµότητα των κυττάρων, µπορεί να οδηγήσει σε ανώµαλη κυτταρική διαίρεση και τελικά σε ανευπλοειδία, ένα χαρακτηριστικό γνώρισµα του καρκίνου.Κατά τα τελευταία χρόνια, έχουν αναφερθεί γενετικές και επιγενετικές αλλοιώσεις σε έναν σηµαντικόαριθµό µιτωτικών γονιδίων σε ανθρώπινους όγκους. Οι αλλοιώσεις αυτές που παρουσιάζονται σε µιτωτικά γονίδια και σχετίζονται µε τον καρκίνο φαίνεται να επηρεάζουν κυρίως τα επίπεδα γονιδιακής έκφρασης. Η πλειοψηφία των µιτωτικών γονιδίων που παρουσιάζουν αλλοιώσεις στον καρκίνο περιλαµβάνει κυρίως κινάσες, πρωτεΐνες του σηµείου ελέγχου της ατράκτου και δοµικές πρωτεΐνες της ατράκτου συµπεριλαµβανοµένων των πρωτεϊνών που προσδένονται σε µικροσωληνίσκους και των κινεσινών.Για το σκοπό αυτό κατευθυνθήκαµε προς την µελέτη µίας πρωτεΐνης που προσδένεται σε µικροσωληνίσκους µε ρόλο-κλειδί στον σχηµατισµό ατράκτου, την πρωτεΐνη HURP (Hepatoma UpRegulated Protein) καθώς και προς τη µελέτη του πιθανού της ρόλου στον καρκίνο. Η HURP έχειαρχικά αναγνωριστεί ως µέρος ενός RanGTP εξαρτώµενoυ πολυ-πρωτεϊνικού σύµπλοκου που προάγει την συγκρότηση της διπολικής ατράκτου σε εκχυλίσµατα αυγών του βατράχου Xenopus και αποτελείται από δύο επιπλέον πρωτεΐνες που προσδένονται σε µικροσωληνίσκους την TPX2 και τηνXMAP215, την κινεσίνη Eg5 και την κινάση Aurora Α. Η HURP ρυθµίζεται αυστηρά κατά τη διάρκειατης µίτωσης ως προς τα επίπεδα έκφρασής της και ως προς τον εντοπισµό της. Τα επίπεδα της πρωτεΐνης HURP φτάνουν στο µέγιστο κατά τη φάση G2/M του κυτταρικού κύκλου ενώ ο εντοπισµός της πρωτεΐνης περιορίζεται σε ένα υποσύνολο των µικροσωληνίσκων της ατράκτου, τους µικροσωληνίσκους του κινητοχώρου (k-ΜΤs). Στη µεταφασική άτρακτο η HURP εντοπίζεται κυρίως στους µικροσωληνίσκους του κινητοχώρου σε διαβάθµιση συγκέντρωσης κατά µήκος του άξονα των δύο πόλων παρουσιάζοντας υψηλότερη συγκέντρωση σε περιοχές κοντά στην χρωµατίνη, φθίνονταςπρος τους πόλους. Η HURP αποτελεί υπόστρωµα της Aurora Α και η φωσφορυλίωσή της διεγείρει τον ογκογόνο µετασχηµατισµό καλλιεργηµένων κυττάρων.Για να κατανοήσουµε καλύτερα το ρόλο της HURP στη συναρµολόγηση της ατράκτου αναπτύξαµε µικρά-µόρια αναστολείς της κινάσης Aurora Α. Σε αυτή τη µελέτη, από ένα σύνολο 105 πιθανών µικροµοριακών αναστολέων, δύο ενώσεις ο Tripolin Α και ο Tripolin Β, βρέθηκαν να αναστέλλουν την δραστικότητα της Aurora A κινάσης in vitro. Ωστόσο, σε ανθρώπινα καρκινικά κύτταρα, µόνο ο TripolinΑ δρα ως αναστολέας της Aurora Α. Συνδυάσαµε βιοχηµικά πειράµατα in vitro και µελέτες συνεστιακής µικροσκοπίας µεµονοµένων κυττάρων in vivo, για να εξετάσουµε τη βιολογική δραστικότητα τουTripolin Α. Τα ευρήµατά µας αποδεικνύουν ότι ο Tripolin Α είναι ένας αναστολέας της Aurora Α, µη-ανταγωνιστικός του ΑΤΡ. Ο Tripolin Α µειώνει τον εντοπισµό της Aurora Α στους µικροσωληνίσκουςτης ατράκτου, επηρεάζει την ακεραιότητα των κεντροσωµάτων, επηρεάζει τον σχηµατισµό και το µήκος της ατράκτου, καθώς και τη δυναµική των µικροσωληνίσκων στην µεσόφαση, αποτελέσµατα που συνάδουν µε απαλοιφή της Aurora A. Ενδιαφέρον παρουσιάζει ότι ο Tripolin Α επηρεάζει την διαβάθµιση συγκέντρωσης της HURP προς τα χρωµοσώµατα, αλλά όχι την πρόσδεσή της στους µικροσωληνίκους. Πειράµατα ανάκτησης φθορισµού µετά από αποχρωµατισµό FRAP (FluorescenceRecovery After Photobleaching, FRAP) δείχνουν ότι η Aurora A ρυθµίζει την ανταλλαγή της HURP στις θέσεις πρόσδεσής της στους µικροσωληνίκους της ατράκτου. Ως εκ τούτου, καταλήγουµε στο συµπέρασµα ότι η φωσφορυλίωση από την Aurora A διατηρεί την διαβάθµιση συγκέντρωσης του εντοπισµού της HURP στην άτρακτο.Προγενέστερες µελέτες γονιδιακής έκφρασης έχουν εντοπίσει υπερέκφραση της HURP σε ανθρώπινους καρκίνους όπως τον καρκίνο του ήπατος και της ουροδόχου κύστης. Για το σκοπό αυτό εξετάσαµε τα επίπεδα γονιδιακής έκφρασης της HURP σε µια οµάδα κυτταρικών σειρών που αντιπροσωπεύουν την πολυσταδιακή χηµική καρκινογένεση της επιδερµίδας του ποντικού,προκειµένου να αναπαραστήσουµε την διαδικασία πολλαπλών βηµάτων για την ανάπτυξη του καρκίνου, σε ένα µοντέλο κυτταρικών σειρών. Οι κυτταρικές σειρές αντιπροσωπεύουν την ανάπτυξη των τριών διαφορετικών στάδιων της καρκινογένεσης, την έναρξης, την προώθηση και την εξέλιξης του όγκου και προέρχονται από όγκους του δέρµατος ποντικού που επάγονται σε δύο στάδια, µε επίδρασηDMBA / ΤΡΑ (πολυκυκλικός αρωµατικός υδρογονάνθρακας / φορβολικός εστέρας). Εφαρµόσαµε λοιπόν, ανάλυση γονιδιακής έκφρασης χρησιµοποιώντας ποσοτική PCR πραγµατικού χρόνου(Quantitative Real Time PCR, qPCR) και αποτύπωµα κατά Western καi βρήκαµε ότι τα επίπεδα mRNAκαι πρωτεΐνης του γονιδίου της Hurp είναι αυξηµένα στις κυτταρικές σειρές που προέρχονται απόκαρκινώµατα πλακώδων κυττάρων και από αναπλαστικά ατρακτοειδή καρκίνωµατα σε σύγκριση µε τις κυτταρικές σειρές που προέρχονται από καλοήθη θηλώµατα. Συλλογικά, προτείνουµε ότι τα επίπεδα έκφρασης της HURP στον καρκίνο δεν συσχετίζονται µε την καλοήθη υπερπλασία αλλά αυξάνουν µε την κακοήθη εξαλλαγή.Στο σύνολό τους τα αποτελέσµατα αυτής της µελέτης αποκάλυψαν µια νέα ρυθµιστική λειτουργία της κινάσης Aurora A στην πρωτεΐνη HURP πάνω στη µιτωτική άτρακτο. Επιπλέον έχουµε δώσει νέες κατευθύνσεις σχετικά µε το ρόλο της HURP στην εξέλιξη του καρκίνου.

Initiation of eukaryotic genome duplication begins when a six-subunit origin recognition complex (ORC) binds to DNA. However, the mechanism by which this occurs in vivo and the roles played by individual subunits appear to differ significantly among organisms. Previous studies identified a soluble human ORC(2–5) complex in the nucleus, an ORC(1–5) complex bound to chromatin, and an Orc6 protein that binds weakly, if at all, to other ORC subunits. Here we show that stable ORC(1–6) complexes also can be purified from human cell extracts and that Orc6 and Orc1 each contain a single nuclear localization signal that is essential for nuclear localization but not for ORC assembly. The Orc6 nuclear localization signal, which is essential for Orc6 function, is facilitated by phosphorylation at its cyclin-dependent kinase consensus site and by association with Kpna6/1, nuclear transport proteins that did not co-purify with other ORC subunits. These and other results support a model in which Orc6, Orc1, and ORC(2–5) are transported independently to the nucleus where they can either assemble into ORC(1–6) or function individually.

ABSTRACT The “classical” nuclear protein import pathway depends on importin α and importin β. Importin α binds nuclear localization signal (NLS)-bearing proteins and functions as an adapter to access the importin β-dependent import pathway. In humans, only one importin β is known to interact with importin α, while six α importins have been described. Various experimental approaches provided evidence that several substrates are transported specifically by particular α importins. Whether the NLS is sufficient to mediate importin α specificity is unclear. To address this question, we exchanged the NLSs of two well-characterized import substrates, the seven-bladed propeller protein RCC1, preferentially transported into the nucleus by importin α3, and the less specifically imported substrate nucleoplasmin. In vitro binding studies and nuclear import assays revealed that both NLS and protein context contribute to the specificity of importin α binding and transport.

Human cytomegalovirus (HCMV) DNA-binding protein pUL56 is thought to be involved in the cleavage/packaging process of viral DNA and therefore needs to be transported into the nucleus. By using indirect immunofluorescence analysis, HCMV pUL56 (p130) was found to be localized predominantly in the nucleus of infected cells. Solitary expression of wild-type as well as epitope-tagged pUL56 also resulted in nuclear distribution after transfection, suggesting the presence of an endogenous nuclear localization signal (NLS). Deletion of a carboxy-terminal stretch of basic amino acids (aa 816–827) prevented nuclear translocation, indicating that the sequence RRVRATRKRPRR of HCMV pUL56 mediates nuclear targetting. The signal character of the NLS sequence was demonstrated by successful transfer of the NLS to a reporter protein chimera. Furthermore, sequential substitutions of pairs of amino acids by alanine in the context of the reporter protein as well as substitutions within the full-length pUL56 sequence indicated that residues at positions 7 and 8 of the NLS (R and K at positions 822 and 823 of pUL56) were essential for nuclear translocation. In order to identify the transport machinery involved, the potential of pUL56 to bind importin α (hSRP1α) was examined. Clear evidence of a direct interaction of a carboxy-terminal portion as well as the NLS of pUL56 with hSRP1α was provided by in vitro binding assays. In view of these findings, it is suggested that nuclear translocation of HCMV pUL56 is mediated by the importin-dependent pathway.

ABSTRACT The nuclear import of proteins bearing a basic nuclear localization signal (NLS) is dependent on karyopherin α/importin α, which acts as the NLS receptor, and karyopherin β1/importin β, which binds karyopherin α and mediates the nuclear import of the resultant ternary complex. Recently, a second nuclear import pathway that allows the rapid reentry into the nucleus of proteins that participate in the nuclear export of mature mRNAs has been identified. In mammalian cells, a single NLS specific for this alternate pathway, the M9 NLS of heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1), has been described. The M9 NLS binds a transport factor related to karyopherin β1, termed karyopherin β2 or transportin, and does not require a karyopherin α-like adapter protein. A yeast homolog of karyopherin β2, termed Kap104p, has also been described and proposed to play a role in the nuclear import of a yeast hnRNP-like protein termed Nab2p. Here, we define a Nab2p sequence that binds to Kap104p and that functions as an NLS in both human and yeast cells despite lacking any evident similarity to basic or M9 NLSs. Using an in vitro nuclear import assay, we demonstrate that Kap104p can direct the import into isolated human cell nuclei of a substrate containing a wild-type, but not a defective mutant, Nab2p NLS. In contrast, other NLSs, including the M9 NLS, could not function as substrates for Kap104p. Surprisingly, this in vitro assay also revealed that human karyopherin β1, but not the Kap104p homolog karyopherin β2, could direct the efficient nuclear import of a Nab2p NLS substrate in vitro in the absence of karyopherin α. These data therefore identify a novel NLS sequence, active in both yeast and mammalian cells, that is functionally distinct from both basic and M9 NLS sequences.
