Abstract—The present study examined Empathizing-Systemizing Theory (E-S Theory) in Indonesian Children. E-S Theory has known as a theory that is attempted to explain the cluster of both the social and non-social features in autism spectrum conditions. Children’s version of the Empathy Quotient (EQ-C) and the Systemizing Quotient (SQ-C) were administered to 372 Indonesia parents who had typically developed children aged 4-11 years old (boys = 182, girls = 190). The results showed that the girls scored higher that boys on EQ-C . In the case of SQ-C, there were no differences between boys and girls. However, the proportion of cognitive style shows clear individual differences between boys and girls. The proportion of participants with empathizing dominant cognitive sytle was shown higher by girls than boys, which distributed to E and Extreme E brain types, while the proportion of systemizing dominant cognitive styles is higher in boys whom brain types spread across the Type S and Extreme S categories. This study explaining inherited biological factors of autism and  underscore the vulnerability among  boys to develop autism spectrum condition if compared to girls. E-S Theory dikenal sebagai sebuah teori yang digunakan untuk melihat gugus fitur sosial dan non-sosial kondisi spektrum autisme. Subjek penelitian ini adalah 372 orang tua Indonesia dengan anak berusia 4-11 tahun (anak laki-laki = 182, perempuan = 190), M usia = 7 Tahun, SD= 2.423 yang diminta untuk mengisi skala Empathizing Quotient (EQ) dan Systemizing Quotient (SQ). Hasil penelitian menunjukkan bahwa bahwa tingkat empati anak perempuan lebih tinggi daripada tingkat empati anak laki-laki. Dalam hal mensistemasi tidak ditemukan adanya perbedaan kemampuan antara anak laki-laki dan perempuan. Namun, proporsi gaya kognitif menunjukkan adanya perbedaan individual yang signifikan antara anak laki-laki dan perempuan. Gaya kognitif dominan berempati lebih tinggi pada anak perempuan, yang tersebar pada kategori tipe otak E dan Ekstrim E, sedangkan proporsi gaya kognitif mensistemasi lebih tinggi pada anak laki-laki dengan tipe otak yang tersebar pada kategori Tipe S dan Ekstrim S.  Hasil penelitian ini memberikan bukti adanya faktor biologis bawaan dari autism ditinjau dari jenis kelamin dan menunjukkan kerentanan bawaan anak laki-laki terhadap kondisi autism jika dibandingkan dengan anak perempuan. Autism spectrum conditions (ASCs) are a category of neurodevelopmental disorders with symptoms of communication and social impairment, and the exhibition of restrictive and repetitive behaviours. Their occurrence is greater in males than females and this sex difference has played an important part in hypothesizing their etiology. The Extreme Male Brain (EMB) theory is a cognitive model proposed by Simon Baron-Cohen to explain the aforementioned sex differences and potential cause of ASCs. It is based upon his Empathizing Systemizing theory, which classifies individuals into one of five cognitive profiles (Type S, Type E, Type B, Extreme Type S and Extreme Type E). These cognitive profiles determine an individual’s ability to systemize and empathize. Systemizing is the ability to understand and derive the rules of a system, and requires deductive and analytical skills. Empathizing relates to understanding human emotion and behaviour, thus requires social and communication skills. Males tend to systemize better than empathize while females have an opposite profile. Based upon the EMB theory, autistic individuals would possess an Extreme Type S profile as their impairments in social communication can be explained by a deficit in empathizing, while their preoccupation with patterns and detail-oriented behaviour can be related to their high systemizing. Together, these cognitive models have resulted in the Foetal Testosterone (fT) Theory, which implicates high prenatal testosterone as a risk factor for the associated hypermasculinized cognitive profile of individuals with ASCs. This review paper assesses the validity of the EMB and fT theories by reviewing the literature relating fT with autistic traits in the general population. The seven studies confirmed a correlation between higher fT levels and an increase in autistic traits, but limitations need to be considered when generalizing this information to an ASC sample.

Weather is important to all people, including vulnerable populations (those whose circumstances include cognitive processing, hearing, or vision differences, physical disability, homelessness, and other scenarios and factors). Autism spectrum conditions (ASC) affect information-processing and areas of neurological functioning that potentially inhibit the reception of hazardous weather information, and is of particular concern for weather messengers. People on the autism spectrum tend to score highly in tests of systemizing, a psychological process that heavily entails attention to detail and revolves around the creation of logical rules to explain things that occur in the world. This article reports the results of three preliminary studies examining weather salience–psychological attention to weather–and its potential relationships with systemizing in autistic people. Initial findings suggest that enhanced weather salience exists among autistic individuals compared to those without the condition, and that this may be related to systemizing. These findings reveal some possible strategies for communicating weather to autistic populations and motivate future work on a conceptual model that blends systemizing and chaos theory to better understand weather salience.

Abstract Background The inability to observe relevant biological processes in vivo significantly restricts human neurodevelopmental research. Advances in appropriate in vitro model systems, including patient-specific human brain organoids and human cortical spheroids (hCSs), offer a pragmatic solution to this issue. In particular, hCSs are an accessible method for generating homogenous organoids of dorsal telencephalic fate, which recapitulate key aspects of human corticogenesis, including the formation of neural rosettes—in vitro correlates of the neural tube. These neurogenic niches give rise to neural progenitors that subsequently differentiate into neurons. Studies differentiating induced pluripotent stem cells (hiPSCs) in 2D have linked atypical formation of neural rosettes with neurodevelopmental disorders such as autism spectrum conditions. Thus far, however, conventional methods of tissue preparation in this field limit the ability to image these structures in three-dimensions within intact hCS or other 3D preparations. To overcome this limitation, we have sought to optimise a methodological approach to process hCSs to maximise the utility of a novel Airy-beam light sheet microscope (ALSM) to acquire high resolution volumetric images of internal structures within hCS representative of early developmental time points. Results Conventional approaches to imaging hCS by confocal microscopy were limited in their ability to image effectively into intact spheroids. Conversely, volumetric acquisition by ALSM offered superior imaging through intact, non-clarified, in vitro tissues, in both speed and resolution when compared to conventional confocal imaging systems. Furthermore, optimised immunohistochemistry and optical clearing of hCSs afforded improved imaging at depth. This permitted visualization of the morphology of the inner lumen of neural rosettes. Conclusion We present an optimized methodology that takes advantage of an ALSM system that can rapidly image intact 3D brain organoids at high resolution while retaining a large field of view. This imaging modality can be applied to both non-cleared and cleared in vitro human brain spheroids derived from hiPSCs for precise examination of their internal 3D structures. This process represents a rapid, highly efficient method to examine and quantify in 3D the formation of key structures required for the coordination of neurodevelopmental processes in both health and disease states. We posit that this approach would facilitate investigation of human neurodevelopmental processes in vitro.

AbstractThe potential etiological role of early acetaminophen exposure on Autism Spectrum Conditions (ASC) and Attention-Deficit/Hyperactivity Disorder (ADHD) is inconclusive. We aimed to study this association in a collaborative study of six European population-based birth/child cohorts. A total of 73,881 mother–child pairs were included in the study. Prenatal and postnatal (up to 18 months) acetaminophen exposure was assessed through maternal questionnaires or interviews. ASC and ADHD symptoms were assessed at 4–12 years of age using validated instruments. Children were classified as having borderline/clinical symptoms using recommended cutoffs for each instrument. Hospital diagnoses were also available in one cohort. Analyses were adjusted for child and maternal characteristics along with indications for acetaminophen use. Adjusted cohort-specific effect estimates were combined using random-effects meta-analysis. The proportion of children having borderline/clinical symptoms ranged between 0.9 and 12.9% for ASC and between 1.2 and 12.2% for ADHD. Results indicated that children prenatally exposed to acetaminophen were 19% and 21% more likely to subsequently have borderline or clinical ASC (OR = 1.19, 95% CI 1.07–1.33) and ADHD symptoms (OR = 1.21, 95% CI 1.07–1.36) compared to non-exposed children. Boys and girls showed higher odds for ASC and ADHD symptoms after prenatal exposure, though these associations were slightly stronger among boys. Postnatal exposure to acetaminophen was not associated with ASC or ADHD symptoms. These results replicate previous work and support providing clear information to pregnant women and their partners about potential long-term risks of acetaminophen use.

In a special issue that focuses on complex presentations related to Autism, we ask the question in this editorial whether an Autism Spectrum Condition without complexity is a disorder, or whether it represents human diversity? Much research into Autism Spectrum Conditions (ASCs) over the years has focused on comparisons between neuro-typical people and people with Autism Spectrum Conditions. These comparisons have tended to draw attention to ‘deficits’ in cognitive abilities and descriptions of behaviours that are characterised as unwanted. Not surprisingly, this is reflected in the classification systems from the World Health Organisation and the American Psychiatric Association. Public opinion about ASC may be influenced by presentations in the media of those with ASC who also have intellectual disability. Given that diagnostic systems are intended to help us better understand conditions in order to seek improved outcomes, we propose a more constructive approach to descriptions that uses more positive language, and balances descriptions of deficits with research finding of strengths and differences. We propose that this will be more helpful to individuals on the Autism Spectrum, both in terms of individual self-view, but also in terms of how society views Autism Spectrum Conditions more positively. Commentary has also been made on guidance that has been adjusted for people with ASC in relation to the current COVID-19 pandemic.
