Presence of Newcastle disease viruses of sub-genotypes Vc and VIn in backyard chickens and in apparently healthy wild birds from Mexico in 2017

Aim: Newcastle disease (ND) is one of the most important avian diseases. Virulent strains of Newcastle disease virus (NDV) have the potential of rapid spread, and may cause serious economic impact and international trade restrictions for the poultry industry. The objective was to study the clinical, gross and histo-pathological and immunohistopathological changes of Newcastle disease infection in apparently healthy and sick indigenous chickens, ducks, pigeons and some wild birds in Plateau State. Methodology: The indigenous chickens used in this study were randomly selected from apparently healthy and from those with suggestive clinical signs of ND. A total of 638 birds were used for the study. Out of the total number of birds sampled, 349 were indigenous chickens, 98 pigeons, 96 ducks and 95 from different species of wild birds. Out of the number sampled from indigenous chickens, 169 (44.01%) were live birds, while 180 (46.90%) were carcasses. Tissues were collected from indigenous chickens, pigeons, ducks and some wild birds from both sick, and apparently healthy unvaccinated flocks to screen for the presence of NDV by immunohistochemical (IHC) techniques.  The histopathology and immunohistochemistry were done using standard laboratory procedures. Results: Clinical signs observed in live birds generally varied from weakness, greenish watery diarrhoea, respiratory difficulty, anorexia and coughing, torticollis, droopy wings, paralysis, partial leg paralysis, and opisthotonos. Generally, the gross lesions in euthanized and dead birds were mostly hyperaemia, hepatomegaly, splenomegaly, moderate enlargement of the heart, petechial haemorrhages on the mucosa surface of the proventriculus and haemorrhagic tracheitis, congestion and moderate enlargement of the pancreas, pulmonary congestion and congested kidneys. Histopathological changes include lymphoid depletion and connective tissue proliferation, enteritis, pulmonary congestion and splenitis. A total of six samples (1.56%) out of 349 from indigenous chickens had positive staining for NDV antigen using IHC technique. While there were only two samples out of the 95  wild birds samples positive by IHC, both of the positive samples were from Red-eyed Dove (Streptopelia semitorquata), resulting in a 25% (2/8) positive rate from this species; had positive staining for NDV by immunohistochemistry. All IHC positive cases in this study, in both wild birds and indigenous chickens shared similar staining patterns. Conclusion: The study also shows that NDV antigens in wild birds and indigenous chickens concentrate more in the spleen, pancreas, trachea and proventriculus. This study, presents for the first time to the best of our knowledge that viral antigens (NDV) in wild birds and indigenous chickens were demonstrated by immunohistochemical technique in Plateau State, Nigeria.

ABSTRACTNewcastle disease (ND), caused by the virulent Newcastle disease virus (NDV), is one of the most important viral diseases of birds globally, but little is currently known regarding enzootic trends of NDV in northeastern China, especially for class I viruses. Thus, we performed a surveillance study for NDV in northeastern China from 2013 to 2015. A total 755 samples from wild and domestic birds in wetlands and live bird markets (LBMs) were collected, and 10 isolates of NDV were identified. Genetic and phylogenetic analyses showed that five isolates from LBMs belong to class I subgenotype 1b, two (one from wild birds and one from LBMs) belong to the vaccine-like class II genotype II, and three (all from wild birds) belong to class II subgenotype Ib. Interestingly, the five class I isolates had epidemiological connections with viruses from southern, eastern, and southeastern China. Our findings, together with recent prevalence trends of class I and virulent class II NDV in China, suggest possible virus transmission between wild and domestic birds and the potential for an NDV epidemic in the future.

Introduction:Newcastle Disease (ND), caused by Avian avulavirus 1 (AAvV 1, avulaviruses), is a notifiable disease throughout the world due to the economic impact on trading restrictions and its embargoes placed in endemic regions. The feral birds including aquatic/migratory birds and other wild birds may act as natural reservoir hosts of ND Viruses (NDVs) and may play a remarkable role in the spread of the virus in environment. In addition, other 19 avulaviruses namely: AAvV 2 to 20, have been potentially recognized from feral avian species.Expalantion:Many previous studies have investigated the field prevailing NDVs to adapt a wide range of susceptible host. Still the available data is not enough to declare the potential role of feral birds in transmission of the virus to poultry and/or other avian birds. In view of the latest evidence related to incidences of AAvVs in susceptible avian species, it is increasingly important to understand the potential of viruses to transmit within the domestic poultry and other avian hosts. Genomic and phylogenomic analysis of several investigations has shown the same (RK/RQRR↓F) motif cleavage site among NDV isolates with same genotypes from domestic poultry and other wild hosts. So, the insight of this, various semi-captive/free-ranging wild avian species could play a vital role in the dissemination of the virus, which is an important consideration to control the disease outbreaks. Insufficient data on AAvV 1 transmission from wild birds to poultry and vice versa is the main constraint to understand about its molecular biology and genomic potential to cause infection in all susceptible hosts.Conclusion:The current review details the pertinent features of several historical and contemporary aspects of NDVs and the vital role of feral birds in its molecular epidemiology and ecology.
