Nigrospora oryzae was isolated as an endophytic fungus from the leaves of Coccinia grandis, a popular medicinal plant used to control diabetes. Fermentation of the fungus in potato dextrose broth and chromatographic purification of the ethyl acetate extracts of the broth and mycelium yielded two phenazine secondary metabolites, which were identified as phenazine-1-carboxylic acid (1) and phenazine-1-carboxamide (2) by comparing their spectral data with those reported in the literature. Compound 2, isolated in high yield (1 g/4 L medium), showed strong antifungal activity against the plant pathogen Cladosporium cladosporioides. This is the first report of the isolation of N. oryzae as an endophytic fungus of C. grandis. These phenazines have never been isolated from any fungal source. Antifungal activity of 2 against C. cladosporioides is reported for the first time.

AbstractThis study was conducted to evaluate anti-acetylcholinesterase and insecticidal and antifungal activities of the endophytic fungus Trichoderma sp, isolated from Ricinus communis L. leaves, against Locusta migratoria L. and Botrytis cinerea Pers.: Fr.. To evaluate the insecticidal and antifungal activities, different concentrations of the fungal extract were applied against L. migratoria (0.2, 0.3, 0.4 g/l) and against B. cinerea (1, 2, 3 g/l). It was found that the mortality of the targeted insects was positively proportional to fungal extract concentration and time after exposure (24, 48, 72 hours). The concentration 0.4 g/l appeared to be the most effective after 72 hours with mortality rate of 56.52%. Regarding antifungal activity, the concentration 3 g/l was the most effective against B. cinerea after 7 days, with an inhibition rate of 92.06% (excellent antifungal activity). Moreover, it was found that at 4 ug/ml the fungal extract had a maximum inhibitory capacity of Ache of 80% for acetylcholenesterase. Preliminary phytochemical analyses revealed the presence of alkaloids, flavonoids, phenols and saponins. In addition the colony of this endophytic fungus produced chitinases and proteases, which explained its important antifungal and insecticidal activities.

A new macrolactone glycoside, lecythomycin (1), 23-methyl-3-(1- O-mannosyl)-oxacyclotetracosan-1-one, was isolated from the endophytic fungus Lecythophora sp. (code 30.1), an endopyte of the Indonesian plant Alyxia reinwardtii. The structure of 1 was elucidated on the basis of NMR spectroscopic and mass spectrometric data. The isolated compound displayed antifungal activity against strains of Aspergillus fumigatus and Candida kruzei at minimal inhibitory concentrations (MIC) of 62.5 – 125 μg/mL.
