Drought has been a big problem and damaged seriously to rice cultivation and production in Vietnam and the Mekong Delta region; evaluating drought tolerance of rice is a major objective for the rice improvement programmes in Can Tho University. Fifty-two collected rice varieties including resistant and susceptible control varieties were screened for water stress under the artificial drought condition. Marker RM223 was used to identify the drought tolerance genotypes for some selected varieties with good and moderate tolerant scores. After 30 days of water stress, the results were 6 varieties of good tolerant, 8 varieties of moderate tolerance, 36 varieties of moderately susceptible and 2 varieties of susceptible to drought. Analyses of PCR showed that 10 varieties expressed the similar bands with the resistant control variety. Four varieties (LH8, MTL812, Lua Canh and VB1) with good tolerant to drought were recommended to use for genetic materials of rice breeding program and applying in alternative wetting and drying irrigation technique for rice cultivation.

Rice (Oryza sativa L.) is a species of crops which is highly sensitive to drought. The sensitivity to water stress varies from genotypes to genotypes of rice. An experiment was performed to compare twenty-five different rice genotypes for drought tolerance in the seedling stage at Gokuleshwor Agriculture and Animal Science College, Baitadi Nepal from July to August 2018. The genotypes were tested under two conditions viz., water stress and non-water stress. The research work was carried out in a completely random design (CRD) with three replications. The results exhibited the height of seedling, length & breadth of leaf, number of leaves/plant decreased, and number of leaves shedding increased under water stress condition. Clustering was done by the method of average linkage, and genotypes were categorized into five clusters. The genotype, namely Radha 11 grouped in Cluster-V had higher plant height, bigger length & breadth of leaf, maximum number of leaves/plant and minimum number of leaf shedding. Thus, this genotype showed the best performance for morphological traits under water stress condition. Therefore, this genotype Radha 11 can be utilized further for developing rice variety with drought-tolerance.

The pandan scent in aromatic rice has been known as the result of 8 bp deletions and 3 Single Nucleotide Polymorphisms (SNPS) in BADH2 gene, which produce non-functional betaine aldehyde dehydrogenase (BADH) enzyme. Several DNA markers for aromatic character based on mutation in BADH2 gene have been developed. In our experiment, we analysed the presence of aromatic character in four local rice variety such as Merah Wangi, Pendok, Genjah Arum, and Mentik Wangi Susu using KOH method and DNA molecular method using three DNA markers to detect mutation that responsible for the development of aromatic character. Phenotype analysis using KOH method showed that Merah Wangi, Genjah Arum, and Mentik Wangi Susu produce pandan scents. PCR analysis using Bradbury and Badex7-5, and RM223 markers showed the presence of BADH2 mutation in Merah Wangi and Mentik Wangi Susu, whereas Pendok and Genjah Arum did not show BADH2 mutation using those used three markers. Our results indicate that among four investigated local rice, only Merah Wangi and Mentik Wangi Susu are categorized as aromatic rice whereas Pendok and Genjah Arum are non-aromatic.Keywords: genetic analysis, aromatic rice, specific markers, genetic mutation.

Abstract The aromatic rice cultivars possess excellent aroma generally when grown in their favourable and specific environments. An early maturing selection from a Kerala aromatic local landrace with short grains, named ‘Biriyanicheera’, when grown in normal tropical conditions was sufficiently fragrant. The present study focused on the analysis of aroma in ‘Biriyanicheera’ rice genotype through molecular methods. The seeds of two aromatic rice varieties viz., Biriyanicheera and Gandhakasala (from Palakkad, Thrissur and Ernakulam districts) along with one non-aromatic rice variety Triveni (control) were used for the study. The BADH2 gene was amplified in all the three rice varieties. Upon sequencing the amplified PCR products of genomic DNA, the mutation in BADH2 gene was detected. The sequencing results of aromatic varieties revealed the presence of 8 base pair mutation in exon 7 in Biriyanicheera and Gandhakasala, whereas this mutation was absent in the non-aromatic variety Triveni. This indicated that aroma production in Biriyanicheera variety is due to similar mutation in BADH2 gene as that of the popular scented rice Basmati.

Drought remains the most important factor that affects rice productivity, especially in rainfed areas, worldwide. Upland rice is one of the crop choices of farmers in the rainfed environment. Although upland rice varieties require less water than lowland rice varieties, yields often remain limited by drought, particularly in the period of early growth. The aims of this study were to identify the traits related to early drought tolerance in upland rice varieties, and to identify the potential sources of germplasm for early drought tolerance. A total of sixty upland rice varieties were planted in a factorial experiment with a randomized complete block design with 3 replications in the rainy seasons of 2011 and 2012, under greenhouse conditions. Based on the drought tolerance index (DTI), the test germplasm sources were classified into three groups: (i) susceptible; (ii) moderately tolerant; (iii) tolerant to drought stress. Grain yield (GY) showed significant negative correlations with the leaf rolling score (r= − 0.623, P< 0.01), the leaf death score (LDS) (r= − 0.673, P< 0.01) and the recovery score (r= − 0.746, P< 0.01), while leaf dry matter (r= 0.698, P< 0.01) and leaf water potential (r= 0.618, P< 0.01) had significant positive correlations with GY. These findings indicate the suitability of the DTI as the selection criteria for early drought tolerance in a breeding programme. In addition, the upland rice germplasm accessions KKU-ULR011, KKU-ULR012, KKU-ULR125, KKU-ULR199 and KKU-ULR292 were identified as having high levels of stability for drought tolerance in both the 2011 and 2012 experiments, suggesting their potential for further use for rice variety improvement for drought tolerance.
