<p><strong>Objective: </strong>Total investigation of the macro-and micromorphological features of <em>Citrus maxima</em> (Burm.) Merrill. In addition, DNA fingerprinting in this study was to estimate genetic polymorphism and relationships with another citrus viz: <em>Citrus paradise</em><strong> </strong>and <em>Citrus sinensis</em> based on RAPD and ISSR markers.</p><p><strong>Methods: </strong>For the establishment of macro-and micromorphological investigation and genetic estimation, this study presents a total investigation of the macro-and micromorphological features of the roots, stems, leaves, flowers, fruits and seeds of the plants through microscopical investigation of the prepared entire, transverse sections and powdered forms of different organs under study. Furthermore, the DNA of the three plants was extracted from leaf samples and two types of DNA markers RAPD and ISSR were assayed.</p><p><strong>Results: </strong>Macro-and micromorphological features of different organs were investigated. On the other hand Genetic polymorphism and relationships among <em>Citrus maxima</em>, <em>Citrus paradisi</em> and <em>Citrus sinensis</em> was revealed. The highest genetic similarity was between <em>Citrus maxima </em>and <em>Citrus paradisi</em> while the lowest similarity was between <em>Citrus paradisi</em> and <em>Citrus sinensis</em> as revealed by RAPD, ISSR and combined data.</p><p><strong>Conclusion: </strong>For the present study, macro and micro-morphological characters, as well as, DNA fingerprinting can be considered as the identifying parameters for authentication of <em>Citrus maxima</em> (Burm.) Merrill.</p>

Grapevine cultivars diversity is vast and full of synonyms and homonyms. Up to few decades ago characterization of grapevine was based on morphological characters. In the last decades, molecular markers were developed and have been used as tools to study genetic diversity in a range of different plant species. Fifty-six Portuguese accessions representative of ‘Vinhos Verdes’ and ‘Douro’ Controlled Designations of Origin (DOC) were analysed through DNA fingerprints generated by Random Amplified Polymorphic DNA (RAPD) and Inter Simple Sequence Repeat (ISSR). The study aimed to compare the effectiveness of RAPD and ISSR molecular techniques in the detection of synonyms, homonyms and misnames. RAPD and ISSR analysis enabled the detection of 36 different band patterns, reducing in about 36% the initial material. Several accessions grown under different names, between and within collections, were confirmed as the same genotype, namely Gouveio/Verdelho, Sousão Douro/Vinhão and Arinto Oeste/Pedernã. Similarly, some homonyms/misnames were also identified, namely within Azal Tinto and Rabigato accessions. RAPD and ISSR markers revealed to be adequate molecular techniques for grapevine varieties fingerprinting with advantages over other molecular procedures, contributing for a good management of grapevine collections.

In this study, thirty one (31) morphologically distinct selections of saffron crop were used for molecular characterization. Molecular characterization was done through SSR, ISSR and RAPD markers. RAPD and ISSR markers showed significant variation; however, SSR markers did not reveal any variation between the selected clones. The Jaccard’s similarity coefficient ranged from 0.94 to 1.00 with an average of 0.98 among all 31 selections used. Minimum similarity value (0.94) was observed between CITH-S-107 and PAM-S-116 selections. The study provides sufficient knowledge to identify clones with better stigma characteristics for further crop improvement programs.
