The identification and characterization of new proteins involved in spermatogenesis is fundamental, considering that good-quality gametes are basic in ensuring proper reproduction. Here, we further analyzed the temporal and spatial localization during the first spermatogenic wave of rat testis of EHBP1L1, which is involved in vesicular trafficking due to the CH and bMERB domains, which bind to actin and Rab8/10, respectively. Western blot and immunofluorescence analyses showed that EHBP1L1 protein expression started at 21 days post-partum (dpp) concomitantly with the appearance of primary spermatocytes (I SPC). In subsequent stages, EHBP1L1 specifically localized together with actin in the perinuclear cytoplasm close to the acrosomal and Golgian regions of spermatids (SPT) during the different phases of acrosome biogenesis (AB). Moreover, it was completely absent in elongated SPT and in mature spermatozoa, suggesting that its role was completed in previous stages. The combined data, also supported by our previous report demonstrating that EHBP1L1 mRNA was expressed by primary (I) and secondary (II) SPC, lead us to hypothesize its specific role during AB. Although these results are suggestive, further studies are needed to better clarify the underlying molecular mechanisms of AB, with the aim to use EHBP1L1 as a potential new marker for spermatogenesis.

Abstract We report the result of the first search for multipoint in situ and imaging observations of interplanetary coronal mass ejections (ICMEs) starting with the first Solar Orbiter (SolO) data in 2020 April–2021 April. A data exploration analysis is performed including visualizations of the magnetic-field and plasma observations made by the five spacecraft SolO, BepiColombo, Parker Solar Probe (PSP), Wind, and STEREO-A, in connection with coronagraph and heliospheric imaging observations from STEREO-A/SECCHI and SOHO/LASCO. We identify ICME events that could be unambiguously followed with the STEREO-A heliospheric imagers during their interplanetary propagation to their impact at the aforementioned spacecraft and look for events where the same ICME is seen in situ by widely separated spacecraft. We highlight two events: (1) a small streamer blowout CME on 2020 June 23 observed with a triple lineup by PSP, BepiColombo and Wind, guided by imaging with STEREO-A, and (2) the first fast CME of solar cycle 25 (≈1600 km s−1) on 2020 November 29 observed in situ by PSP and STEREO-A. These results are useful for modeling the magnetic structure of ICMEs and the interplanetary evolution and global shape of their flux ropes and shocks, and for studying the propagation of solar energetic particles. The combined data from these missions are already turning out to be a treasure trove for space-weather research and are expected to become even more valuable with an increasing number of ICME events expected during the rise and maximum of solar cycle 25.

Cladribine tablets (CladT) preferentially reduce B and T lymphocyte levels. As aging is associated with a decline in immune function, the effect of CladT on lymphocyte levels may differ by age. This post hoc analysis combined data from the Phase 3 CLARITY, CLARITY Extension, and ORACLE-MS studies to examine the effect of age (≤50 or >50 years) on lymphopenia following CladT 3.5 mg/kg (CladT3.5; cumulative dose over 2 years) treatment over 96 weeks. Both CladT3.5 and placebo were given over Weeks 1 and 5 (Year 1 treatment) and Weeks 48 and 52 (Year 2 treatment) from the start of the studies. Absolute lymphocyte count (ALC) and levels of lymphocyte subsets were examined in 1564 patients (Age ≤50 [placebo: N=566; CladT3.5: N=813]; Age >50 [placebo: N=75; CladT3.5: N=110]). In both age groups, following CladT3.5 treatment, nadir for ALC occurred at Week 9 (8 weeks following start of Year 1 treatment) and Week 55 (7 weeks following start of Year 2 treatment) of the 96-week period; for CD19+ B lymphocytes, nadir occurred at Week 9 (Year 1) and Week 52 (Year 2). For CD4+ T lymphocytes, nadir occurred at Week 16 (Year 1) in both age groups, and at Weeks 60 and 72 (Year 2) in the Age ≤50 and >50 groups, respectively. Nadir for CD8+ T lymphocytes occurred at Week 16 (Year 1) and Week 72 (Year 2) in the Age ≤50 group and levels remained in the normal range; nadir occurred at Week 9 (Year 1) and Week 96 (Year 2) in the Age >50 group. Lymphocyte recovery began soon after nadir following CladT3.5 treatment and median levels reached normal range by end of the treatment year in both age groups. By Week 96, ~25% of patients treated with CladT3.5 reported ≥1 episode of Grade ≥3 lymphopenia (Gr≥3L). The rate of certain infections was numerically higher in older versus younger patients who experienced Gr≥3L. In conclusion, CladT3.5 had a similar effect on ALC and lymphocyte subsets in both younger and older patient groups.

Medical prediagnosis systems are now available online to give users quick and preliminary diagnosis information. The need for such a system has become particularly evident in areas with insufficient health professionals. Due to the privacy of patient medical information and the sensitivity of cloud diagnosis models, it is necessary to protect the security of data, models, and communications. These existing diagnosis systems can hardly provide a satisfied diagnosis accuracy while ensuring comprehensive security and high efficiency. In order to solve these problems, we proposed Relief- k minimum Wasserstein distance (Relief- k MW) classification method, which combined data encryption and BLS signature to form a privacy-preserving efficient online multiparty interactive medical prediagnostic scheme (OMPD). Theoretical analysis shows our OMPD effectively provides high-precision prediagnosis services. Extensive experimental results demonstrate that OMPD not only greatly improves the diagnostic accuracy but also reduces the computational and communication overhead.

The marine red algal order Halymeniales currently includes two families, the Halymeniaceae and Tsengiaceae, and consist of 38 genera and about 358 species. Phylogenetic analyses on specific taxa of the order are common, but not comprehensive, leaving the many intra-ordinal relationships within the Halymeniales unresolved. To reassess the phylogeny of the Halymeniales, we conducted extensive phylogenetic analyses based on 207 rbcL sequences and multigene analyses (rbcL, psaA, psbA, cox1, and LSU) using 47 taxa from the order. The combined data set fully supports the monophyly of the Grateloupia sensu lato clade. Phylogenetic assessment of the reproductive structures in the order using the type of auxiliary cell ampullae, pericarp origin, and tetrasporangial development characters, supports a Grateloupia sensu lato clade distinct from the Halymeniaceae exemplified by the generitype Halymenia. As a result, we propose to reinstate the family Grateloupiaceae Schmitz based on the Grateloupia sensu lato clade and including Grateloupia and eight other genera: Dermocorynus, Mariaramirezia, Neorubra, Pachymeniopsis, Kintokiocolax, Phyllymenia, Prionitis, and Yonagunia. The emended Grateloupiaceae is distinguished from the Halymeniaceae by the following three characteristics; (i) simple unbranched and unilateral type of auxiliary cell ampullae, (ii) pericarp formed densely by the fusion of secondary medullary filaments from subcortical cells and lateral ampullary filaments from a fusion cell complex, (iii) tetrasporangia originating laterally from the outer cortex. The Halymeniales comprises the monophyletic Grateloupiaceae, Halymeniaceae sensu lato (which requires further study), and the Tsengiaceae.

Spaciousness is an important quality of space which is affected by both the volume of space and the lighting environment. This study’s aim is to create the calculation model to quantify the effect of lighting environment on spaciousness. 3D luminance mapping, which is the combined data of luminance of surface and its distance to the observer, is used for the analysis of lighting environment, because we hypothesized that the three-dimensional localization of light affects the spaciousness. From the result of a subjective experiment to evaluate spaciousness with different lighting environment, it is revealed that spaciousness can be quantified from average luminance and “Dark-part-reduced visible volume” which is the reduced visible volume according to the distribution of dark pixels in the 3D luminance mapping. We also compared average illuminance of whole image and of ceiling, walls, floor, and concluded that the whole average illuminance best describes the effect.

AbstractRed pepper is enriched in antioxidant components, such as carotenoids, phenolic compounds, and vitamins. In this study, we investigated the natural variability in the content of carotenoids and phenolic acids in 11 red pepper cultivars grown in two locations in South Korea during 2016, 2017, and 2018. Seven carotenoids and six phenolic acids, including soluble and insoluble forms, were detected in the red fruit pericarps. The major carotenoids were β-carotene (40%) and capsanthin (20%). The content of insoluble phenolic acids was higher than that of soluble phenolic acids because of the large amount of insoluble p-coumaric acid. The statistical analysis of combined data showed significant differences among varieties, locations, and years for most of the measured components. The results from variance component analysis indicated that the effects of location, year and the interaction of location and year mainly accounted for the variation in carotenoids, whereas variations in phenolic acid content were attributed to year and variety. In addition, the results of principal component analysis and orthogonal partial least-squares discriminant showed that carotenoids were well discriminated by location and year, whereas phenolic acids were distinctively separated only by year. The data from this study could explain the natural variation in the content of carotenoids and phenolic acids in red pepper fruits by genotype and environment.

Abstract We use magnetic field measurements by the Juno spacecraft to catalog and investigate interplanetary coronal mass ejections (ICMEs) beyond 1 au. During its cruise phase, Juno spent about 5 yr in the solar wind between 2011 September and 2016 June, providing measurements of the interplanetary magnetic field (IMF) between 1 and 5.4 au. Juno therefore presents the most recent opportunity for a statistical analysis of ICME properties beyond 1 au since the Ulysses mission (1990–2009). Our catalog includes 80 such ICME events, 32 of which contain associated flux-rope-like structures. We find that the dependency of the mean magnetic field strength of the magnetic flux ropes decreases with heliocentric distance as r −1.24±0.43 between 1 and 5.4 au, in good agreement with previous relationships calculated using ICME catalogs at Ulysses. We combine the Juno catalog with the HELCATS catalog to create a data set of ICMEs covering 0.3–5.4 au. Using a linear regression model to fit the combined data set on a double-logarithmic plot, we find that there is a clear difference between global expansion rates for ICMEs observed at shorter heliocentric distances and those observed farther out beyond 1 au. The cataloged ICMEs at Juno present a good basis for future multispacecraft studies of ICME evolution between the inner heliosphere, 1 au, and beyond.
