Molecular diagnosis is based on target detection associated with many diseases and abnormalities. Common analytical techniques have disadvantages in the diagnosis. In contrast, biosensors are performed quick and selective detection of various markers. Surface plasmon resonance (SPR) biosensor allows qualitative and quantitative measurements without labelling and frequently used in medical applications. Recent developments in various emerging disciplines have allowed the design of novel bioreceptors for SPR biosensors. In recent years, the use of bioreceptors combined with nanotechnology and molecular imprinting technologies has increased in SPR instruments. Molecularly imprinted polymers (MIPs) are synthetic polymeric materials and provide specific and selective recognition sites for the target of interest. MIPs as bioreceptor show unique features compared to natural receptors. Therefore, MIP-based bioreceptors in SPR sensors have gained more attention recently. This chapter primarily focuses on the status and applications of nano-MIP-based SPR biosensors for the molecular diagnosis.

In the last decades,in vitrodiagnostic devices (IVDDs) became a very important tool in medicine for an early and correct diagnosis, a proper screening of targeted population, and also assessing the efficiency of a specific therapy. In this review, the most recent developments regarding different configurations of surface plasmon resonance affinity biosensors modified by using several nanostructured materials forin vitrodiagnostics are critically discussed. Both assembly and performances of the IVDDs tested in biological samples are reported and compared.

Surface plasmon resonance (SPR) and localized surface plasmon resonance (LSPR) are among the most common and powerful label-free refractive index-based biosensing techniques available nowadays. Focusing on LSPR sensors, their performance is highly dependent on the size, shape, and nature of the nanomaterial employed. Indeed, the tailoring of those parameters allows the development of LSPR sensors with a tunable wavelength range between the ultra-violet (UV) and near infra-red (NIR). Furthermore, dealing with LSPR along optical fiber technology, with their low attenuation coefficients at NIR, allow for the possibility to create ultra-sensitive and long-range sensing networks to be deployed in a variety of both biological and chemical sensors. This work provides a detailed review of the key science underpinning such systems as well as recent progress in the development of several LSPR-based biosensors in the NIR wavelengths, including an overview of the LSPR phenomena along recent developments in the field of nanomaterials and nanostructure development towards NIR sensing. The review ends with a consideration of key advances in terms of nanostructure characteristics for LSPR sensing and prospects for future research and advances in this field.

Two optical methods, namely surface plasmon resonance imaging and frustrated total internal reflection, are described in the paper in terms of comparing their sensitivity to change of refractive index of a thin boundary layer of an investigated medium. It is shown that, despite the fact that the theoretically calculated sensitivity is higher for the frustrated total internal reflection method, and the fact that usually in practice the surface plasmon resonance method, on the contrary, is considered more sensitive, under the same experimental conditions both methods show a similar result.
