Lipid profile of in vitro embryos produced from Bos indicus cows with low and high antral follicle counts

Two experiments in Nelore cows subjected to conventional protocol for timed AI (TAI) were designed to compare the influence of antral follicle count (AFC; 1) on the ovarian follicular dynamics and (2) conception rates. First, multiparous cows with high (G-high, ≥45 follicles; n = 43) and low AFC (G-low, ≤15 follicles; n = 32) were selected to receive a conventional oestradiol/progesterone-based TAI protocol for monitoring ovarian follicular dynamics. Second, AFC from 962 multiparous suckling cows (40 ± 15 days postpartum) subjected to a conventional protocol for TAI were classified as G-high (≥45 follicles; n = 194), G-intermediate (≥20 and ≤40 follicles, n = 397), and G-low AFC (≤ 15 follicles; n = 243). Pregnancy status was evaluated 30 to 35 days later. Data were analysed using ANOVA followed by the Tukey test and model of logistic regression (P ≤ 0.05). In the first study, the AFC were greater (P ≤ 0.05) in G-high than in G-low, but remained stable in both groups during the evaluations. The ovarian diameter, perimeter, and area were greater (P ≤ 0.05) in G-high than in G-low (28.3 ± 3.9 v. 20.5 ± 3.2 mm, 100.9 ± 13.2 v. 73.8 ± 13.9 mm, and 67.5 ± 16.4 v. 37.7 ± 11.7 mm2; respectively). Follicular growth rate, time of ovulation, corpus luteum diameter, and progesterone concentration were similar (P > 0.05) for cows with low or high AFC. Dominant follicles showed greater (P ≤ 0.05) diameter in G-low than G-high at D4 (7.3 ± 2.2 v. 6.2 ± 1.4 mm; P = 0.069, tendency), D8 (11.2 ± 1.8v. 9.5 ± 1.8 mm), D9 (12.3 ± 1.7 v. 10.6 ± 1.7 mm), and D10 (13.4 ± 1.3 v. 12.2 ± 1.8 mm), and the estimated diameter of the ovulatory follicle tended (P = 0.08) to be higher in G-low than in G-high (14.4 ± 1.5 v. 13.4 ± 2.1 mm). In the second study, AFC of G-low cows showed higher (P ≤ 0.05) conception rate (61.7%) than those of G-intermediate (52.9%) and G-high (49.5%) cows. In Nelore cows subjected to ovulation synchronization protocol, cows with high AFC showed higher ovarian measurements, but those with low AFC had large follicular diameters. Additionally, the cows with low AFC showed higher conception rate than groups with intermediate and high counts.

Embryo production may be enhanced when associated with cows selected on the basis of fertility markers, which should be easy to measure, such as antral follicle count (AFC) and genital tract morphometrics. The objective was to evaluate the effects of AFC class on oocyte 24-h outcome and in vitro embryo production in Bos indicus beef cows. Brahman (n = 151) cows (2-13 years old, 344-803 kg of BW, and 7-9 BCS). Low (LAFC), intermediate (IAFC), and high (HAFC) antral follicle classes were defined as follows: LAFC ≤ 30; IAFC 30-49; and HAFC ≥50 AFC. All follicles ≥3 mm in diameter were aspirated by conventional ovum pick-up technique. Only cumulus–oocyte complexes with at least 2 layers of granulosa cells and homogeneous cytoplasm were used for in vitro culture. They were matured in TCM-199 plus supplements for 24 h at 38.7°C in a 5% CO2 humidified atmosphere. After 24 h of maturation, a subset of oocytes (n = 319) was fixed and analysed under fluorescent microscopy and oocyte outcome was evaluated by classification, as follows: germinal vesicle (GV), germinal vesicle breakdown (GVBD), metaphase I/anaphase I/telophase I (MIAITI), and metaphase II (MII). The second subset of oocytes (n = 797) was fertilized in Ferti-TALP (10-15 oocytes per 60-µL drop) with frozen–thawed semen (18-22 h at 38.7°C in 5% CO2 after Percoll) from a single bull previously tested for good in vitro fertility. Presumptive zygotes were cultivated in CR2 medium for 48 h at 37.8°C in 5% CO2. For the remaining 96 h, embryos were transferred to 10% FCS-supplemented TCM-199 drops until the final evaluation. Data were analysed by the GENMOD, GLM, and CORR procedures of SAS (SAS Institute Inc., Cary, NC, USA). Viable oocytes, total embryos, and embryo production efficiency (viable oocyte/total embryos produced; P < 0.05) to AFC in various degrees (r2 = 0.87, 0.86, 0.30, respectively). The proportion of oocytes in GV, GVBD, MIAITI, and MII were different (P < 0.05) between LAFC, IAFC, and HAFC classes [GV: 12.3% (13/106)a, 3.1% (3/96)a and 4.3% (5/117)b, respectively]; [GVBD: 32.1% (34/106)a, 8.3% (8/96)a and 6.0 (7/117)b]; [MIAITI: 14.2% (15/106)a, 26.0% (25/96)b and 8.5% (10/117)c, respectively] and [MII: 41.5% (44/106)b, 62.5% (60/96)a and 81.2% (95/117)c, respectively). In conclusion, high AFC is positively related to better in vitro embryo fertility and to 24-h oocyte outcome after in vitro maturation.

This study aimed to evaluate the potential of Girolando (Bos taurus × Bos indicus) cows with high and low antral follicle counts (AFC) for the in vitro production of bovine embryos (IVEP), as well as the pregnancy rates of the recipients of these embryos. Girolando cows (Bos taurus × Bos indicus) were classified as high and low AFC when they had 35-52 (n = 13) and 11-17 follicles (n = 15), respectively. All animals were subjected to repeated follicular aspiration [Ovum pick-up (OPU)] and subsequent IVEP sessions. The synchronization protocol of the recipients was performed on a random day of the estrous cycle (Day 0) with the implantation of progesterone, estradiol benzoate, and prostaglandin. The high AFC group had higher aspirated oocyte/OPU (42.6 ± 5.2 vs. 14.6 ± 1.9; p < 0.01) and cultured oocyte/OPU (38.1 ± 6.6 vs. 12.3 ± 2.8; p < 0.01) averages as well as a higher blastocyst percentage on D7 (23.0 ± 1.0% vs. 18.4 ± 1.5%; p < 0.05) and higher pregnancy rate (42.7 ± 2.7% vs. 39.7 ± 4.6%; p < 0.05) than the low AFC group. Thus, we can conclude that animals with high AFC had better IVEP success rates than animals with low AFC.

The demand for calves as oocyte donors for in vitro embryo production is growing. However, Bos indicus have a late puberty, and some aspects of the reproductive physiology during the prepubertal period remain unclear. We characterized endocrine and morpho-functional reproductive features in Nelore calves (n=8) at 2- 5 (early prepubertal period, EPP) and from 8-11 months old (mo., intermediate prepubertal period, IPP). The calves’ ovaries and uterus were B-mode transrectal ultrasonography examined, and blood samples were collected every second week. The antral follicles number and size, and ovarian and uterine horn diameters, were recorded, and plasma FSH and LH concentrations were measured (RIA). Non-pregnant, non-lactating cyclic Nelore cows (n=8) were used as controls for endocrine endpoints. Somatic development was monitored by monthly weighing, and 3D scanning of the rump area. The somatic and endocrine endpoints were compared within and between EPP and IPP, and between each period and control cows. Associations were determined by the Spearman correlation method, and the developmental rates were determined by non-linear regression. All morphological endpoints, except antral follicle count, increased (P < 0.001) from the EPP to the IPP. However, within each period differences occurred only at EPP. During the EPP LH and FSH plasma concentrations were similar (P > 0.05), whereas during the IPP LH was lower (P < 0.05) and FSH was higher (P < 0.001) than control cows. The EPP calves showed moderate to high positive correlations among ovarian, uterine, and somatic endpoints. Conversely, the IPP such correlations were mostly weak. In summary, distinct ovarian activity and development patterns of primary and secondary sexual characteristics occurred in Nelore calves at 2- 5 mo compared to 8-11 mo.

Aim: This study aimed to evaluate the antidiabetic and antihyperlipidemic effects of Asteriscus graveolens. Background: Asteriscus graveolens (Asteraceae) is a medicinal plant widely used by the Moroccan population to treat various diseases including diabetes. Objective: This work aimed to assess the capacity of flavonoids extracted from Asteriscus graveolens (FEE) to improve diabetes mellitus and dyslipidemia in normal and STZ-induced diabetic rats. Methods: Flavonoids were extracted from A. graveolens using the Soxhlet apparatus and using different organic solvents. Normal and streptozotocin-induced diabetic rats were treated orally by the extract of A. graveolens at a dose of 10 mg/kg. The oral treatment during 15 days was used to evaluate the effect of the flavonoids extracted from A. graveolens on blood glucose level and lipid profile in normal and diabetic rats. The oral glucose tolerance test as well as the analysis of histopathological examination of liver was performed. The antioxidant activity of FEE was also assessed by the method of trapping of free radical 2,2-diphenyl-1 picrylhydrazyl (DPPH), in order to estimate the mechanisms of action involved by FEE to improve hyperglycemia and lipid profile in normal and diabetic rats. Results: FEE reduced serum glucose concentrations in both normal and diabetic rats and exhibited in the last group lowering total cholesterol and triglycerides effects as well as improvement of the HDL-cholesterol serum level. In addition, a remarkable influence on glucose tolerance was also noticed after FEE treatment. Moreover, FEE was able to improve histopathological status of liver and possess a potential antioxidant effect in vitro. Conclusion: In conclusion, this study demonstrates the hypoglycemic and antihyperlipidemic effects of FEE in rats supporting then its traditional use for the management of diabetes.
