In vitro and in vivo acaricide action of juvenoid analogs produced from the chemical modification of Cymbopogon spp. and Corymbia citriodora essential oil on the cattle tick Rhipicephalus (Boophilus) microplus

Essential oils from species of the genus Hyptis are well-known for their significant biological properties, including antimicrobial and acaricidal activities. The essential oil from the aerial parts of H. crenata was obtained by hydrodistillation; borneol (17.8%), 1,8-cineol (15.6%) and p-cimene (7.9%) were characterized by GC-MS as its major constituents. The essential oil was evaluated in vitro for its antimicrobial activities against six fungal and five bacterial strains, by measuring the respective MICs, MFCs and MBCs, using broth microdilution methods. The strongest bactericidal activities were shown against Staphylococcus aureus and Enterococcus faecalis, while the strongest fungicidal activities were against Cryptococcus neoformans, Candida glabrata and Candida tropicalis. The oil was also assessed for its anti-tick properties and, at a concentration of 2.5%, it significantly inhibited in vivo oviposition of engorged females of the cattle tick Rhipicephalus (Boophilus) microplus, using the adult immersion test., with an effectiveness of 94.4%.

SummaryCircumstantial evidence suggests that the earlier detachment of Boophilus microplus larvae from highly resistant cattle follows the release of histamine at the attachment site. In vivo and in vitro experiments show that a proportion of the larvae will detach following injection or infusion of histamine. Other mediators such as bradykinin, prostaglandin E2, 5-hydroxytryptamine and dopamine have little or no effect on tick behaviour in vivo. Sensitivity to histamine declines as larval attachments stabilize, and repeated injections have no effect on the weight of larvae after 3 days on the host. Response to histamine is discussed in relation to host resistance, histology of the feeding lesion and larval behaviour.

B. micro plus larvae of the organophosphorus-resistant Biarra and Mt Alford strains were respectively 8 x and 35 x resistant to bromophos-ethyl compared with larvae of the standard organophosphorussusceptible Yeerongpilly strain. Both resistant strains had acetylcholinesterase with decreased sensitivity to inhibition by the oxon of bromophos-ethyl in vitro; this was the only resistance mechanism apparent in the Biarra strain but Mt Alford larvae were protected additionally by increased metabolism of the oxon in vivo to water-soluble products. Total degradation rates for the parent chemical were similar in all strains and relatively slow. Both bromophos-ethyl and its oxon were potent inhibitors of mixed-function oxidase in vivo and it seemed that the slow oxidative metabolism of bromophos-ethyl (the major pathway) could be attributed to substrate and/or product inhibition. No phenolic metabolites were detected and the major water-soluble metabolite was identified electrophoretically as 2,5-dichlorophenyl-o-ethyl phosphate. Some debrominated oxon was detected in all samples of larvae after dosage with bromophos-ethyl or with its oxon, indicating that oxidation of bromophos-ethyl to the oxon, debromination of the oxon followed by deethylation was a major degradative sequence.

Esta pesquisa foi conduzida com o objetivo de avaliar o efeito in vitro e in vivo do óleo de eucalipto (Corymbia citriodora) sobre o carrapato bovino (Rhipicephalus (Boophilus) microplus). Na experimentação in vitro, foi utilizado o grupo controle negativo e oito concentrações de óleo de eucalipto (0,5; 1; 2; 5; 10; 20; 50; 100%), em fêmeas ingurgitadas de carrapato. A eficácia de controle foi de 0; 30,5; 75,5; 91; 100; 100; 100; 100 e 100%, respectivamente. Para a experimentação in vivo, foram constituídos três grupos (controle negativo; óleo de eucalipto a 3,5% - nível estimado mediante análise de regressão, correspondendo a 95% de eficácia de controle do carrapato da pesquisa in vitro e amitraz a 0,025%), com dezoito vacas da raça Holandesa. Antes (média dos dias -3, -2,-1) e após a aplicação do produto (1, 2, 3, 5, 7, 10, 14, 21dias), foram contadas fêmeas ingurgitadas de carrapato. A eficácia de controle foi de 0; 96,4 e 69%, respectivamente, 21 dias após o tratamento. Na 1ª e na 2ª ordenha após a aplicação dos tratamentos, foram avaliadas variáveis fisiológicas e coletadas amostras de leite para avaliar as propriedades organolépticas no leite e no iogurte (controle negativo x tratamento fitoterápico). O teste de aceitação sensorial do leite e das variáveis fisiológicas avaliadas foram similares entre os tratamentos.

Abstract Background Silexan, a special essential oil from flowering tops of lavandula angustifolia, is used to treat subsyndromal anxiety disorders. In a recent clinical trial, Silexan also showed antidepressant effects in patients suffering from mixed anxiety-depression (ICD-10 F41.2). Since preclinical data explaining antidepressant properties of Silexan are missing, we decided to investigate if Silexan also shows antidepressant-like effects in vitro as well as in vivo models. Methods We used the forced swimming test (FST) in rats as a simple behavioral test indicative of antidepressant activity in vivo. As environmental events and other risk factors contribute to depression through converging molecular and cellular mechanisms that disrupt neuronal function and morphology—resulting in dysfunction of the circuitry that is essential for mood regulation and cognitive function—we investigated the neurotrophic properties of Silexan in neuronal cell lines and primary hippocampal neurons. Results The antidepressant activity of Silexan (30 mg/kg BW) in the FST was comparable to the tricyclic antidepressant imipramine (20 mg/kg BW) after 9-day treatment. Silexan triggered neurite outgrowth and synaptogenesis in 2 different neuronal cell models and led to a significant increase in synaptogenesis in primary hippocampal neurons. Silexan led to a significant phosphorylation of protein kinase A and subsequent CREB phosphorylation. Conclusion Taken together, Silexan demonstrates antidepressant-like effects in cellular as well as animal models for antidepressant activity. Therefore, our data provides preclinical evidence for the clinical antidepressant effects of Silexan in patients with mixed depression and anxiety.

ABSTRACT: Rhipicephalus (Boophilus) microplus, known as the cattle tick, is a cause of great economic losses for dairy cattle farming because of its high frequency of occurrence and the difficulty in controlling it. The aim of this study was to evaluate the in vitro activity of Chenopodium ambrosioides extract on R. (B.) microplus. For this purpose, 125 females were selected and classified into five groups according to their weight, in order to ensure that the females used presented homogeneous weight. The treatments comprised 40 and 60% extracts of Chenopodium ambrosioides, distilled water, ethanol (70ºGL) and 12.5% amitraz. The extracts of C. ambrosioides (40 and 60%) showed effectiveness of 99.7 and 100% and higher percentages of dead females than the other treatments: 64 and 96%, respectively (p<0.001). In the groups exposed to distilled water and ethanol (70º GL), 92 and 88% of the females maintained oviposition. In the females exposed to 40 and 60% extracts, oviposition of 36 and 4% occurred, respectively. It was concluded that the extract of Chenopodium ambrosioides, at both concentration evaluated, had high efficiency against engorged females of cattle ticks.
