Effects of repeated asenapine in a battery of tests for anxiety-like behaviours in mice

ObjectiveA number of atypical antipsychotic drugs were demonstrated to have anxiolytic effects in patients and in animal models. These effects were mostly suggested to be the consequence of the drugs’ affinity to the serotonin system and its receptors. Asenapine is a relatively new atypical antipsychotic that is prescribed for schizophrenia and for bipolar mania. Asenapine has a broad pharmacological profile with significant effects on serotonergic receptors, hence it is reasonable to expect that asenapine may have some anxiolytic effects. The present study was therefore designed to examine possible effects of asenapine on anxiety-like behaviour of mice.MethodMale ICR mice were repeatedly treated with 0.1 or 0.3 mg/kg injections of asenapine and then tested in a battery of behavioural tests related to anxiety including the open-field test, elevated plus-maze (EPM), defensive marble burying and hyponeophagia tests. In an adjunct experiment, we tested the effects of acute diazepam in the same test battery.ResultsThe results show that diazepam reduced anxiety-like behaviour in the EPM, the defensive marble burying test and the hyponeophagia test but not in the open field. Asenapine has anxiolytic-like effects in the EPM and the defensive marble burying tests but had no effects in the open-field or the hyponeophagia tests. Asenapine had no effects on locomotor activity.ConclusionThe results suggest that asenapine may have anxiolytic-like properties and recommends that clinical trials examining such effects should be performed.

Human amniotic fluid and a mixture of eight fatty acids (FAT-M) identified in this maternal fluid (C12:0, lauric acid, 0.9 μg%; C14:0, myristic acid, 6.9 μg%; C16:0, palmitic acid, 35.3 μg%; C16:1, palmitoleic acid, 16.4 μg%; C18:0, stearic acid, 8.5 μg%; C18:1cis, oleic acid, 18.4 μg%; C18:1trans, elaidic acid, 3.5 μg%; C18:2, linoleic acid, 10.1 μg%) produce anxiolytic-like effects that are comparable to diazepam in Wistar rats, suggesting the involvement ofγ-aminobutyric acid-A (GABAA) receptors, a possibility not yet explored. Wistar rats were subjected to the defensive burying test, elevated plus maze, and open field test. In different groups, threeGABAAreceptor antagonists were administered 30 min before FAT-M administration, including the competitive GABA binding antagonist bicuculline (1 mg/kg),GABAAbenzodiazepine antagonist flumazenil (5 mg/kg), and noncompetitiveGABAAchloride channel antagonist picrotoxin (1 mg/kg). The FAT-M exerted anxiolytic-like effects in the defensive burying test and elevated plus maze, without affecting locomotor activity in the open field test. TheGABAAantagonists alone did not produce significant changes in the behavioral tests. Picrotoxin but not bicuculline or flumazenil blocked the anxiolytic-like effect of the FAT-M. Based on the specific blocking action of picrotoxin on the effects of the FAT-M, we conclude that the FAT-M exerted its anxiolytic-like effects throughGABAAreceptor chloride channels.

Introduction: Lack of high-quality sleep causes serious side effects like anxiety and changes in plasma concentration of oxalate. The current study aimed to investigate the impact of local extremely low frequency magnetic fields (ELF-MFs) on inducing sleep (sleepiness) and anxiety in male rats. Methods: In this experimental study, 40 male rats were allocated in four groups (n=10). The ELF-MFs exposure (0, 10 and 18 Hz) was applied with intensity 200µT for three days (10 min/day). Sham-treated animal did not receive ELF-MF. Serum level of oxalic acid (OA) and sleepiness were measured both before first and after last exposure to ELF-MF or sham. Anxiety, sleepiness and OA were measured by using elevated plus maze, open-field test (OFT) and ELISA test, respectively. Results: Comparison of oxalate levels between before and after exposure to ELF-MF revealed that ELF-MF (10 Hz) decreased the serum level of oxalate (p<0.05). Comparison of the percent of open:closed arm entry (in elevated plus maze) between before and after exposure to ELF-MF revealed significant differences. Also, frequency, velocity and distance moved were decreased in the open-field test. Conclusion: Results of the present study demonstrated that ELF-MF with short time exposure may modulate the metabolism of OA and may modulate anxiety-like behavior or kind of induction of sleepiness in male rats.

Aim: We conducted this study with the aim to investigate the effect of vitamin D3 on spatial learning and memory in healthy young albino rat. Study Design: Experimental evaluation. Place and Duration of Study: Department of Pharmacology, Smt. Kashibai Navale Medical College and General Hospital (SKNMCGH), Pune, between October 2019 to February 2020. Methodology: All the pharmacological experiments were conducted using Wistar albino rats (n=6), weighing between 100 g – 150 g. Total 18 animals (9 male and 9 female) were screened and marked into 3 different groups (6 in each group) Control (Normal saline 10 ml/kg), Vitamin D (Cholecalciferol 1000 IU/kg) and standard (Piracetam 200 mg/kg). Drugs were administered per os for 21 days. Elevated Plus Maze (Transfer Latency), Open Field Test (Rearing, Locomotion), Radial Arm Maze (Working and Reference memory) were used as amnesic models and their parameters for evaluation of this study. Results: After 21 days of treatment among all the three groups, Transfer Latency (p=9.55) in elevated plus maze, Working memory (p=0.454) and Reference memory (p=0.929) observed in radial 8 arm maze were non significant. In open field apparatus pellets count was significant (0.010), rest all parameters were non significant. Conclusion: The result of study suggests that no significant beneficial effect of Vitamin D was seen on various learning models as assessed by Elevated Plus Maze, Radial Arm Maze, Open Field Test.

Decrease in natural illumination in fall/winter months causes depressive-like seasonal affective disorders in vulnerable individuals. Obesity is another risk factor of depression. The lethal yellow (AY) mutation causes ectopic expression of agouti protein in the brain. Mice heterozygous for AY mutation (AY/a) are obese compared to their wild-type littermates (a/a). The main aims of the study were to investigate the effects of AY mutation, photoperiod and the interaction between these factors on daily activity dynamics, feeding, locomotor and exploratory activities, anxiety-related and depressive-like behaviors in mild stress condition. Six weeks old mouse males of AY/a and a/a lines were divided into four groups eight animals each and exposed to long- (14 h light and 10 h darkness) or short- (4 h light and 20 h darkness) day conditions for 28 days. Then the behavior of these mice was successively investigated in the home cage, open field, elevated plus-maze and forced swim tests. We did not observed any effect of AY mutation on the general activity, water and food consumption in the home cage; locomotion and exploration in the open field test; anxiety-related behavior in the open field and elevated plus-maze tests. At the same time, AY mutation increased depressive-like immobility time in the forced swim test (F1.28 = 20.03, p = 0.00012). Shortday conditions decreased nocturnal activity in the home cage, as well as locomotion (F1.28 = 16.33, p = 0.0004) and exploration (F1.28 = 16.24, p < 0.0004) in the open field test. Moreover, short-day exposition decreased time spent in the center of the open field (F1.28 = 6.57, p = 0.016) and in the open arms of the elevated plus-maze (F1.28 = 12.08, p = 0.0017) tests and increased immobility time in the forced swim test (F1.28 = 9.95, p = 0.0038). However, no effect of the interaction between AY mutation and photoperiod on immobility time in the forced swim test was observed. Therefore, short-day photoperiod and AY mutation increased depressive-like behavior in the forced swim test by means of different mechanisms.

Ageing is a genetically programmed physiological process that is modulated by numerous environmental factors, associated with decreasing physiological function, decreasing reproductive rate and increasing age-related mortality rate. Maintaining mobility performance and physical function in the elderly is the main objective of the successful ageing concept. In this study, we aimed to evaluate the beneficial effect of a novel nutraceutical formulation containing Centella asiatica L. extract, vitamin C, zinc and vitamin D3 (as cholecalciferol) on motor activity and anxiety with the use of a murine model of old animals, as a means of providing proof for clinical use in the elderly, for enhancing physical strength and improving life quality. Eighteen Sprague Dawley 18 months old male rats were divided into three groups and received corn oil (the control group) or 1 capsule/kg bw Reverse supplement (treatment group 1) or 2 capsules/kg bw Reverse supplement (treatment group 2), for a period of 3 months. The Reverse supplement (Natural Doctor S.A, Athens, Greece) contains 9 mg Centella asiatica L. extract, vitamin C (200 mg as magnesium ascorbate), zinc (5 mg as zinc citrate), vitamin D3 (50 µg as cholecalciferol) per capsule. Before and after the treatment, the motor function and behavioral changes for anxiety and depression were evaluated using the open-field test, elevated plus-maze test and rotarod test. The supplementation with Reverse (Natural Doctor S.A) supplement can improve the locomotor activity in old rats in a dose-dependent manner, as demonstrated by an increase in the latency to leave from the middle square, in the number of rearings in the open field test, in the time spent in the open arms and time spent in the center in the elevated plus-maze test and the latency to all in all three consecutive trials in the rotarod test. Stress also decreased significantly in a dose-dependent manner, following the treatment with Reverse supplement, as was demonstrated by the decrease in the number of groomings at the open field test and time spent in the dark and the number of groomings at the elevated plus-maze test.

Background Anxiety disorders are the most common neuropathologies worldwide, but the precise neuronal mechanisms that underlie these disorders remain unknown. The hippocampus plays a role in mediating anxiety-related responses, which can be modeled in rodents using behavioral assays, such as the elevated plus maze. Yet, the molecular markers that underlie affect-related behavior on the elevated plus maze are not well understood. Methods We used herpes simplex virus vector delivery to overexpress extracellular signal-regulated kinase-2, a signaling molecule known to be involved in depression and anxiety, within the dorsal hippocampus of adult Sprague-Dawley male rats. Three days post virus delivery, we assessed anxiety-like responses on the elevated plus maze or general locomotor activity on the open field test. Results When compared to controls, rats overexpressing extracellular signal-regulated kinase-2 in the dorsal hippocampus displayed an anxiolytic-like phenotype, per increases in time spent in the open arms, and less time in the closed arms, of the elevated plus maze. Furthermore, no changes in locomotor activity as a function of virus infusion were observed on the open field test between the experimental groups. Conclusion This investigation demonstrates that virus-mediated increases of extracellular signal-regulated kinase-2 signaling, within the hippocampus, plays a critical role in decreasing anxiogenic responses on the rat elevated plus maze. As such, our data provide construct validity, at least in part, to the molecular mechanisms that mediate anxiolytic-like behavior in rodent models for the study of anxiety.

Purpose. Our previously data on orexigenic peptides (orexin, ghrelin) showed antagonists of peptides receptors as correctors of the emotional-motivational and cognitive spheres. Currently, a close relationship between ghrelin and orexin with neuropetide Y has been shown in feeding and emotional behavior. The aim of this work was to analyze the effect of the NPY antagonist Y1R BMS 193885 on emotional and intraspecies behavior, as well as on the reinforcing properties of ethanol in rats. Methods. We used the open field test, elevated plus-maze, Porsolts forced swimming test, resident intruder test, conditional place preference (CPP). BMS 193885 1 mg/ml, 20 l intranasally did not cause an anxiogenic effect in the elevated plus-maze. Results. In the Porsolts test, there was also no increase in the level of depression. Moreover, there was a significant decrease in the number and time of dives, as an indirect indicator of a decrease in the level of depression. At the same time, in the resident intruder test were decreased protective behavior, as an indicator of a decrease in the stress of intraspecific interaction in the absence of aggression. Moreover, local movements were increased in the open field test as an indicator of the animals activity impaired by fear. BMS 193885 had no effect on the expression of the CPP of ethanol. Conclusion. Thus, it was previously shown that the BMS 193885 is a powerful, selective, brain-penetrating Y1 receptor antagonist, it reduces food intake and body weight in animal models of obesity both after acute and chronic administration. Our data indicate that the decrease in food intake is not associated with the level of anxiety, depression, or with a change in intraspecific interaction. It has been previously shown that NPY reduces alcohol consumption. Our data indicate that the Y1R antagonist of the neuropeptide Y BMS 193885 does not cause a change in the CPP of alcohol.
