O objetivo deste trabalho foi selecionar entre 300 estirpes de Bacillus thuringiensis as efetivas simultaneamente contra larvas de Spodoptera frugiperda J.E. Smith e Anticarsia gemmatalis Hübner (Lepidoptera: Noctuidae), Anthonomus grandis Boheman (Coleoptera: Curculionidae), Aedes aegypti Linnaeus e Culex quinquefasciatus Say (Diptera: Culicidae). Foram selecionadas duas estirpes de B. thuringiensis, denominadas S234 e S997, que apresentaram atividade contra as três ordens de insetos. As estirpes foram caracterizadas por métodos morfológicos, bioquímicos e moleculares. As mesmas apresentaram duas proteínas principais de 130 e 65 kDa, produtos de reação em cadeia da polimerase de tamanho esperado para a detecção dos genes cry1Aa, cry1Ab, cry1Ac, cry1B e cry2 e cristais bipiramidais, cubóides e esféricos.

ABSTRACT Both Bacillus sphaericus and Bacillus thuringiensis subsp. israelensis produce mosquitocidal toxins during sporulation and are extensively used in the field for control of mosquito populations. All the known toxins of the latter organism are known to be encoded on a large plasmid, pBtoxis. In an attempt to combine the best properties of the two bacteria, an erythromycin resistance-marked pBtoxis plasmid was transferred to B. sphaericus by a mating technique. The resulting transconjugant bacteria were significantly more toxic to Aedes aegypti mosquitoes and were able to overcome resistance to B. sphaericus in a resistant colony of Culex quinquefasciatus, apparently due to the production of Cry11A but not Cry4A or Cry4B. The stability of the plasmid in the B. sphaericus host was moderate during vegetative growth, but segregational instability was observed, which led to substantial rates of plasmid loss during sporulation.

Tujuan dari penelitian ini adalah untuk mengisolasi Bacillus thuringiensis entomopathogenik yang mampu membunuh larva nyamuk Culex quinquefasciatus dan Aedes aegypti dari berbagai habitat di Kota Mataram, Nusa Tenggara Barat. Bakteri diisolasi dari tanah yang diambil dari 7 habitat yang berbeda di sekitar Kota Mataram. Isolat yang didapat kemudian diuji karakter biokimia dan fisiologisnya. Uji toksisitas selektif dilakukan pada larva C. quinquefasciatus dan A. aegypti instar 3%. Dari 7 jenis habitat yang dieksplorasi didapatkan 20 biakan yang teridentifikasi sebagai B. thuringiensis berdasarkan karakteristik morfologi, biokimia dan fisiologinya. Seluruh biakan B. thuringiensis yang diisolasi dari seluruh habitat menunjukkan tingkat toksisitas yang bervariasi. Toksisitas tertinggi terhadap larva C. quinquefasciatus sebesar 15% didapatkan pada biakan B. thuringiensis yang diisolasi dari habitat kebun yang ternaung. Toksisitas tertinggi terhadap larva A. aegypti didapatkan sebesar 23,35% pada B. thuringiensis yang diisolasi dari habitat kolam.

ABSTRACT The interaction of two cytolytic toxins, Cyt1Ab fromBacillus thuringiensis subsp. medellinand Cyt2Ba from Bacillus thuringiensis subsp.israelensis, with Bacillus sphaericus was evaluated against susceptible and resistant Culex quinquefasciatus and the nonsensitive species Aedes aegypti. Mixtures of B. sphaericus with either cytolytic toxin were synergistic, and B. sphaericusresistance in C. quinquefasciatus was suppressed from >17,000- to 2-fold with a 3:1 mixture of B. sphaericusand Cyt1Ab. This trait may prove useful for combating insecticide resistance and for improving the activity of microbial insecticides.

The objective of this work was to evaluate 210 Bacillus thuringiensis strains against Aedes aegypti and Culex quinquefasciatus larvae to select the most effective. These strains were isolated from different regions of Brazil and are stored in a Bacillus spp. collection at Embrapa Recursos Genéticos e Biotecnologia, Brasília, Brazil. The selected strains were characterized by morphological (microscopy), biochemical (SDS-PAGE 10%) and molecular (PCR) methods. Six B. thuringiensis strains were identified as mosquito-toxic after the selective bioassays. None of the strains produced the expected PCR products for detection of cry4, cry11 and cyt1A genes. These results indicate that the activity of mosquitocidal Brazilian strains are not related with Cry4, Cry11 or Cyt proteins, so they could be used as an alternative bioinsecticide against mosquitoes.
