Effects of larval diet on the life-history traits and phenotypic expression of pyrethroid resistance in the major malaria vector Anopheles gambiae s.s.

The success achieved in reducing malaria transmission by vector control is threatened by insecticide resistance. To strengthen the current vector control programmes, the non-genetic factors underlying the emergence of insecticide resistance in Anopheles vectors and its widespread need to be explored. This study aimed to assess the effects of larval diet on some life-history traits and pyrethroid-insecticide susceptibility of Anopheles gambiae s.s. Three (3) An. gambiae strains, namely Kisumu (insecticide susceptible), AcerKis (homozygous ace-1 R G119S resistant) and KisKdr (homozygous kdr R L1014F resistant) were fed with three different diets (low, medium, and high) of TetraMin ® Baby fish food. Pre-imaginal developmental time, larval mortality, adult emergence rate and female wing length were measured. Mosquito females were exposed to insecticide-treated net (ITN) PermaNet 2.0 and PermaNet 3.0. In the three An. gambiae strains, significant differences in adult emergence rates ( F = 1054.2; df = 2; p <0.01), mosquito wing length ( F = 970.5; df = 2; p <0.01) and adult survival post insecticide exposure ( χ2 = 173; df = 2; p <0.01), were noticed among the three larval diets. Larvae fed with the low food diets took more time to develop, were smaller at emergence and displayed a short lifespan, while the specimens fed with a high regime developed faster and into big adults. Although being fed with a high diet, none of An. gambiae strain harbouring the kdr R and ace-1 R allele survived 24 hours after exposure against PermaNet 3.0. This study showed that variation in the larval diet significantly impacts An. gambiae life-history traits such as larval mortality and developmental time, adult wing length, and female susceptibility to pyrethroid insecticides. Further investigations through field-based studies would allow an in-depth understanding of the implications of these non-genetic parameters on the physiological traits of malaria vectors and consequently improve resistance management.

A scaffold-level genome assembly of a minute pirate bug, Orius laevigatus (Hemiptera: Anthocoridae), and a comparative analysis of insecticide resistance-related gene families with hemipteran crop pests

Background Orius laevigatus, a minute pirate bug, is a highly effective beneficial predator of crop pests including aphids, spider mites and thrips in integrated pest management (IPM) programmes. No genomic information is currently available for O. laevigatus, as is the case for the majority of beneficial predators which feed on crop pests. In contrast, genomic information for crop pests is far more readily available. The lack of publicly available genomes for beneficial predators to date has limited our ability to perform comparative analyses of genes encoding potential insecticide resistance mechanisms between crop pests and their predators. These mechanisms include several gene/protein families including cytochrome P450s (P450s), ATP binding cassette transporters (ABCs), glutathione S-transferases (GSTs), UDP-glucosyltransferases (UGTs) and carboxyl/cholinesterases (CCEs). Methods and findings In this study, a high-quality scaffold level de novo genome assembly for O. laevigatus has been generated using a hybrid approach with PacBio long-read and Illumina short-read data. The final assembly achieved a scaffold N50 of 125,649 bp and a total genome size of 150.98 Mb. The genome assembly achieved a level of completeness of 93.6% using a set of 1658 core insect genes present as full-length genes. Genome annotation identified 15,102 protein-coding genes - 87% of which were assigned a putative function. Comparative analyses revealed gene expansions of sigma class GSTs and CYP3 P450s. Conversely the UGT gene family showed limited expansion. Differences were seen in the distributions of resistance-associated gene families at the subfamily level between O. laevigatus and some of its targeted crop pests. A target site mutation in ryanodine receptors (I4790M, PxRyR) which has strong links to diamide resistance in crop pests and had previously only been identified in lepidopteran species was found to also be present in hemipteran species, including O. laevigatus. Conclusion and significance This assembly is the first published genome for the Anthocoridae family and will serve as a useful resource for further research into target-site selectivity issues and potential resistance mechanisms in beneficial predators. Furthermore, the expansion of gene families often linked to insecticide resistance may be an indicator of the capacity of this predator to detoxify selective insecticides. These findings could be exploited by targeted pesticide screens and functional studies to increase effectiveness of IPM strategies, which aim to increase crop yields by sustainably, environmentally-friendly and effectively control pests without impacting beneficial predator populations.

Determinants of Insecticide Resistance Evolution: Comparative Analysis Among Heliothines

It is increasingly clear that pest species vary widely in their propensities to develop insecticide resistance. This review uses a comparative approach to analyze the key pest management practices and ecological and biochemical or genetic characteristics of the target that contribute to this variation. We focus on six heliothine species, three of which, Helicoverpa armigera, Heliothis virescens, and Helicoverpa zea, have developed resistances to many pesticide classes. The three others, Helicoverpa punctigera, Helicoverpa assulta, and Helicoverpa gelotopoeon, also significant pests, have developed resistance to very few pesticide classes. We find that host range and movement between alternate hosts are key ecological traits that influence effective selection intensities for resistance. Operational issues are also critical; area-wide, cross-pesticide management practices that account for these ecological factors are key to reducing selection intensity. Without such management, treatment using broad-spectrum chemicals serves to multiply the effects of host plant preference, preadaptive detoxification ability, and high genetic diversity to create a pesticide treadmill for the three high-propensity species.Without rigorous ongoing management, such a treadmill could still develop for newer, more selective chemistries and insecticidal transgenic crops.

Monitoring of Insecticide Resistance in Anopheles culicifacies in Twelve Districts of Madhya Pradesh, Central India (2017–2019)

Background. Indoor residual spraying (IRS) and long-lasting insecticidal nets (LLINs) are malaria vector control measures used in India, but the development of insecticide resistance poses major impediments for effective vector control strategies. As per the guidelines of the National Vector Borne Disease Control Programme (NVBDCP), the study was conducted in 12 districts of Madhya Pradesh to generate data on insecticide resistance in malaria vectors. Methods. The susceptibility tests were conducted on adult An. culicifacies as per the WHO standard technique with wild-caught mosquitoes. The blood-fed female mosquitoes were exposed in 3 to 4 replicates on each occasion to the impregnated papers with specified discriminating dosages of the insecticides (DDT: 4%, malathion: 5%, deltamethrin: 0.05%, and alphacypermethrin: 0.05%), for one hour, and mortality was recorded after 24-hour holding. Results. An. culicifacies was found resistant to DDT 4% in all the 12 districts and malathion in 11 districts. The resistance to alphacypermethrin was also observed in two districts, and possible resistance was found to alphacypermethrin in seven districts and to deltamethrin in eight districts, while the vector was found susceptible to both deltamethrin and alphacypermethrin in only 3 districts. Conclusion. An. culicifacies is resistant to DDT and malathion and has emerging resistance to pyrethroids, alphacypermethrin, and deltamethrin. Therefore, regular monitoring of insecticide susceptibility in malaria vectors is needed for implementing effective vector management strategies. However, studies to verify the impact of IRS with good coverage on the transmission of disease are required before deciding on the change of insecticide in conjunction with epidemiological data.

Evidences for newer insecticides and growth regulators resistance in field populations of Helicoverpa armigera (Lepidoptera: Noctuidae) in Karnataka, India

The prominence of Helicoverpaarmigera (Lepidoptera: Noctuidae) averse insecticide resistance was traversed in the course of 2017 in Karnataka, India. The results divulged typical resistance level prostrating in selected newer insecticides, even though exiguous higher resistance airing in insect growth regulator Novaluranwith LC 50 of and 13.02 to 18.07 ppm and 1.17 to 1.95 folds resistance) compared to newer group insecticides Spinasad, Indoxacarb, Flubendiamide, and Rynaxypyr (3.57 to 10.19 ppm, 1.01 to 1.27 fold). Raichur and Kalaburgi strains comprehend higher resistance to Novaluran and newer insecticides with exception of Flubendiamide (Raichur and Vijayapura strains), respectively, and Spinosad (Kalaburgi and Raichur strains), respectively. The morphometric parameters of larval length, pupal length, and weight were most in RCH stain (2.75 cm, 1.76.18 cm, and 0.511 g, 0.309 g) respectively, which was pursued by Kalaburgi strain. The morphometric correlation revealed that larval length was a significant positive relation with insecticide resistance which might be an influence of resistance but not merely responsible. Among newer insecticides, a significant positive correlation between Rynaxypyr and Indoxacarb was evident, similarly, Nuvaluran with Indoxacarb and Rynaxypyr as well. Usage pattern revealed that 81.67 % of farmers found to use insecticides more than the recommended dose and 70.83% have habit consecutive applications of products from the same chemical group which bears witness to developing resistance.

Current status of insecticide resistance in malaria vectors in the Asian countries: a systematic review

Background: The application of insecticides for malaria vector control has led to a global problem, which is the current trend of increased resistance against these chemicals. This study aimed to review the insecticide resistance status was previously determined in Asia and how to implement the necessary interventions. Moreover, the implications of resistance in malaria vector control in this region were studied. Methods: This systematic review was conducted using a predefined protocol based on PRISMA-retrieved articles from four science databases, namely ProQuest, Science Direct, EBSCO, and PubMed in the last ten years (2009 to 2019). The searching process utilized four main combinations of the following keywords: malaria, vector control, insecticide, and Asia. In ProQuest, malaria control, as well as an insecticide, were used as keywords. The following criteria were included in the filter, namely full text, the source of each article, scholarly journal, Asia, and publication date as in the last ten years. Results: There were 1408 articles retrieved during the initial search (ProQuest=722, Science Direct=267, EBSCO=50, PubMed=285, and Scopus=84). During the screening, 27 articles were excluded because of duplication, 1361 based on title and abstract incompatibility with the inclusion criteria, and 20 due to content differences. In the final screening process, 15 articles were chosen to be analyzed. From the 15 articles, it is known that there was organochlorine (DDT), organophosphate (malathion), and pyrethroids resistance in several Anopheles species with a less than 80% mortality rate. Conclusion: This review found multiple resistance in several Anopheles includes resistance to pyrethroid. The reports of pyrethroid resistance were quite challenging because it is considered effective in the malaria vector control. Several countries in Asia are implementing an insecticide resistance management (IRM) strategy against malaria vectors following the Global Plan for IRM.
