Maribacter aestuarii sp. nov., isolated from tidal flat sediment, and an emended description of the genus Maribacter

A Gram-staining-negative, strictly aerobic bacterial strain, motile by gliding, designated GY20T, was isolated from a tidal flat at Gwangyang Bay, South Korea. Cells were moderately halophilic, catalase- and oxidase-positive rods. Growth of strain GY20T was observed at 10–30 °C (optimum, 25 °C), at pH 6.5–11.0 (optimum, pH 7.0–8.0) and in the presence of 1–5 % (w/v) NaCl (optimum, 2–3 %). MK-6 was detected as the sole isoprenoid quinone, and iso-C15 : 0, iso-C15 : 1 G, summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c), summed feature 9 (iso-C17 : 1ω9c and/or 10-methyl C16 : 0), iso-C17 : 0 3-OH and iso-C15 : 0 3-OH as major fatty acids. Strain GY20T contained phosphatidylethanolamine, one unidentified glycolipid and one unidentified lipid as major polar lipids. The G+C content of the genomic DNA was 39.8 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain GY20T formed a tight phyletic lineage with members of the genus Maribacter . Strain GY20T was most closely related to Maribacter dokdonensis DSW-8T, with sequence similarity of 96.2 %. On the basis of phenotypic, chemotaxonomic and molecular properties, strain GY20T was shown to represent a novel species within the genus Maribacter , for which the name Maribacter aestuarii sp. nov. is proposed. The type strain is GY20T ( = KACC 16440T = JCM 18631T). An emended description of the genus Maribacter is also proposed.

A Gram-staining-negative, yellow-pigmented, strictly aerobic bacterial strain motile by gliding, designated BS12T, was isolated from a tidal flat at Boseong, Korea. Cells were moderately halotolerant, catalase- and oxidase-positive rods. Growth was observed at 5–40 °C (optimum, 25 °C), at pH 5.5–9.0 (optimum, pH 7.0–7.5) and in the presence of 1–11 % (w/v) NaCl (optimum, 2–4 %). The major cellular fatty acids were iso-C15 : 0, iso-C17 : 0 3-OH, iso-C17 : 1ω9c and anteiso-C15 : 0. The polar lipid pattern comprised phosphatidylethanolamine, four unidentified aminolipids and three unidentified lipids. The G+C content of the genomic DNA was 42.3 mol% and the only respiratory quinone was menaquinone 6. Phylogenetic inference based on 16S rRNA gene sequences showed that strain BS12T formed a tight phyletic lineage with members of the genus Gramella . Strain BS12T was most closely related to ‘ Gramella jeungdoensis’ HMD3159 with sequence similarity of 97.5 %, but the DNA–DNA relatedness value between the two strains was only 22.1±2.2 %. On the basis of phenotypic and molecular features, strain BS12T was shown to represent a novel species of the genus Gramella , for which the name Gramella aestuarii sp. nov. is proposed. The type strain is BS12T ( = KACC 16188T = JCM 17790T). An emended description of Gramella echinicola is also proposed.

A Gram-staining-negative, aerobic and rod-shaped bacterial strain, designated HMC4223T, was isolated from a tidal flat in Taean, Republic of Korea. A phylogenetic tree based on 16S rRNA gene sequences showed that strain HMC4223T formed a lineage within the genus Algoriphagus and exhibited highest sequence similarity to Algoriphagus hitonicola 7-UAHT (96.3 %). The major fatty acids of strain HMC4223T were iso-C15 : 0 and summed feature 3 (comprising C16 : 1ω6c and/or C16 : 1ω7c). The major respiratory quinone was MK-7. The major polar lipids were phosphatidylethanolamine and five unidentified polar lipids. The DNA G+C content of strain HMC4223T was 48.4 mol%. On the basis of the evidence presented in this study, strain HMC4223T represents a novel species of the genus Algoriphagus , for which the name Algoriphagus taeanensis sp. nov. is proposed, with the type strain HMC4223T ( = KCTC 22709T = NBRC 105728T). An emended description of Algoriphagus hitonicola is also presented.

A Gram-staining-negative, strictly aerobic, white-colony-forming bacterium, designated strain 5-10T, was isolated from forest soil of Bac Kan Province in Vietnam. Cells were non-motile rods or coccoids, showing oxidase- and catalase-positive reactions. Growth was observed at 10–37 °C (optimum, 30 °C), at pH 5.0–9.0 (optimum, pH 7.0) and in the presence of 0–1.0 % (w/v) NaCl (optimum, 0–0.5 %). The major cellular fatty acids were summed feature 3 (comprising C16 : 1ω6c and/or C16 : 1ω7c), C16 : 0, C10 : 0 3-OH and summed feature 8 (comprising C18 : 1ω6c and/or C18 : 1ω7c). The G+C content of the genomic DNA was 69.9 mol% and the only respiratory quinone detected was ubiquinone 8 (Q-8). Phylogenetic analysis based on 16S rRNA gene sequences showed that strain 5-10T formed a tight phyletic lineage with members of the genus Ramlibacter . Strain 5-10T was most closely related to Ramlibacter tataouinensis TTB310T (97.3 %), but the DNA–DNA relatedness level between the two strains was 38.2±1.8 %. Based on phenotypic, chemotaxonomic and molecular features, strain 5-10T was shown to represent a novel species of the genus Ramlibacter , for which the name Ramlibacter solisilvae sp. nov. is proposed. The type strain is 5-10T ( = KACC 17567T = JCM 19319T). An emended description of the genus Ramlibacter is also proposed.

An orange, rod-shaped, gliding bacterium, designated strain PSC33T, was isolated from the gut microflora of a mussel collected from Gwangyang Bay, South Sea (Republic of Korea). Cells were Gram-reaction-negative, strictly aerobic, and catalase- and oxidase-positive. The major fatty acids were iso-C15 : 0, iso-C17 : 0 3-OH, iso-C15 : 1 G, C15 : 0 3-OH and iso-C17 : 1ω9c. The only isoprenoid quinone was menaquinone-6 (MK-6). The DNA G+C content of strain PSC33T was 37.9 mol%. A phylogenetic tree based on 16S rRNA gene sequences showed that strain PSC33T formed an evolutionary lineage within the radiation encompassing members of the genus Aquimarina with Aquimarina macrocephali JAMB N27T as its nearest neighbour (96.3 % sequence similarity). A number of phenotypic characteristics distinguished strain PSC33T from recognized members of the genus Aquimarina . On the basis of the data presented in this study, strain PSC33T is considered to represent a novel species of the genus Aquimarina , for which the name Aquimarina mytili sp. nov. is proposed. The type strain is PSC33T ( = KCTC 23302T = JCM 17454T). An emended description of A. macrocephali is also provided.

A novel Gram-staining-positive, rod-shaped, motile, strictly aerobic, endospore-forming bacterium, designated WCC 4585T, was isolated from a pharmaceutical production line. The organism grew optimally at 30 °C, at pH 8 and in the presence of 0.5 % (w/v) NaCl. Oval endospores were formed subterminally and terminally in swollen sporangia. The cell-wall diamino acid was meso-diaminopimelic acid (type A1γ) and the genomic DNA G+C content was 38.7 mol%. The major menaquinone was MK-7. The cellular fatty acid profile contained major amounts of iso-C15 : 0, anteiso-C15 : 0 and anteiso-C17 : 0, and the cellular phospholipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and aminophospholipid. The isolate was most closely related to Bacillus oceanisediminis H2T, Bacillus infantis SMC 4352-1T, Bacillus firmus NCIMB 9366T, Bacillus circulans ATCC 4513T and Bacillus horneckiae DSM 23495T with which it shared less than 98.0 % 16S rRNA gene sequence similarity. DNA–DNA relatedness values between strain WCC 4585T and five type strains of related species were ≤27 % and sequence similarity values based on groEL sequences were ≤88.7 %. On the basis of the characteristics presented, strain WCC 4585T is proposed to represent a novel species, Bacillus gottheilii sp. nov. The type strain is WCC 4585T( = DSM 23668T = CCUG 59876T = LMG 25856T).

A novel yellow-pigmented bacterial strain, designated ARSA-19T, was isolated from a freshwater reservoir in Cheonan, Korea. Cells were Gram-staining-negative, rod-shaped, strictly aerobic and positive for catalase and oxidase. The phylogenetic tree based on 16S rRNA gene sequences showed that strain ARSA-19T belonged to the genus Flavobacterium and was related to Flavobacterium macrobrachii an-8T (96.5 % sequence similarity), F. koreense KACC 14969T (96.2 %), F. chungnamense KACC 14971T (96.1 %), F. cheonanense KACC 14972T (96.4 %) and F. aquatile ATCC 11947T (95.6 %). The isolate contained MK-6 as the predominant menaquinone and iso-C15 : 0 (21.8 %), iso-C16 : 0 3-OH (10.4 %), iso-C15 : 1 G (9.3 %), summed feature 3 (iso-C15 : 0 2-OH and/or C16 : 1ω7c; 8.2 %), iso-C15 : 0 3-OH (7.7 %) and iso-C16 : 0 (7.3 %) as the major fatty acids. The polar lipids of strain ARSA-19T were phosphatidylethanolamine, four unknown aminolipids and two unidentified polar lipids. The DNA G+C content of strain ARSA-19T was 33.3 mol%. Based on its phenotypic and genotypic characteristics and on the phylogenetic evidence presented, strain ARSA-19T is considered to represent a novel species in the genus Flavobacterium , for which the name Flavobacterium dankookense sp. nov. is proposed. The type strain is ARSA-19T ( = KCTC 23179T  = JCM 17065T  = KACC 14968T). Emended descriptions of Flavobacterium cheonanense , Flavobacterium chungnamense , Flavobacterium koreense and Flavobacterium aquatile are also proposed.

Colonies were opaque, white, smooth and circular on marine agar. Cells were moderately halophilic, non-motile rods showing catalase- and oxidase-positive reactions. Growth of strain BS14T was observed at 5–40 °C (optimum: 30 °C), pH 6.5–9.5 (optimum: 7.0–7.5) and 0–10 % (w/v) NaCl (optimum: 1–1.5 %). The G+C content of the genomic DNA was 61.6 mol%. Strain BS14T contained ubiquinone-10 (Q-10) as the sole respiratory quinone and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c), C18 : 0 3-OH, C10 : 0 3-OH and C18 : 0 as the major fatty acids. The polar lipid pattern comprised phosphatidylethanolamine, diphosphatidylglycerol, an unidentified aminolipid, an unidentified phospholipid and an unidentified polar lipid. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain BS14T formed a tight phylogenetic lineage with Defluviimonas denitrificans D9-3T with a bootstrap value of 100 %. The 16S rRNA gene sequence similarity between strain BS14T and D. denitrificans D9-3T was 97.4 % and their DNA–DNA relatedness was 19.1±3.6 %. Based on the phenotypic and genotypic studies, strain BS14T represents a novel species of the genus Defluviimonas , for which the name Defluviimonas aestuarii sp. nov. is proposed. The type strain is BS14T ( = KACC 16442T = JCM 18630T). An emended description of the genus Defluviimonas Foesel et al. 2011 is also proposed.

Growth of strain G3T was observed between 15 and 40 °C (optimum 30 °C) and between pH 5.5 and 9.0 (optimum pH 6.5–7.5). Strain G3T contained Q-8 as the predominant lipoquinone and iso-C15 : 0, iso-C17 : 1ω9c, iso-C16 : 0 and iso-C11 : 0 as the major fatty acids. The G+C content of the genomic DNA was 69.6 mol%. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain G3T formed a tight phylogenetic lineage with Luteimonas mephitis B1953/27.1T within the genus Luteimonas and was most closely related to L. mephitis B1953/27.1T with 98.0 % 16S rRNA gene sequence similarity. The DNA–DNA relatedness between strain G3T and L. mephitis B1953/27.1T was 35.2±3.3 %. On the basis of chemotaxonomic data and molecular properties, strain G3T represents a novel species of the genus Luteimonas, for which the name Luteimonas lutimaris sp. nov. is proposed. The type strain is G3T ( = KACC 14929T = JCM 16916T).

A novel facultatively anaerobic, Gram-stain-negative bacterium, designated strain HT7-2T, was isolated from Ulva prolifera collected from the intertidal zone of Qingdao sea area, China, during its bloom. Cells were rod-shaped (1.9–3.5×0.4–0.6 µm), non-sporulating and motile by gliding. Strain HT7-2T was able to grow at 4–50 °C (optimum 40–42 °C), pH 5.5–8.5 (optimum pH 7.0), 0–8 % (w/v) NaCl (optimum 2–3 %) and 0.5–10 % (w/v) sea salts (optimum 2.5 %). The genomic DNA G+C content was 38.8 mol%. The phylogenetic analysis based on 16S rRNA gene sequences revealed that strain HT7-2T belonged to the genus Maribacter with sequence similarity values of 94.5–96.6 %, and was most closely related to Maribacter aestuarii GY20T (96.6 %). Chemotaxonomic analysis showed that the main isoprenoid quinone was MK-6 and the major fatty acids were iso-C15 : 0 and unknown equivalent chain-length 13.565. The polar lipids of strain HT7-2T consisted of one phosphatidylethanolamine, four unidentified lipids and one unidentified aminolipid. On the basis of the phenotypic, phylogenetic and chemotaxonomic characteristics, strain HT7-2T ( = CGMCC 1.12207T = JCM 18466T) is concluded to represent a novel species of the genus Maribacter , for which the name Maribacter thermophilus sp. nov. is proposed. An emended description of the genus Maribacter is also proposed.

Phosphatidylethanolamine, one unidentified aminolipid and two unidentified polar lipids were found as major polar lipids. A phylogenetic tree based on 16S rRNA gene sequences showed that strain PSC32T forms an evolutionary lineage within the genus Aquimarina and is closely related to Aquimarina spongiae A6T (97.0 % similarity) and to other members of the genus Aquimarina (94.4–96.5 %). Genomic DNA–DNA relatedness between strain PSC32T and A. spongiae A6T was 40.7 %. A number of phenotypic characteristics distinguished strain PSC32T from described members of the genus Aquimarina . On the basis of the evidence presented in this study, strain PSC32T represents a novel species, for which the name Aquimarina gracilis sp. nov. is proposed. The type strain is PSC32T ( = KCTC 23301T = JCM 17453T). An emended description of Aquimarina spongiae is given.
