Terriglobus aquaticus sp. nov., isolated from an artificial reservoir

A pink-pigmented, chemo-organotrophic bacterium, designated strain 03SUJ4T, was isolated from the freshwater of Juam reservoir, Republic of Korea (35° 03′ 43′′ N 127° 14′ 15′′ E). Cells were aerobic, Gram-reaction-negative and non-motile rods. Strain 03SUJ4T grew at pH 6–7 (optimum, pH 6) and at 15–30 °C (optimum, 25 °C). Phylogenetic analysis based on 16S rRNA gene sequences indicated that the isolate belonged to the genus Terriglobus , showing sequence similarities of 97.09 % and 96.82 % to Terriglobus roseus DSM 18391T and Terriglobus saanensis SP1PR4T, respectively. Low rpoB gene sequence similarity with members of the genus Terriglobus and different fingerprints with the repetitive primers BOX, ERIC and REP indicated that the isolate represented a novel species of the genus Terriglobus . The major cellular fatty acids were iso-C15 : 0, C16 : 0, C20 : 1ω9c, C14 : 0 and summed feature 3 (C16 : 1ω7c/C16 : 1ω6c). The DNA G+C content of strain 03SUJ4T was 63.2±0.1 mol% (mean±sd of three determinations). The predominant menaquinone was MK-8. The major polar lipids were phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylglycerol and two unidentified phospholipids. Several phenotypic characteristics served to differentiate the novel isolate from recognized members of the genus Terriglobus . On the basis of the evidence presented in this study, a novel species, Terriglobus aquaticus sp. nov. is proposed for strain 03SUJ4T ( = KCTC 23332T = JCM 17517T).

Enrichment cultures inoculated with hydrothermally influenced nearshore sediment from Papua New Guinea led to the isolation of an arsenic-tolerant, acidophilic, facultatively aerobic bacterial strain designated PNG-AprilT. Cells of this strain were Gram-stain-negative, rod-shaped, motile and did not form spores. Strain PNG-AprilT grew at temperatures between 4 °C and 40 °C (optimum 30–37 °C), at pH 3.5 to 8.3 (optimum pH 5–6) and in the presence of up to 2.7 % NaCl (optimum 0–1.0 %). Both arsenate and arsenite were tolerated up to concentrations of at least 0.5 mM. Metabolism in strain PNG-AprilT was strictly respiratory. Heterotrophic growth occurred with O2 or nitrate as electron acceptors, and aerobic lithoautotrophic growth was observed with thiosulfate or nitrite as electron donors. The novel isolate was capable of N2-fixation. The respiratory quinones were Q-8 and Q-7. Phylogenetically, strain PNG-AprilT belongs to the genus Burkholderia and shares the highest 16S rRNA gene sequence similarity with the type strains of Burkholderia fungorum (99.8 %), Burkholderia phytofirmans (98.8 %), Burkholderia caledonica (98.4 %) and Burkholderia sediminicola (98.4 %). Differences from these related species in several physiological characteristics (lipid composition, carbohydrate utilization, enzyme profiles) and DNA–DNA hybridization suggested the isolate represents a novel species of the genus Burkholderia , for which we propose the name Burkholderia insulsa sp. nov. The type strain is PNG-AprilT ( = DSM 28142T = LMG 28183T).

A non-motile and rod-shaped bacterium, designated strain 02SUJ3T, was isolated from freshwater collected from the Juam Reservoir (Republic of Korea). Cells were Gram-stain-negative, aerobic, oxidase-negative and catalase-positive. The major fatty acids were iso-C15 : 0, iso-C15 : 1 G and iso-C17 : 0 3-OH. The strain contained MK-7 as the major isoprenoid quinone. The main polar lipid was phosphatidylethanolamine. The DNA G+C content was 46.4 mol%. A phylogenetic tree based on 16S rRNA gene sequences showed that strain 02SUJ3T forms an independent lineage within the genus Flavisolibacter with low sequence similarity to Flavisolibacter ginsengiterrae Gsoil 492T and Flavisolibacter ginsengisoli Gsoil 643T (95.7 %). Phenotypic characteristics distinguished strain 02SUJ3T from members of the genus Flavisolibacter . On the basis of the evidence presented in this study, strain 02SUJ3T is considered to represent a novel species, for which the name Flavisolibacter rigui sp. nov. is proposed. The type strain is 02SUJ3T ( = JCM 17515T = KCTC 23328T). An emended description of the genus Flavisolibacter is also provided.

A polyphasic analysis was undertaken of seven independent isolates of Gram-negative cocci collected from pathological clinical samples from New York, Louisiana, Florida and Illinois and healthy subgingival plaque from a patient in Virginia, USA. The 16S rRNA gene sequence similarity among these isolates was 99.7–100 %, and the closest species with a validly published name was Neisseria lactamica (96.9 % similarity to the type strain). DNA–DNA hybridization confirmed that these isolates are of the same species and are distinct from their nearest phylogenetic neighbour, N. lactamica . Phylogenetic analysis of 16S and 23S rRNA gene sequences indicated that the novel species belongs in the genus Neisseria . The predominant cellular fatty acids were C16 : 0, summed feature 3 (C16 : 1ω7c and/or iso-C15 : 0 2-OH) and C18 : 1ω7c. The cellular fatty acid profile, together with other phenotypic characters, further supports the inclusion of the novel species in the genus Neisseria . The name Neisseria oralis sp. nov. (type strain 6332T  = DSM 25276T  = LMG 26725T) is proposed.

A novel endophytic actinobacterium, designated strain YIM 68236T, was isolated from healthy leaves of Camptotheca acuminata. and characterized by using a polyphasic approach. Cells of this strain occurred singly, in pairs or in tetrads. It grew at 10–45 °C, at pH 5.0–8.0 (optimum pH 7.0) and in the presence of 0–3 % (w/v) NaCl. The DNA G+C content was 71.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain YIM 68236T belongs to the genus Blastococcus . However, it differed from its closest relatives, Blastococcus aggregatus DSM 4725T, Blastococcus saxobsidens DSM 44509T and Blastococcus jejuensis DSM 19597T in many phenotypic characteristics. Moreover, the DNA–DNA relatedness values between the novel isolate and the three above-mentioned type strains were 49.0±1.6 %, 46.1±3.2 % and 39.8±1.5 %, respectively. Based on comparative analysis of physiological and chemotaxonomic data, strain YIM 68236T represents a novel species of the genus Blastococcus , for which the name Blastococcus endophyticus sp. nov. is proposed. The type strain is YIM 68236T ( = CCTCC AA 209045T = DSM 45413T = KCTC 19998T).

A Gram-stain-negative, aerobic, non-motile, rod- and coccus-shaped bacterium, designated strain B6-12T, was isolated from sediment collected from the River Geumho in South Korea. In comparative 16S rRNA gene sequence analysis, the novel strain appeared to be affiliated with the class Alphaproteobacteria and to be most closely related to Kaistia adipata KCTC 12095T, Kaistia dalseonensis DSM 18800T, Kaistia geumhonensis DSM 18799T, Kaistia granuli KCTC 12575T, Kaistia soli KACC 12605T and Kaistia terrae KACC 12910T, with sequence similarities of 96.2–99.1 %. The predominant ubiquinone in the isolate was Q-10, major fatty acids were C18 : 0, C18 : 1ω7c and C19 : 0ω8c cyclo, and genomic DNA G+C content was 63.0 mol%. Based on the phylogenetic and chemotaxonomic evidence and the results of DNA–DNA hybridizations, strain B6-12T represents a novel species in the genus Kaistia , for which the name Kaistia defluvii sp. nov. is proposed. The type strain is B6-12T ( = KCTC 23766T  = JCM 18034T).

A novel endophytic actinomycete, designated strain NEAU-TX2-2T, was isolated from moss and characterized using a polyphasic approach. The isolate was found to have morphological characteristics typical of the genus Microbispora . The isolate formed longitudinally paired spores on the tips of short sporophores that branched from aerial hyphae. Analysis of the 16S rRNA gene sequence supported the assignment of the novel strain to the genus Microbispora , and strain NEAU-TX2-2T exhibited 99.08 and 98.62 % gene sequence similarities to Microbispora amethystogenes JCM 3021T and Microbispora rosea subsp. rosea JCM 3006T, respectively. However two tree-making algorithms supported the position that strain NEAU-TX2-2T formed a distinct clade with M. rosea subsp. rosea JCM 3006T. A low level of DNA–DNA relatedness allowed the isolate to be differentiated from M. amethystogenes JCM 3021T and M. rosea subsp. rosea JCM 3006T. Moreover, strain NEAU-TX2-2T could also be distinguished from its closest phylogenetic relatives by morphological and physiological characteristics. Therefore, it is proposed that strain NEAU-TX2-2T represents a novel species of the genus Microbispora for which the name Microbispora bryophytorum sp. nov. is proposed. The type strain is NEAU-TX2-2T ( = CGMCC 4.7138T = DSM 46710T).

Strain SulfLac1T, a thermophilic, anaerobic and slightly halophilic, rod-shaped bacterium with a sheath-like outer structure (toga), was isolated from a whey digester in Tunisia. The strain’s non-motile cells measured 3–30×1 µm and appeared singly, in pairs or as long chains. The novel strain reduced thiosulfate and elemental sulfur, but not sulfate or sulfite, into sulfide. It grew at 37–65 °C (optimum 55 °C), at pH 6.5–7.9 (optimum pH 6.9) and with 0.2–3 % (w/v) NaCl (optimum 0.5 %). The G+C content of the strain’s genomic DNA was 33.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain SulfLac1T was most closely related to Petrotoga mobilis (91.4 % sequence similarity). Based on phenotypic, phylogenetic and chemotaxonomic evidence, strain SulfLac1T represents a novel species of a new genus within the order Thermotogales , for which the name Defluviitoga tunisiensis gen. nov., sp. nov. is proposed. The type strain of the type species is SulfLac1T ( = DSM 23805T  = JCM 17210T).

A novel actinomycete, designated strain NEAU-GRX6T, was isolated from mucky soil collected from a stream of Jinlong Mountain in Harbin, Heilongjiang Province, north China, and characterized using a polyphasic approach. The isolate formed irregular sporangia containing motile sporangiospores on the substrate mycelium. The whole-cell sugars were xylose, glucose and galactose. The predominant menaquinones were MK-9(H6), MK-10(H4) and MK-9(H4). The major fatty acids were C16 : 0, C15 : 0, C18 : 1ω9c, C17 : 1ω7c and C18 : 0. The phospholipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol and phosphatidylinositol. The DNA G+C content was 67 mol%. 16S rRNA gene sequence similarity studies showed that strain NEAU-GRX6T belonged to the genus Actinoplanes , being most closely related to Actinoplanes palleronii IFO 14916T (97.80 % similarity) and Actinoplanes missouriensis NBRC 102363T (97.76 %). However, the low observed levels of DNA–DNA relatedness allowed the isolate to be differentiated from the above-mentioned species of the genus Actinoplanes . Moreover, strain NEAU-GRX6T could also be distinguished from A. palleronii IFO 14916T and A. missouriensis NBRC 102363T by phenotypic characteristics. Therefore, it is proposed that strain NEAU-GRX6T represents a novel species of the genus Actinoplanes , for which the name Actinoplanes lutulentus sp. nov. is proposed. The type strain is strain NEAU-GRX6T ( = CGMCC 4.7090T = DSM 45883T).

A Gram-stain-negative, aerobic, orange-pigmented, rod-shaped and non-motile bacterium, designated strain A6B8T, was isolated from seawater of the Mariana Trench. The isolate grew at 4–50 °C (optimum 30–35 °C), at pH 6.5–8.0 (optimum pH 7.5) and with 0.5–4.0 % (w/v) NaCl (optimum 1.0–2.0 %). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain A6B8T was related most closely to the genus Muriicola and shared highest sequence similarity of 97.7 % with Muriicola jejuensis EM44T. Chemotaxonomic analysis showed menaquinone 6 (MK-6) was the predominant isoprenoid and iso-C15 : 0, iso-C15 : 1 G and iso-C17 : 0 3-OH were the major cellular fatty acids. The polar lipid profile of strain A6B8T included phosphatidylethanolamine, three unidentified aminolipids and four unidentified lipids. The genomic DNA G+C content was 47.1 mol%. The DNA–DNA relatedness value (23.3 %) clearly demonstrated that strains A6B8T and M. jejuensis EM44T were representatives of two different species. Based on the phenotypic, phylogenetic and chemotaxonomic characterizations, A6B8T ( = CGMCC 1.12606T = KCTC 32436T) is considered to be the type strain of a novel species of the genus Muriicola , for which the name Muriicola marianensis sp. nov. is proposed.

A novel psychrotolerant, strictly aerobic, non-motile, rod-shaped bacterial strain, designated IM13T, was isolated from a sample taken from prehistoric guano paintings in Magura Cave, northwest Bulgaria and subjected to a polyphasic taxonomic study. Strain IM13T formed yellow colonies on LB agar plates and was Gram-staining-negative, heterotrophic and alkalitolerant. It grew optimally at pH 7.5 and 30 °C in the absence of NaCl. Phylogenetic analysis of the whole 16S rRNA gene revealed that strain IM13T branched with representatives of the genus Myroides with sequence similarity of 93–94 % with other species of the genus. The novel isolate contained iso-C15 : 0 (49.1 %), iso-C17 : 1ω9c (18.2 %) and iso-C17 : 0 3-OH (14.0 %) as dominant fatty acids. The DNA G+C content of strain IM13T was 33.5 mol%. Based on phylogenetic inference and phenotypic characteristics, it was concluded that strain IM13T represents a novel species of the genus Myroides , for which the name Myroides guanonis sp. nov. is proposed. The type strain is IM13T ( = DSM 26542T = NBIMCC 8736T).
