Expectations have an important role in guiding behavior and the interpretation of novel information, but can contain negative biases such as is the case in anxiety disorders. Positive future thinking may provide an accessible way to attenuate these negatively biases. However, much is still unclear about the optimal form of such positive interventions, and it is unknown if the effects go beyond subjective experience. Here, we used a positive future thinking intervention to adapt the way a stressful event is experienced. Participants imagined either task-relevant (N = 21) or irrelevant (N = 21) positive future events before being subjected to the Trier Social Stress Task, or did not receive the intervention (N= 20). We recorded resting state EEG during the anticipation and recovery phases of the TSST to assess intervention and trait anxiety related differences in the level of frontal delta-beta coupling, which is considered a neurobiological substrate of emotion regulation. Results show that the intervention reduces event-related stress and anxiety, and increases social fixation behavior and task performance, but only if future thinking is task relevant. Paradoxically, task-irrelevant positive future thoughts enhance negative perceptual biases and stress reactivity. This increase in stress reactivity in the task-irrelevant group was corroborated by the elevated levels frontal delta-beta coupling during event anticipation, especially for those with high trait anxiety. This suggests an increased demand for emotion regulation following the task-irrelevant intervention, possibly due to the contextual incongruence between positive imagery and the stressor. Together, these results show that positive future thinking can mitigate the negative emotional, behavioral and neurobiological consequences of a stressful event, but that it should not be applied indiscriminately. Task-relevant positive future thinking can be an accessible way to boost efficacy of exposure therapy for pathological anxiety, and can help people deal with negative anticipation in daily life.

AbstractAnxiety and stress-related disorders are highly prevalent and debilitating conditions that impose an enormous burden on society. Sensitive measurements that can enable early diagnosis could mitigate suffering and potentially prevent onset of these conditions. Self-reports, however, are intrusive and vulnerable to biases that can conceal the true internal state. Physiological responses, on the other hand, manifest spontaneously and can be monitored continuously, providing potential objective biomarkers for anxiety and stress. Recent studies have shown that algorithms trained on physiological measurements can predict stress states with high accuracy. Whether these predictive algorithms generalize to untested situations and participants, however, remains unclear. Further, whether biomarkers of momentary stress indicate trait anxiety – a vulnerability foreshadowing development of anxiety and mood disorders – remains unknown. To address these gaps, we monitored skin conductance, heart rate, heart rate variability and EEG in 39 participants experiencing physical and social stress and compared these measures to non-stressful periods of talking, rest, and playing a simple video game. Self-report measures were obtained periodically throughout the experiment. A support vector machine trained on physiological measurements identified stress conditions with ~96% accuracy. A decision tree that optimally combined physiological and self-report measures identified individuals with high trait anxiety with ~84% accuracy. Individuals with high trait anxiety also displayed high baseline state anxiety but a muted physiological response to acute stressors. Overall, these results demonstrate the potential for using machine learning tools to identify objective biomarkers useful for diagnosing and monitoring mental health conditions like anxiety and depression.

Objective: Mindfulness interventions, which train practitioners to monitor their present-moment experience with a lens of acceptance, are known to buffer stress reactivity. Little is known about the active mechanisms driving these effects. We theorize that acceptance is a critical emotion regulation mechanism underlying mindfulness stress reduction effects. Method: In this three-arm parallel trial, mindfulness components were dismantled into three structurally equivalent 15-lesson smartphone-based interventions: (1) training in both monitoring and acceptance (Monitor+Accept), (2) training in monitoring only (Monitor Only), or (3) active control training (Coping control). 153 stressed adults (mean age = 32 years; 67% female; 53% white, 21.5% black, 21.5% Asian, 4% other race) were randomly assigned to complete one of three interventions. After the intervention, cortisol, blood pressure, and subjective stress reactivity were assessed using a modified Trier Social Stress Test. Results: As predicted, Monitor+Accept training reduced cortisol and systolic blood pressure reactivity compared to Monitor Only and control trainings. Participants in all three conditions reported moderate levels of subjective stress.Conclusions: This study provides the first experimental evidence that brief smartphone mindfulness training can impact stress biology, and that acceptance training drives these effects. We discuss implications for basic and applied research in contemplative science, emotion regulation, stress and coping, health, and clinical interventions.

Abstract Objectives Several studies have investigated the relationship between contemplative approaches and psychobiological stress response; however, this area of research is still new, the mechanisms of the relationship between the stress response attenuation and contemplative training have not been fully delineated, and little is known about the effects of contemplative practice on the ways psychological stress is experienced. This study aimed to explore the first-person experience of psychological stress in meditation practitioners. Methods We conducted short semi-structured interviews with twenty-five meditation practitioners and twenty meditation-naïve controls immediately after they had undergone a laboratory task (the Trier Social Stress Test). A mixed-method approach was used to analyze the interviews. Thematic analysis was combined with descriptive statistics of the qualitative information that had been converted to quantitative data. Results Experiences instantiating main themes were identified as follows: (1) primary experiences encountered, describing the most salient experiences associated with the task; (2) reasons for stress, delineating the analyses of why the task was stressful; (3) affect, dealing with emotional experiences during the task; (4) emotion regulation; and (5) attention allocation describing regulatory strategies employed by the participants. Responses to subjective stress experience in meditation practitioners included use of humor, presence of positive affect, combinations of different types of emotion regulation strategies, and adaptive attention allocation. Conclusions This study elucidates particularities of meditators’ subjective experience of psychological stress, provides new insights on the mechanisms of meditation effect on the stress response, and proposes new directions for research.

In recent years, it has become increasingly clear that common forms of psychopathology derive from complex interactions among neurobiological vulnerabilities and environmental adversities. These interactions can alter neurobehavioral development to yield progressively intractable forms of psychopathology across childhood and adolescence. This chapter focuses on neurobiological mechanisms of trait impulsivity, trait anxiety, stress reactivity, and emotion regulation/executive function. How these traits confer vulnerability to externalizing disorders, internalizing disorders, heterotypic comorbidity, and heterotypic continuity is described. Next, neurobiological mechanisms of treatment response are considered. Trait impulsivity and trait anxiety are highly heritable and derive initially from subcortical structures that mature early in life. In contrast, emotion regulation and executive function, which modulate trait impulsivity and trait anxiety, are more sensitive to environmental influence and derive from cortical structures that mature into young adulthood. Neurobiological mechanisms of psychosocial treatment response are represented largely in the cortex and its neuromodulatory connections with the subcortex.
