Kemampuan Indonesia dalam memenuhi kebutuhan beras nasional sangat penting. Akan tetapi, usaha pemenuhan kebutuhan beras ini tidak selamanya berjalan dengan lancar karena terganggunya produktivitas padi. Salah satu penyebab turunnya produktivitas padi di Indonesia karena adanya serangan serangga hama. Beberapa kendala yang menyebabkan gagalnya petani dalam mengendalikan serangga hama karena petani masih belum melakukan identifikasi serangga hama dan gejala serangan dengan baik. Penelitian yang dilaksanakan akhir tahun 2012 ini yang berlokasi di Lahan Persawahan Padi Dataran Tinggi Desa Sukawening, Kecamatan Ciwidey, Kabupaten Bandung, Jawa Barat bertujuan meng inventarisasi dan mendapatkan nilai keragaman jenis serangga hama pada tanaman padi. Penangkapan serangga hama yang dilakukan dengan menggunakan perangkap kuning, perangkap jaring dan perangkap cahaya. Hasil penangkapan serangga diidentifikasi di laboratorium. Indeks keragaman serangga dianalisis dengan menggunakan perhitungan Shannon- Weinner. Serangga hama yang dominan diperoleh pada fase vegetative yaitu Scirpophaga incertulas, Thaia oryzivora, dan Orselia oryzae. Serangga hama minor diperoleh yaitu Dicladispa armigera, Leptispa pygmaea, dan Melanitis ledaismene. Serangga hama yang dominan diperoleh pada fase generative yaitu Leptocorisa acuta, Scirpophaga incertulas, dan Thaiaoryzivora. Serangga hama minor diperoleh yaitu Nilaparvata lugens, Sogatella furcifera, dan Cofana spectra. Indeks keragaman serangga hama yang diperoleh pada ketiga lahan percobaan ada diantara sedang hingga tinggi.Pada fase vegetatif indeks keragaman tertinggi sebesar 4,74 pada pengamatan ke 6 dan terendah pada pengamatan ke 1 sebesar 2,22. Pada fase generatif indeks keragaman tertinggi pada pengamatan ke 9 sebesar 4,86 dan terendah pada pengamatan ke 12 sebesar 1,37.   Kata kunci :indeks keragaman, serangga hama, padi, Kabupaten Bandung.

ABSTRACT The brown planthopper Nilaparvata lugens (Hemiptera) is a major pest of rice crops in Asia. Artificial transinfections of Wolbachia have recently been used for reducing host impacts, but transinfections have not yet been undertaken with another important endosymbiont, Cardinium. This endosymbiont can manipulate the reproduction of hosts through phenotypes such as cytoplasmic incompatibility (CI), which is strong in the related white-backed planthopper, Sogatella furcifera (Hemiptera). Here, we stably infected N. lugens with Cardinium from S. furcifera and showed that it exhibits perfect maternal transmission in N. lugens. The density of Cardinium varied across developmental stages and tissues of the transinfected host. Cardinium did not induce strong CI in N. lugens, likely due to its low density in testicles. The infection did decrease fecundity and hatching rate in the transinfected host, but a decrease in fecundity was not apparent when transinfected females mated with Wolbachia-infected males. The experiments show the feasibility of transferring Cardinium endosymbionts across hosts, but the deleterious effects of Cardinium on N. lugens limit its potential to spread in wild populations of N. lugens in the absence of strong CI. IMPORTANCE In this study we established a Cardinium-infected N. lugens line that possessed complete maternal transmission. Cardinium had a widespread distribution in tissues of N. lugens, and this infection decreased the fecundity and hatching rate of the host. Our findings emphasize the feasibility of transinfection of Cardinium in insects, which expands the range of endosymbionts that could be manipulated for pest control.

AbstractMany species of insects associated with cultivated rice do not over-winter in Korea and Japan, but migrate into these areas each year. To understand better the origins of these immigrations as well as the geographic structure of rice pests in Asian rice growing regions, intraspecific variation in two species of delphacid planthoppers, Nilaparvata lugens (Stål) and Sogatella furcifera Horvath, was examined. An 850 base pair region of mitochondrial DNA cytochrome oxidase-I (CO-I) was sequenced from a total of 71 individuals collected from 11 localities in seven countries: Korea, Philippines, China, Bangladesh, Malaysia, Vietnam and Thailand. In N. lugens, three haplotypes were found and all populations sampled shared a dominant haplotype. Localities in Korea contained two haplotypes and localities in China and the Philippines contained three. However, in samples from the Indochina peninsula no variation was detected either within or between populations, consistent with a hypothesis of regular migration and gene flow. These populations did not contain some haplotypes found in Korea, suggesting they were not the source of yearly immigration into Korea and, by extension, Japan. Populations from China did share haplotypes with Korea, which was consistent with the hypothesis that China was the source for yearly immigration into Korea. There was insufficient resolution to distinguish among populations in China. For N. lugens, the data suggested that populations south of the Red River Valley in Vietnam experienced regular mixing and were distinct from populations to the north which contributed to yearly immigrations. In S. furcifera, there was less differentiation among populations. Two haplotypes were found in all populations except Malaysia. The results for both species were consistent with seasonal weather data and indicated that more detailed analysis of DNA sequence data will be fruitful.
