In ectotherm species, environmental temperature plays a key role in development, growth, and survival. Thus, determining how temperature affects fish populations is of utmost importance to accurately predict the risk of climate change over fisheries and aquaculture, critical to warrant nutrition and food security in the coming years. Here, the potential effects of abnormal thermal regimes (24, 28 and 32 °C; TR24, TR28, and TR32, respectively) exclusively applied during embryogenesis in tropical gar (Atractosteus tropicus) has been explored to decipher the potential consequences on hatching and growth from fertilization to 16 days post-fertilization (dpf), while effects on skeletal development and body morphology were explored at fertilization and 16 dpf. Egg incubation at higher temperatures induced an early hatching and mouth opening. A higher hatching rate was obtained in eggs incubated at 28 °C when compared to those at 24 °C. No differences were found in fish survival at 16 dpf, with values ranging from 84.89 to 88.86%, but increased wet body weight and standard length were found in larvae from TR24 and TR32 groups. Thermal regime during embryogenesis also altered the rate at which the skeletal development occurs. Larvae from the TR32 group showed an advanced skeletal development, with a higher development of cartilaginous structures at hatching but reduced at 16 dpf when compared with the TR24 and TR28 groups. Furthermore, this advanced skeletal development seemed to determine the fish body morphology. Based on biometric measures, a principal component analysis showed how along development, larvae from each thermal regime were clustered together, but with each population remaining clearly separated from each other. The current study shows how changes in temperature may induce craniofacial and morphological alterations in fish during early stages and contribute to understanding the possible effects of global warming in early development of fish and its ecological implications.

Bradysia odoriphaga is an agricultural pest in China’s vegetable industry. In this study, pupae and adults were exposed to various non-lethal high-temperatures. The results demonstrated a decreased rate of eclosion once the pupae were exposed to temperatures exceeding 37 °C for 1 h. No effect on the lifespan of unmated female adults was observed after exposure to temperature stress, while unmated male adult lifespan decreased (>37 °C for 2 h). The size of the testis and ovaries for unmated male and female adults decreased, as did the fecundity and egg hatching rate for mated females. Compared with the control group (25 °C), the testis size of unmated male adults decreased after high-temperature stress followed by recovery at 25 °C for 1 h, though the size of the ovaries of female adults did not change. Additionally, the size of the testis and ovaries for unmated male and female adults decreased following high-temperature stress and 24 h of recovery at 25 °C. High temperatures affected males more than females; 37 °C is the critical temperature to control the population of B. odoriphaga. These results lay the foundation for the future development of environmentally friendly high-temperature prevention and pest-control strategies.

Yun7Ge is a giant egg mutant found in the silkworm variety Yun7. In comparison with the giant mutant Ge, the eggs of Yun7Ge are larger. The number of laid eggs and hatching rate of Yun7Ge are reduced, which is not conducive to reproduction. In this work, the target gene controlling giant egg trait is located on the Z chromosome and was determined through genetic analysis. Transcriptome results showed that phytanoyl-CoA dioxygenase domain-containing protein 1 (PHYHD1) on the Z chromosome was silenced, and the 25 chorion genes on chromosome 2 were remarkably downregulated. Sequence analysis showed that the 73.5 kb sequence including the PHYHD1 was replaced by a ~3.0 kb sequence. After knocking out the PHYHD1 by using CRISPR/Cas9, the chorion genes were significantly downregulated. Hence, the silencing of PHYHD1 leads to the downregulation of many chorion protein genes, thus directly causing giant eggs.

Abstract Background: The Lehmann Funnel Entry Trap has proven to be effective in catching and killing up to 70% of mosquitoes even in a high mosquito density setting. A medium-sized prototype was selected and deployed at large scale in Vallée du Kou 3 (VK3) in the Southwest of Burkina Faso to assess its entomological and sociological impact.Method: Overall, 1,313 traps impregnated with Pyriproxyfen (PPF), were deployed. Of them, 12 traps were randomly selected across the intervention village compared to houses without traps in the control village, Vallée du Kou 5 (VK5). Traps were placed at the windows while doors were blocked with curtains. Mosquitoes were collected in traps and matching houses in VK3 and in houses only in VK5, for nine days per month from July to October 2015. Collected mosquitoes were morphologically identified, counted, and preserved in 80% ethanol vials for subsequent analyses, including resistance genes and female mosquito age structure. The impact of the trap on mosquito density at community level was assessed by performing a pyrethrum spray catch (PSC) with bioassays to assess the effect of PPF. Results: Overall mosquito density was reduced by ~90% in all houses equipped with traps in VK3. At the community level, while mosquito density before intervention was 33% higher in VK3 than in VK5, it was 47% higher in VK5 after the intervention. Old female mosquito numbers increased in VK5 by 12% in October but not in VK3, indicating that the traps were cumulatively killing old females. The additional effect of PPF was to limit egg-laying, with a smaller number of eggs counted, and with a low hatching rate. Mosquitoes were highly resistant to pyrethroids with ~0.9 frequency of the kdr mutation. The trap was well accepted by the communities as 85.4% and 93.8% of interviewees in VK3 found the traps reducing mosquito bites with peaceful sleep respectively. Conclusion: The Lehmann Funnel Entry Trap has real potential to control malaria mosquito populations and can be widely used to sustain the global effort of malaria elimination.

Background: cattle tick Rhipicephalus (Boophilus) microplus poses serious problems for farmers in Brazil, especially because the parasite easily develops resistance to pesticide agents. For this reason, together with other factors including environmental, human and animal contamination and costs, alternative approaches have been sought for. Aims: this study sough to evaluate the efficiency of a tick biotherapic on tick-infested cows. Methods: 34 dairy Dutch cows were divided in 2 groups: one group received 100g/day of mineral salt supplement impregnated with tick biotherapic 12cH for 6 months, and then in alternate days with tick biotherapic 30cH to complete 28 months of treatment; the other group (control) received only the mineral salt supplement. After 28 months of treatment, engorged Rhipicephalus (boophilus) microplus females were collected in both groups, counted and weighed; in vitro tests were carried out to assess mass of ticks; egg mass; egg-hatching rate; and reproductive efficiency. Results: There was significant difference between both groups for all parameters evaluated; tick-mass (p = 0.0008); egg mass (p=0.0044); egg-hatching rate (p= 0.0017); and reproductive efficiency (p = 0.0044). Conclusion: treatment with tick biotherapic significantly decreased the mass of engorged females, deposition and hatching rate of eggs, resulting consequently in the decrease of the reproductive efficiency of ticks.

The purpose of the research is study of some reproductive characteristics of the biology of ticks of the family Ixodidae which are widespread in the southeast of the North Caucasus.Materials and methods. In 2000–2010, 9 series of experiments were carried out in the conditions of the plain belt in Dagestan. The first, second, and third series of experiments were intended to find out the duration of the tick searching for a place to fix on the animal, the duration of the skin incised and the proboscis inserted into the wound, and the engorgement duration; and the fourth series was intended to clarify the tick wintering on the animal. Such experiments were performed on two-year-old calf bulls on the Educational and Experimental Farm of the Dagestan State Agrarian University with Boophilus annulatus, Rhipicephalus bursa, Hyalomma detritum, H. scupense, and H. anatolicum in 2000. Five hungry adult female ticks were placed onto all 5 calf bulls in the middle third of the neck. We conducted experiments on searching for a place for egg-laying, egg-laying duration in the environment, number of eggs in a clutch, duration of the larva formed and the hatching rate, distance of the larvae from the hatching place, and larvae death rate in five specially equipped sites of pastures of 1 m2 . Five adult female ticks of B. annulatus, R. bursa, H. detritum, H. scupense, and H. anatolicum were placed after engorgement in each biological site.Results and discussion. Ixodid ticks perform their natural physiological functions associated with feeding and reproduction with significant time fluctuations. The search for a place to fix on the animal took 4–8 hours; the skin incised and proboscis inserted in the wound took 2–5 hours; the engorgement took 4–6 hours; the search for an egg-laying place took 10– 22 hours; the egg-laying duration in the environment was 18–23 hours; the number of eggs in a clutch was 4–13K; the duration of the larva formed and the hatching rate was 10–15 days and 58.0–75% respectively; the distance of the larva from the hatching place was 0.5–1.5 m; death rate of the larva in captivity was 100%; and the tick wintering on the animal was near the neck, ears, in the dewlap, the flank and the udder. These features are typical for the analyzed tick species in the southeast of the North Caucasus and, probably, for other taxa of these genera.

Carbendazim (CAR) and enrofloxacin (ENF) are frequently detected in fruits and meat products, respectively. Since most people consume fruits, vegetables, and meat products, combined exposure is possible, necessitating further evaluation of toxic interactions. In this study, the developmental toxicity of separate and combined exposure was examined in zebrafish embryos. Carbendazim exposure at 0.79 mg/L and above significantly affected developmental parameters, while enrofloxacin alone had no substantial effects on these developmental parameters within the selected concentration range (0.10–0.40 mg/L). Surprisingly, ENF antagonized the CAR-evoked reduction in the 48 hpf (hours post-fertilization) hatching rate and the increases in the 96 hpf malformation and lethality rates. The results revealed that the antagonism might be associated with reciprocal effects of these compounds on metabolism-related genes, such as cyp7a1 and apoa1a. These results reveal a complex interaction between ENF and CAR on metabolic regulation during development and highlight the importance of combined assessment for agents with the potential for simultaneous exposure.

Difenoconazole (DIF) and dimethomorph (DIM) are widely used pesticides frequently detected together in environmental samples, so the deleterious effects of combined exposure warrant detailed examination. In this study, the individual and combined effects of DIM and DIF on conventional developmental parameters (hatching rate, deformity rate, lethality) and gene expression were measured in embryonic zebrafish. Both DIF and DIM interfered with normal zebrafish embryo development, and the most sensitive toxicity index for both was 96 h post-fertilization (hpf) deformity rate (BMDL10 values of 0.30 and 1.10 mg/L, respectively). The combination of DIF and DIM had mainly synergistic deleterious effects on 96 hpf deformity and mortality rates. Transcriptome analysis showed that these compounds markedly downregulated expression of mcm family genes, cdk1, and cdc20, thereby potentially disrupting DNA replication and cell cycle progression. Enhanced surveillance for this pesticide combination is recommended as simultaneous environmental exposure may be substantially more harmful than exposure to either compound alone.

The RING-type E3 ubiquitin ligases play an important role in plant growth, development, and defense responses to abiotic stresses and pathogens. However, their roles in the resistance of plants to herbivorous insects remain largely unknown. In this study, we isolated the rice gene OsJMJ715, which encodes a RING-domain containing protein, and investigated its role in rice resistance to brown planthopper (BPH, Nilaparvata lugens). OsJMJ715 is a nucleus-localized E3 ligase whose mRNA levels were upregulated by the infestation of gravid BPH females, mechanical wounding, and treatment with JA or ABA. Silencing OsJMJ715 enhanced BPH-elicited levels of ABA, JA, and JA-Ile as well as the amount of callose deposition in plants, which in turn increased the resistance of rice to BPH by reducing the feeding of BPH and the hatching rate of BPH eggs. These findings suggest that OsJMJ715 negative regulates the BPH-induced biosynthesis of ABA, JA, and JA-Ile and that BPH benefits by enhancing the expression of OsJMJ715.

Ikan koi merupakan salah satu komoditas ikan hias yang populer dibudidayakan di Indonesia. Ikan ini memiliki daya tarik karena warnanya yang sangat indah dan sering dijadikan sebagai ikan kontes. Setiap tahunnya permintaan ikan koi terus meningkat baik di pasar dalam maupun di luar negeri, sehingga ikan ini memiliki prospek usaha yang menjanjikan. Salah satu prospeknya yaitu pada kegiatan pembenihan yang meliputi kegiatan persiapan wadah, pemeliharaan induk, pemijahan induk, penetasan telur, pemeliharaan larva, pemeliharaan benih, pemeliharaan sangkal, pengelolaan kualitas air, pencegahan hama dan penyakit, serta pemanenan. Salah satu sentra pembenihan ikan koi yaitu Mizumi Farm yang berlokasi di Sukabumi, Jawa Barat. Produktivitas ikan koi di lokasi tersebut diantaranya jumlah telur yang dihasilkan sekitar 116.452 butir, fertilization rate (FR) 81 %, hatching rate (HR) 75 %, survival rate (SR) larva 63%, SR benih/putihan sebesar 63% dan SR sangkal sebesar 74%. Ikan yang dihasilkan berjumlah 634 ekor dengan ukuran 5 cm. Pemasaran benih ikan koi di Mizumi Farm diantaranya mitra binaan, penghoby daerah Sukabumi dan kota lain.
