Polygalacturonases represent an important member of pectinases group of enzymes with immense industrial applications. The activity of exo-polygalacturonase produced by Aspergillus niger was studied in solid state fermentation (SSF) using Nephrolepis biserrata leaves as substrate. Central composite design (CCD) was used to optimize four significant variables resulted from the screening process that has been initially analyzed for the production of exo-polygalacturonase which are incubation time, temperature, concentration of pectin and moisture content. The optimum exo-polygalacturonase production obtained was 54.64 U/g at 120 hours of incubation time, temperature at 340C, 5.0 g/L of pectin concentration and 75.26% of moisture content. For partial characterization of exo-polygalacturonase, the optimum temperature and pH were obtained at 50°C and pH 4.0, respectively. SDS-PAGE analysis showed that molecular weight of exo-polygalacturonase were 35 and 71 kDa. This study has revealed a significant production of exo-polygalacturonase by A. niger under SSF using cheap and easily available substrate and thus could found immense potential application in industrial sectors and biotechnology

Amyloglucosidase (AMG) and exo-polygalacturonase (exo-PG) were simultaneously produced by two different strains of Aspergillus niger in solid-state fermentation (SSF) using defatted rice-bran as substrate. The effect of Aspergillus niger strain (t0005/007-2 and/or CCT 3312), inoculum type (spore suspension or fermented bran) and addition of inducers (pectin and/or starch) to the culture media was studied using a 3² x 2¹ factorial experimental design. The production of AMG and exo-PG was significantly affected by fungal strain and inoculum type but inducers had no effect. The maximum yields obtained were 1310 U/g dm for AMG using a spore suspension of A. niger CCT 3312 and 50.2 U/g dm for exo-PG production, using A. niger t0005/007-2 and fermented bran as inoculum. The yields obtained represented acceptable values in comparison with data available in the literature and indicated that defatted rice-bran was a good nutrient source.

The purpose of this research is to evaluate the coffee pulp, a by-product of coffee processing, as substrate for polygalacturonase production by solid state fermentation. In addition, it is a way to take advantage of the coffee pulp. Characterization of the coffee pulp revealed a high content of nutrients for fungi growth, such as reducing sugars (5.4% of dry pulp), proteins (9.4% of dry pulp), pectins (20.5% of dry pulp), which are inducers of pectic hydrolases production and source of carbon after degradation, and caffeine (1.4% of dry pulp), among others. The characterization of the cell-wall of coffee pulp revealed, after polysaccharides fractionation, the content in cell-wall pectins (25.5%), hemicelluloses (11.5%) and cellulosic residue (44%). A strain of Aspergillus niger, called van Thiegem, has been selected as a good producer of polygalacturonases (60 U/ml) using the coffee pulp as the growth substrate. During fermentation, reducing sugars, caffeine and phenolic compounds were consumed till almost exhaustion. A partial characterization of the polygalacturonase using high methoxyl pectin as substrate indicates an optimal pH of 4.0 and 45oC as optimal temperature, which are good values for the use of the enzyme in vegetable processing, including coffee processing.

The study demonstrates the starter construction and its solid state fermentation (SSF) using rice straw as substrate for lignocellulases production. The starter is constructed by natural microflora growing on rice straw additionally reinforced with Aspergillus niger ZJU-RYD1, using the SSF substrate rice straw with 90% moisture content, successfully produced the integrate and high active lignocellulases at about 72 h, i.e. CMCase 12.44 U/g, cellobiase 12.05 U/g, FPAase 3.18 U/g, xylanase 365.61 U/g, lassase 2.48 U/g. In this study the strategy is satisfactorily developed to construct the starter for high effective and low-cost production of lignocellulases using rice straw as substrate, based on natural microflora and additional reinforcing with special strain.

Nowadays, a lot of interest has been given to the development of cost-effective and efficient enzyme production technologies. Laccase enzymes are widely used in biotechnological, environmental and industrial sectors. Due to the cost-effectiveness of the solid-state fermentation (SSF) process, it is widely used to produce a broad range of biological products. In this study, optimization of moisture content, temperature, pH, and inoculum size were studied to enhance laccase production ability of Pleurotus sajor-caju in SSF by using One Factor At Time (OFAT) and Response Surface Methodology (RSM). OFAT was used as a baseline study for deducing the experimental design of RSM. The highest production of laccase enzyme (1450 U/g) by Pleurotus sajor-caju on wheat straw was observed at 26°C, 6.0 pH, 72.5 % moisture content, 7.5% inoculum size, 1% fructose and 0.5 % peptone. Unlike the conventional inoculum preparation method, here the inoculum was generated by the spawning method for SSF. The molecular weight of partially purified laccase from Pleurotus sajor-caju was estimated to be around 62 K Da using SDS PAGE. The activity staining of laccase was observed as a zymogram on Native PAGE using ABTS as a substrate. Lignin degradation of wheat straw and its structural disruption due to laccase was observed by Scanning Electron Microscopy (SEM).

AbstractObjective: Glucoamylase is a hydrolyzing enzyme with several industrial applications. Glucoamylase was produced via a solid state fermentation by three naturally occurring zygomycetes fungi of Mucor indicus, Mucor hiemalis, and Rhizopus oryzae on wheat bran.Methods: The effects of cultivation temperature, medium moisture content, and cultivation time on the enzyme production were investigated. Experiments were designed with an orthogonal central composite design on the three variables using response surface methodology (RSM).Results: For glucoamylase production, the optimum temperature and medium moisture content for the three fungi were 26.6°C and 71.8%, respectively. The optimum cultivation time for M. hiemalis and R. oryzae was 33.1 h, while it was 66.8 h for M. indicus. At optimum conditions, glucoamylase production by M. indicus, M. hiemalis, and R. oryzae was respectively 255.3, 272.3, and 1545.3 U per g dry substrate.Conclusion: R. oryzae is a suitable candidate for industrial production of glucoamylase.

Exo-polygalacturonase was produced by Aspergillus niger ATCC 120120 in a solid-state fermentation using Nephrolepis biserrata leaves. Factors affecting the production of exo-polygalacturonase were determined using a two-level fractional factorial design. The screening process for six factors; pH, incubation time, temperature, pectin concentration, inoculum size and moisture content, that influence the production of exo-polygalacturonase by A. niger was performed. The result of variance analysis (ANOVA) suggested that there were four statistically significant (P < 0.005) factors in the production of exo-polygalacturonase by A. niger. These factors were incubation time, temperature, pectin concentration and moisture content. The statistical analysis shows that the linear mathematical model is significant with coefficient of determination (R2) value of 0.9711. The optimum production of exo-polygalacturonase obtained using the model in this study was at 40.00 U/g.
