Induced Cytochrome P4501A in Winter Flounder, Pleuronectes americanus, from Offshore and Coastal Sites

Cytochrome P4501A (CYP1A), Aroclor 1254 (A1254), and 3,3′,4,4′-tetrachlorobiphenyl (TCB) were measured in liver of winter flounder, Pleuronectes americanus, from Boston Harbor, Mass., Hempsted Harbor, N.Y., Niantic, Conn., and an offshore site, Georges Bank. We also measured CYP1A content and activity in flounder from Passamaquoddy Bay, N.B. Concentrations of A1254 and TCB were the least in fish from Georges Bank (0.46 and 0.002 μg∙g dry weight−1, respectively); concentrations in fish from Boston, Niantic, and Hempsted ranged from 7.6 to 11.3 μg∙g−1 and from 0.013 to 0.024 μg∙g−1. Immunodetected microsomal CYP1A contents (expressed as scup P450E equivalents) were 0.17 and 0.19 nmol∙mg−1 in fish from Georges Bank and Passamaquoddy and 0.25–0.41 nmol∙mg−1 in fish from Boston, Niantic, and Hempsted. Ethoxyresorufin-O-deethylase specific activities likewise were greater in fish from Boston, Niantic, and Hempsted (1.7–2.4 nmol∙min−1∙mg−1) than in fish from Georges Bank or Passamaquoddy (0.83 and 0.61 nmol∙min−1∙mg−1). CYP1A content and activity were correlated with hepatic concentrations of A1254 and TCB. These data, together with data reported in previous studies, indicate that strong induction of CYP1A protein occurs in winter flounder populations along most of the industrialized east coast and that induction of CYP1A is common, but less strong, at sites distant from the urban centers of the Northeast.

7-Ethoxyresorufin-O-deethylase (EROD), benzo[a]pyrene hydroxylase (BaPH), and cytochromes P-450 (cyt-P450) and b5 (cyt-b5) varied annually in winter flounder (Pleuronectes americanus) collected in August of 1987, 1988, and 1989 from a coal tar contaminated estuary (Sydney, Nova Scotia, Canada). For August 1989, with fish available from all estuary areas, these indices correlated strongly with a spatial (along estuary) gradient in PAH in bottom sediments (7.19 ± 6.59–191 ± 184 μg g dry weight−1). Mean EROD activities in flounder near the coal tar source were up to seven times those in other estuary areas and paralleled sediment PAH loadings; however, standard deviations were high. Correlations for all MFO indices and sediment PAH were obtained in female flounder (P < 0.01: EROD, cyt-b5, cyt-P450; P < 0.02: BaPH). For male flounder the trend was similar, but only cyt-P450 correlated with sediment PAH (P < 0.017). BaPH activity was highest near the coal tar source but was more variable and less sensitive to pollutant levels than EROD activity. Somatic indices in fish from Sydney estuary and St. George's Bay were similar. Winter flounder are vulnerable to PAH-induced MFO activities from coal tar contaminated sediments, but MFO induction does not occur equally in all fish; single-season or single-year data must be interpreted with caution.

The present article describes changes in specific activities and fluctuations in the ratio of natural 40K and artificial 137Cs radionuclides in soil samples taken from different places of Lithuanian territory. The samples of soil have been selected from the districts polluted after the accident in Chernobyl nuclear plant performing nuclear testing operations. The study has established the main physical and chemical properties of soil samples and their impact on the concentration of 40K activities. 137Cs/40K specific activities in soil have been observed under the dry weight of the sample that varied from 0.0034 to 0.0240. The results of the study could be used for establishing and estimating 137Csand 40K transfer in the system “soil-plant”. Santrauka Straipsnyje nagrinėjama gamtinės (40K) ir dirbtinės (137Cs) kilmės radionuklidų savitųjų aktyvumų ir jų santykio kaita skirtinguose Lietuvos teritorijos dirvožemiuose. Dirvožemio mėginiai parinkti iš vietovių, kurios buvo labiau užterštos po Černobylio atominės elektrinės avarijos ir buvusių branduolinių bandymų. Tyrimo metu nustatytos pagrindinės fizinės cheminės dirvožemio savybės ir jų poveikis 40K aktyvumų koncentracijai. 137Csir 40K savitieji aktyvumai dirvožemyje tirti esant sausam mėginio svoriui. 137Cs savitieji aktyvumai sausame dirvožemyje svyravo nuo 1,1±1,0 iki 14,3±0,9 Bq/kg, o 40K savitieji aktyvumai – nuo 326±29 iki 740±15 Bq/kg. 137Csir 40K savitųjų aktyvumų santykis skirtingų vietovių dirvožemiuose kito nuo 0,0034 iki 0,0240 Bq/kg. Tyrimo rezultatai gali būti panaudoti, nustatant ir įvertinant 137Csir 40K pernašą sistemoje dirvožemis–augalai.

Wang (Can. J. Botany, 38, 635–642 (1960)) concluded that the accumulation of radioactivity observed on radioautographs at infection sites on rusted leaves fed with C14-labelled substances was 'apparent' rather than real. The ‘accumulation ratio’ is defined as the ratio of the specific activities (c.p.m./mg dry weight of intact tissue) of rust-infected to uninfected areas of infected leaves. Theoretical considerations relating to the radioautography of leaves labelled with C14 and to the measurement of ‘accumulation ratios’ by extraction of C14-labelled substances from rusted and uninfected segments of infected leaves, as well as experimental data, show that Wang's conclusion is not generally applicable.Experimentally, it was shown using polymethacrylate C14 sources that differences in distance between sources and X-ray film of the order of 100 μ had no effect on the intensity of autoradiographs. Rust-infected leaves, fed with radioactive glucose, were radiographed between X-ray plates. Localization of radioactivity at infection sites was observed on both ‘dorsal’ and ‘ventral’ radiographs, indicating a real accumulation per unit area. Ventral were more radioactive than dorsal surfaces. The main development of the fungus occurred on the former. Radioautography revealed that C14 from glucose-1-C14, glucose-6-C14, and uniformly labelled glucose fed to excised wheat leaves became localized at 10-day-old rust infections in 2 hours. ‘Accumulation ratios’ calculated from the specific activity of leaf segments remained close to 1.0 for at least 6 hours after introduction of the tracer, but increased to more than 2 after 24 hours. When ‘accumulation ratios’ were calculated from the specific activities of individual pustules (excised with a punch 1 mm in diameter) and interpustular disks, values greater than 1 were observed in 2 hours, thus confirming the results of autoradiography. Differences between the ‘accumulation ratios’ observed with glucose-6-C14 and glucose-1-C14 were consistent with an increased role of the pentose phosphate pathway at infection sites. Incorporation of C14 from uniformly labelled glucose into the alcohol-insoluble fraction of rusted leaf segments was 2.5-fold that in uninfected segments in 6 hours and 3.65-fold in 24 hours. The humin formed during hydrochloric acid hydrolysis accounted for approximately 50% of the activity of the alcohol-insoluble material. The ‘accumulation ratio’ for the alcohol-soluble material was only 1.56 after 24 hours.All the results support the view (Shaw and Samborski, Can. J. Botany, 34, 389–405 (1956)) that there is a quantitative, metabolically dependent accumulation of C14 from radioactive glucose at vigorous rust infections. The relative roles of fungus and host in this process are discussed briefly.
