RAPD-based genetic linkage maps of yellow passion fruit (Passiflora edulis Sims. f. flavicarpa Deg.)

A single cross between two clones of passion fruit (Passiflora edulis Sims. f. flavicarpa Deg., 2n = 18) was selected for genetic mapping. The mapping population was composed of 90 F1 plants derived from a cross between 'IAPAR 123' (female parent) and 'IAPAR 06' (male parent). A total of 380 RAPD primers were analyzed according to two-way pseudo-testcross mapping design. The linkage analysis was performed using Mapmaker version 3.0 with LOD 4.0 and a maximum recombination fraction (θ) of 0.30. Map distances were estimated using the Kosambi mapping function. Linkage maps were constructed with 269 loci (2.38 markers/primer), of which 255 segregated 1:1, corresponding to a heterozygous state in one parent and null in the other. The linkage map for 'IAPAR123' consisted of 135 markers. A total of nine linkage groups were assembled covering 727.7 cM, with an average distance of 11.20 cM between framework loci. The sizes of the linkage groups ranged from 56 to 144.6 cM. The linkage map for 'IAPAR 06' consisted of 96 markers, covering 783.5 cM. The average distance between framework loci was 12.2 cM. The length of the nine linkage groups ranged from 20.6 to 144.2 cM. On average, both maps provided 61% genome coverage. Twenty-four loci (8.9%) remained unlinked. Among their many applications, these maps are a starting point for the identification of quantitative trait loci for resistance to the main bacterial disease affecting passion fruit orchards in Brazil, caused by Xanthomonas campestris pv. passiflorae, because parental genotypes exhibit diverse responses to bacterial inoculation.Key words: Passiflora, genetic mapping, molecular markers, pseudo-testcross mapping strategy.

AbstractGenomic datasets continue to increase in size and ease of production for a wider selection of species including non-model organisms. For many of these species highly contiguous and well-annotated genomes are unavailable due to their prohibitive complexity and cost. As a result, a common starting point for genomic work in non-model species is the production of a linkage map, which involves the grouping and relative ordering of genetic markers along the genome. Dense linkage maps facilitate the analysis of genomic data in a variety of ways, from broad scale observations regarding genome structure e.g. chromosome number and type or sex-related structural differences, to fine scale patterns e.g. recombination rate variation and co-localisation of differentiated regions. Here we present both a sex-averaged and sex-specific linkage maps for Coregonus sp. “Albock” containing 5395 single nucleotide polymorphism (SNP) loci across 40 linkage groups to facilitate future investigation into the genomic basis of whitefish adaptation and speciation. The map was produced using restriction-site associated digestion (RAD) sequencing data from two wild-caught parents and 156 F1 offspring in Lep-MAP3. We discuss the differences between our sex-avagerated and sex-specific maps and identify synteny between C. sp. “Albock” linkage groups and the Atlantic salmon (Salmo salar) genome. Our synteny analysis confirms that many patterns of homology observed between Atlantic salmon and Oncorhynchus and Salvelinus species are also shared by members of the Coregoninae subfamily.

Green manures can replace or supplement mineral fertilization and add organic matter to the soils, ensuring greater sustainability to fruit growing in semiarid regions. Biological fixation, transfer and balance of nitrogen were determined on an irrigated yellow passion fruit orchard (Passiflora edulis Sims) intercropped separately with three cover crops: sunn hemp, Crotalaria juncea (L.); pigeon pea, Cajanus cajan (L.) Mill; and jack bean, Canavalia ensiformis (L.) DC. In a fourth treatment, legumes were not planted, but spontaneous vegetation was left to grow freely. The legumes were croped for 90 days in three lines (0.5 m apart) inside the passion fruit plant lines (2.5 m apart). Fixation and transfers were determined by the 15N natural abundance technique, using sunflower as a reference plant. The three planted legumes nodulated abundantly and fixed nitrogen in high proportions (between 50 and 90% of their N), forming symbiosis with bacteria naturally established in the soil. Jack bean produced more biomass than sunn hemp and pigeon pea, and as much as the spontaneous plants, of which 23% were legumes. The amounts of fixed N (150, 43, 30 and 29 kg ha-1) were determined mainly by the biomass of legumes. More than 40% of the N of passion fruit plants came from the biological nitrogen fixation of the intercropped jack bean, which provided an amount of N higher than that exported in the fruits, generating a positive balance of more than 100 kg ha-1. Therefore, it is recommended to intercrop jack bean in irrigated passion fruit orchards.

O alagamento no solo é um fenômeno que reduz a sobrevivência, o crescimento e o desenvolvimento de plantas, podendo ocasionar queda na produtividade quando essas são agricultáveis. O maracujazeiro (Passiflora edulis Sims) é uma frutífera comercial com propriedades medicinais cuja produção incentiva a irrigação; e não são encontrados estudos que apresentem efeitos do excesso de água no solo sobre estas plantas. O objetivo deste trabalho foi verificar alterações morfoanatômicas e fisiológicas no desenvolvimento inicial de plântulas de maracujazeiro expostas a diferentes níveis de água no solo. As plântulas foram organizadas em três tratamentos: solo com capacidade de campo, pré-submergido e alagado. Após sete dias, foram feitas análises de trocas gasosas, potencial hídrico, quantificação de prolina e cortes anatômicos de raízes. O solo alagado influencia as trocas gasosas e afeta negativamente a fotossíntese. Não houve alterações no potencial hídrico nem na quantificação de prolina entre os tratamentos. Os tratamentos pré-submergido e alagado apresentaram aerênquima e aumento no diâmetro das raízes e na espessura das barreiras apoplásticas e do córtex. O maracujazeiro apresentou alterações fisiológicas e anatômicas que permitiram sua sobrevivência em condições pré-submergidas e alagadas, o que significa que plântulas jovens dessa espécie suportam maiores quantidades de água no solo.

A evapotranspiração máxima da cultura (ETc) e a evapotranspiração de referência (ETo) foram medidas em lisímetros de nível de lençol freático constante, durante o período de 29 de setembro de 2000 a 20 de julho de 2001. Os valores da ETc e ETo foram de 954,98 mm e  1.069,21 mm, respectivamente, durante todo o período medido. The maximum crop water demand (ETc) and the Reference Evapotranspiration (ETo), measured by lysimeters, were 954.98 mm and 1,069.21 mm, respectively, during all period measured.
