Efficient photocatalytic oxygenation of alkenes by water soluble sensitizer in organic–water biphasic media

A new green, environmentally and economically photooxygenation of olefins by molecular oxygen in the presence of 4-carboxyl tetra phenyl porphyrin (H2TCPP), Rose Bengal (RB), Methylene Blue (MB) and some metallosensitizers (MS) such as cobalt phthalocyaninesulfonate (Coph(SO3H)[Formula: see text], MnTCPPCl, FeTCPPCl and SbTPP(OH)2 as water–soluble sensitizers in organic–water biphasic media at room temperature under visible light was illustrated. The products were obtained with 100% conversion by H2TCPP and 100% selectivity by Coph(SO3H)4. Also in this study, singlet oxygen production as the major route for oxidation of olefins was proved.

A series of porphyrin derivatives with one to four maltohexaose moieties in their meso positions have been synthesized. Zinc or free-base m-THPP (5,10,15,20-tetrakis(m-hydroxyphenyl)-porphyrin) was used as the porphyrin platform. The reaction of m-THPP with 3-iodopropyl nonadecaacetylmaltohexaoside afforded a mixture of all possible combinations of glycoconjugated porphyrins having one to four maltohexaose moieties; monoglycosylated (Ac-1), bisglycosylated (Ac-cis-2 and Ac-trans-2), triglycosylated (Ac-3), and tetraglycosylated (Ac-4) porphyrins were obtained in 11–26% yield. Removal of acetyl groups at maltohexaose moiety afforded highly water-soluble glycoconjugated porphyrins 1–4. Zinc derivatives were synthesized in a similar manner. These maltohexaose-linked porphyrins exhibit remarkable water-solublity (530 mg/mL for 4). The singlet oxygen production ability upon visible light irradiation is not affected by the maltohexaose substitution. Photo- and dark cytotoxicities of the maltohexaose-conjugated porphyrins 1–4 and Zn-1–4 were examined against a HeLa cell line, which showed that the mono-maltohexaosylated derivative (1 and Zn-1) was the most effective photosensitizer for PDT.

Computational tools have been used to study the photophysical and photochemical features of photosensitizers in photodynamic therapy (PDT) –a minimally invasive, less aggressive alternative for cancer treatment. PDT is mainly based by the activation of molecular oxygen through the action of a photoexcited sensitizer (photosensitizer). Temoporfin, widely known as mTHPC, is a second-generation photosensitizer, which produces the cytotoxic singlet oxygen when irradiated with visible light and hence destroys tumor cells. However, the bioavailability of the mostly hydrophobic photosensitizer, and hence its incorporation into the cells, is fundamental to achieve the desired effect on malignant tissues by PDT. In this study, we focus on the optical properties of the temoporfin chromophore in different environments –in <i>vacuo</i>, in solution, encapsulated in drug delivery agents, namely cyclodextrin, and interacting with a lipid bilayer.

Novel polymeric photosensitizers for singlet oxygen production in water are described. The polymers contain rose bengal (4,5,6,7-tetrachloro-2',4',5',7'­tetraiodo-fluorescein) chromophores covalently attached to the polymer chain. The quantum yields of singlet oxygen formation and photooxidation of phenol in water have been determined. The methods of separation of photosensitizer from the reactants after completion of reaction have been established.

Herein, we describe a copper photoredox catalyzed synthesis of acetamide via regioselective C–N coupling of arylamines with terminal alkynes using molecular oxygen (O2) as an oxidant at room temperature under visible light irradiation (47 examples).
