Previous studies in neurologically intact subjects have shown that motor coordination can be described by task-dependent combinations of a few muscle synergies, defined here as a fixed pattern of activation across a set of muscles. Arm function in severely impaired stroke survivors is characterized by stereotypical postural and movement patterns involving the shoulder and elbow. Accordingly, we hypothesized that muscle synergy composition is altered in severely impaired stroke survivors. Using an isometric force matching protocol, we examined the spatial activation patterns of elbow and shoulder muscles in the affected arm of 10 stroke survivors (Fugl-Meyer <25/66) and in both arms of six age-matched controls. Underlying muscle synergies were identified using non-negative matrix factorization. In both groups, muscle activation patterns could be reconstructed by combinations of a few muscle synergies (typically 4). We did not find abnormal coupling of shoulder and elbow muscles within individual muscle synergies. In stroke survivors, as in controls, two of the synergies were comprised of isolated activation of the elbow flexors and extensors. However, muscle synergies involving proximal muscles exhibited consistent alterations following stroke. Unlike controls, the anterior deltoid was coactivated with medial and posterior deltoids within the shoulder abductor/extensor synergy and the shoulder adductor/flexor synergy in stroke was dominated by activation of pectoralis major, with limited anterior deltoid activation. Recruitment of the altered shoulder muscle synergies was strongly associated with abnormal task performance. Overall, our results suggest that an impaired control of the individual deltoid heads may contribute to poststroke deficits in arm function.

The musculoskeletal redundancy of the body provides multiple solutions for performing motor tasks. We have proposed that the nervous system solves this unconstrained problem through the recruitment of motor modules or functional muscle synergies that map motor intention to action. Consistent with this hypothesis, we showed that trial-by-trial variations in muscle activation for multidirectional balance control in humans were constrained by a small set of muscle synergies. However, apparent muscle synergy structures could arise from characteristic patterns of sensory input resulting from perturbations or from low-dimensional optimal motor solutions. Here we studied electromyographic (EMG) responses for balance control across a range of biomechanical contexts, which alter not only the sensory inflow generated by postural perturbations, but also the muscle activation patterns used to restore balance. Support-surface translations in 12 directions were delivered to subjects standing in six different postural configurations: one-leg, narrow, wide, very wide, crouched, and normal stance. Muscle synergies were extracted from each condition using nonnegative matrix factorization. In addition, muscle synergies from the normal stance condition were used to reconstruct muscle activation patterns across all stance conditions. A consistent set of muscle synergies were recruited by each subject across conditions. When balance demands were extremely different from the normal stance (e.g., one-legged or crouched stance), task-specific muscle synergies were recruited in addition to the preexisting ones, rather generating de novo muscle synergies. Taken together, our results suggest that muscle synergies represent consistent motor modules that map intention to action, regardless of the biomechanical context of the task.

We recently showed that four muscle synergies can reproduce multiple muscle activation patterns in cats during postural responses to support surface translations. We now test the robustness of functional muscle synergies, which specify muscle groupings and the active force vectors produced during postural responses under several biomechanically distinct conditions. We aimed to determine whether such synergies represent a generalized control strategy for postural control or if they are merely specific to each postural task. Postural responses to multidirectional translations at different fore-hind paw distances and to multidirectional rotations at the preferred stance distance were analyzed. Five synergies were required to adequately reconstruct responses to translation at the preferred stance distance—four were similar to our previous analysis of translation, whereas the fifth accounted for the newly added background activity during quiet stance. These five control synergies could account for >80% total variability or r2 > 0.6 of the electromyographic and force tuning curves for all other experimental conditions. Forces were successfully reconstructed but only when they were referenced to a coordinate system that rotated with the limb axis as stance distance changed. Finally, most of the functional muscle synergies were similar across all of the six cats in terms of muscle synergy number, synergy activation patterns, and synergy force vectors. The robustness of synergy organization across perturbation types, postures, and animals suggests that muscle synergies controlling task-variables are a general construct used by the CNS for balance control.

AbstractWalking and running are mechanically and energetically different locomotion modes. For selecting one or another, speed is a parameter of paramount importance. Yet, both are likely controlled by similar low-dimensional neuronal networks that reflect in patterned muscle activations called muscle synergies. Here, we investigated how humans synergistically activate muscles during locomotion at different submaximal and maximal speeds. We analysed the duration and complexity (or irregularity) over time of motor primitives, the temporal components of muscle synergies. We found that the challenge imposed by controlling high-speed locomotion forces the central nervous system to produce muscle activation patterns that are wider and less complex relative to the duration of the gait cycle. The motor modules, or time-independent coefficients, were redistributed as locomotion speed changed. These outcomes show that robust locomotion control at challenging speeds is achieved by modulating the relative contribution of muscle activations and producing less complex and wider control signals, whereas slow speeds allow for more irregular control.

Several recent studies have used matrix factorization algorithms to assess the hypothesis that behaviors might be produced through the combination of a small number of muscle synergies. Although generally agreeing in their basic conclusions, these studies have used a range of different algorithms, making their interpretation and integration difficult. We therefore compared the performance of these different algorithms on both simulated and experimental data sets. We focused on the ability of these algorithms to identify the set of synergies underlying a data set. All data sets consisted of nonnegative values, reflecting the nonnegative data of muscle activation patterns. We found that the performance of principal component analysis (PCA) was generally lower than that of the other algorithms in identifying muscle synergies. Factor analysis (FA) with varimax rotation was better than PCA, and was generally at the same levels as independent component analysis (ICA) and nonnegative matrix factorization (NMF). ICA performed very well on data sets corrupted by constant variance Gaussian noise, but was impaired on data sets with signal-dependent noise and when synergy activation coefficients were correlated. Nonnegative matrix factorization (NMF) performed similarly to ICA and FA on data sets with signal-dependent noise and was generally robust across data sets. The best algorithms were ICA applied to the subspace defined by PCA (ICAPCA) and a version of probabilistic ICA with nonnegativity constraints (pICA). We also evaluated some commonly used criteria to identify the number of synergies underlying a data set, finding that only likelihood ratios based on factor analysis identified the correct number of synergies for data sets with signal-dependent noise in some cases. We then proposed an ad hoc procedure, finding that it was able to identify the correct number in a larger number of cases. Finally, we applied these methods to an experimentally obtained data set. The best performing algorithms (FA, ICA, NMF, ICAPCA, pICA) identified synergies very similar to one another. Based on these results, we discuss guidelines for using factorization algorithms to analyze muscle activation patterns. More generally, the ability of several algorithms to identify the correct muscle synergies and activation coefficients in simulated data, combined with their consistency when applied to physiological data sets, suggests that the muscle synergies found by a particular algorithm are not an artifact of that algorithm, but reflect basic aspects of the organization of muscle activation patterns underlying behaviors.

There is increasing evidence that including sex as a biological variable is of crucial importance to promote rigorous, repeatable and reproducible science. In spite of this, the body of literature that accounts for the sex of participants in human locomotion studies is small and often produces controversial results. Here, we investigated the modular organization of muscle activation patterns for human locomotion using the concept of muscle synergies with a double purpose: i) uncover possible sex-specific characteristics of motor control and ii) assess whether these are maintained in older age. We recorded electromyographic activities from 13 ipsilateral muscles of the lower limb in young and older adults of both sexes walking (young and old) and running (young) on a treadmill. The data set obtained from the 215 participants was elaborated through non-negative matrix factorization to extract the time-independent (i.e., motor modules) and time-dependent (i.e., motor primitives) coefficients of muscle synergies. We found sparse sex-specific modulations of motor control. Motor modules showed a different contribution of hip extensors, knee extensors and foot dorsiflexors in various synergies. Motor primitives were wider (i.e., lasted longer) in males in the propulsion synergy for walking (but only in young and not in older adults) and in the weight acceptance synergy for running. Moreover, the complexity of motor primitives was similar in younger adults of both sexes, but lower in older females as compared to older males. In essence, our results revealed the existence of small but defined sex-specific differences in the way humans control locomotion and that these strategies are not entirely maintained in older age.

Postural control is a natural behavior that requires the spatial and temporal coordination of multiple muscles. Complex muscle activation patterns characterizing postural responses suggest the need for independent muscle control. However, our previous work shows that postural responses in cats can be robustly reproduced by the activation of a few muscle synergies. We now investigate whether a similar neural strategy is used for human postural control. We hypothesized that a few muscle synergies could account for the intertrial variability in automatic postural responses from different perturbation directions, as well as different postural strategies. Postural responses to multidirectional support-surface translations in 16 muscles of the lower back and leg were analyzed in nine healthy subjects. Six or fewer muscle synergies were required to reproduce the postural responses of each subject. The composition and temporal activation of several muscle synergies identified across all subjects were consistent with the previously identified “ankle” and “hip” strategies in human postural responses. Moreover, intertrial variability in muscle activation patterns was successfully reproduced by modulating the activity of the various muscle synergies. This suggests that trial-to-trial variations in the activation of individual muscles are correlated and, moreover, represent variations in the amplitude of descending neural commands that activate individual muscle synergies. Finally, composition and temporal activation of most of the muscle synergies were similar across subjects. These results suggest that muscle synergies represent a general neural strategy underlying muscle coordination in postural tasks.

The purpose of this study was to investigate muscle activation patterns during a landing task in boys and girls through the use of muscle synergies. Electromyographical data from six lower extremity muscles were collected from 11 boys and 16 girls while they performed single-leg drop-landings. Electromyographical data from six leg muscles were rectified, smoothed, and normalized to maximum dynamic muscle activity during landing. Data from 100 ms before to 100 ms after touchdown were submitted to factor analyses to extract muscle synergies along with the associated activation and weighing coefficients. Boys and girls both used three muscle synergies. The activation coefficients of these synergies captured muscle activity during the prelanding, touchdown, and postlanding phases of the single-leg drop-landing. Analysis of the weighing coefficients indicated that within the extracted muscle synergies the girls emphasized activation of the medial hamstring muscle during the prelanding and touchdown synergy whereas boys emphasized activation of the vastus medialis during the postlanding synergy. Although boys and girls use similar muscle synergies during single-leg drop-landings, they differed in which muscles were emphasized within these synergies. The observed differences in aspects related to the muscle synergies during landing may have implications with respect to knee injury risk.

1. The timing and intensity of phasic muscle activation were related to the distance of reaching movements of the human arm. We dissociated phasic components of muscle activation from complete muscle activation waveforms by subtracting waveforms obtained during very slow movements. 2. We recorded electromyographic (EMG) activity from elbow and/or shoulder muscles as standing subjects reached forward and upward to targets at four distances. Accuracy was deemphasized and no terminal corrections were allowed. In the first part of the experiment subjects were asked to move at their preferred speed. In the second part of the experiment they were asked to move using a range of speeds. 3. In the first part of the experiment subjects moved faster to more distant targets but they also increased movement time as a nearly linear function of target distance. The slope of this function was very similar across subjects. The phasic EMG waveforms for different distances appeared to be similar in shape but of variable duration. EMG time base was quantified using a correlation technique that identified the time base scale factor that best superimposed a given trace with a template. This technique revealed that the slope of the relation between EMG time base and target distance was not the same for all muscles. 4. In the second part of the experiment, where subjects moved to each target at a range of specified speeds, time base scaling was again significantly different for different muscles. The scaling differed most dramatically between anterior deltoid and medial head of triceps. 5. EMG intensity was more strongly related to movement time than to distance. We quantified the correspondence of distance and movement time to phasic EMG intensity using a multiple regression analysis of all distances and speeds, assuming a power relation. Distance exponents were positive and movement time exponents were larger and negative. This implies that movement time is more important than distance in its relation to EMG intensity.

This study compared the kinetics, barbell, and joint kinematics and muscle activation patterns between a one-repetition maximum (1-RM) Smith machine squat and isometric squats performed at 10 different heights from the lowest barbell height. The aim was to investigate if force output is lowest in the sticking region, indicating that this is a poor biomechanical region. Twelve resistance trained males (age: 22 ± 5 years, mass: 83.5 ± 39 kg, height: 1.81 ± 0.20 m) were tested. A repeated two-way analysis of variance showed that Force output decreased in the sticking region for the 1-RM trial, while for the isometric trials, force output was lowest between 0–15 cm from the lowest barbell height, data that support the sticking region is a poor biomechanical region. Almost all muscles showed higher activity at 1-RM compared with isometric attempts (p < 0.05). The quadriceps activity decreased, and the gluteus maximus and shank muscle activity increased with increasing height (p ≤ 0.024). Moreover, the vastus muscles decreased only for the 1-RM trial while remaining stable at the same positions in the isometric trials (p = 0.04), indicating that potentiation occurs. Our findings suggest that a co-contraction between the hip and knee extensors, together with potentiation from the vastus muscles during ascent, creates a poor biomechanical region for force output, and thereby the sticking region among recreationally resistance trained males during 1-RM Smith machine squats.
