Phylogenetic position of Pelusios williamsi and a critique of current GenBank procedures (Reptilia: Testudines: Pelomedusidae)

We re-examine the phylogenetic position of Pelusios williamsi by merging new sequences with an earlier published data set of all Pelusios species, except the possibly extinct P. seychellensis, and the nine previously identified lineages of the closely allied genus Pelomedusa (2054 bp mtDNA, 2025 bp nDNA). Furthermore, we include new sequences of Pelusios broadleyi, P. castanoides, P. gabonensis and P. marani. Individual and combined analyses of the mitochondrial and nuclear data sets indicate that P. williamsi is sister to P. castanoides, as predicted by morphology. This provides evidence for the misidentification of GenBank sequences allegedly representing P. williamsi. Such mislabelled GenBank sequences contribute to continued confusion, because only the original submitter can revise their identification; an impractical procedure impeding the rectification of obvious mistakes. We recommend implementing another option for revising taxonomic identifications, paralleling the century-old best practice of natural history museums for new determinations of specimens. Within P. broadleyi, P. gabonensis and P. marani, there is only shallow genetic divergence, while some phylogeographic structuring is present in the wide-ranging species P. castaneus and P. castanoides.

Abstract Researchers associated with natural history museums have made the collection of genetic resources a priority due to their importance in molecular studies, but often the long-term curation of these collections is difficult due to decentralized curation over multiple storage locations and lack of community best practice guidelines for their stewardship. Unlike traditional natural history specimens, the research utility of genetic samples increases with lower storage temperatures and fewer freeze–thaw events and, in addition, their use is consumptive. Collection managers must, therefore, maximize the research potential of each sample by carefully considering use on a case-by-case basis. This paper presents standardized guidelines accumulated for the management of genetic collections associated with natural history collections. These recommended practices are informed by general standards for biorepositories and augmented by information unique to natural history collections with the goal of providing a foundation for those curating genetic samples. Information pertains to all aspects of genetic sample curation and will assist those in making decisions regarding how to collect, store, track, process, and distribute genetic specimen samples. These guidelines also will allow users to make informed decisions regarding how to apply and improve the curation of their collection given their institution's goals and available resources.

AbstractComparative studies of the relative testes size in animals show that promiscuous species have relatively larger testes than monogamous species. Sperm competition favours the evolution of larger ejaculates in many animals – they give bigger testes. In the view, we presented data on relative testis mass for 17 Chinese species including 3 polyandrous species. We analyzed relative testis mass within the Chinese data set and combining those data with published data sets on Japanese and African frogs. We found that polyandrous foam nesting species have relatively large testes, suggesting that sperm competition was an important factor affecting the evolution of relative testes size. For 4 polyandrous species testes mass is positively correlated with intensity (males/mating) but not with risk (frequency of polyandrous matings) of sperm competition.

Objective:To ascertain the relationship between the germline NF1 gene mutation and glioma development in patients with neurofibromatosis type 1 (NF1).Methods:The relationship between the type and location of the germline NF1 mutation and the presence of a glioma was analyzed in 37 participants with NF1 from one institution (Washington University School of Medicine [WUSM]) with a clinical diagnosis of NF1. Odds ratios (ORs) were calculated using both unadjusted and weighted analyses of this data set in combination with 4 previously published data sets.Results:While no statistical significance was observed between the location and type of the NF1 mutation and glioma in the WUSM cohort, power calculations revealed that a sample size of 307 participants would be required to determine the predictive value of the position or type of the NF1 gene mutation. Combining our data set with 4 previously published data sets (n = 310), children with glioma were found to be more likely to harbor 5′-end gene mutations (OR = 2; p = 0.006). Moreover, while not clinically predictive due to insufficient sensitivity and specificity, this association with glioma was stronger for participants with 5′-end truncating (OR = 2.32; p = 0.005) or 5′-end nonsense (OR = 3.93; p = 0.005) mutations relative to those without glioma.Conclusions:Individuals with NF1 and glioma are more likely to harbor nonsense mutations in the 5′ end of the NF1 gene, suggesting that the NF1 mutation may be one predictive factor for glioma in this at-risk population.

Abstract The evolutionary relationship between the New World quail (Odontophoridae) and other groups of Galliformes has been an area of debate. In particular, the relationship between the New World quail and guineafowl (Numidinae) has been difficult to resolve. We analyzed >8 kb of DNA sequence data from 16 taxa that represent all major lineages of Galliformes to resolve the phylogenetic position of New World quail. A combined data set of eight nuclear loci and three mitochondrial regions analyzed with maximum parsimony, maximum likelihood, and Bayesian methods provide congruent and strong support for New World quail being basal members of a phasianid clade that excludes guineafowl. By contrast, the three mitochondrial regions exhibit modest incongruence with each other. This is reflected in the combined mitochondrial analyses that weakly support the Sibley-Ahlquist topology that placed the New World quail basal in relation to guineafowl and led to the placement of New World quail in its own family, sister to the Phasianidae. However, simulation-based topology tests using the mitochondrial data were unable to reject the topology suggested by our combined (mitochondrial and nuclear) data set. By contrast, similar tests using our most likely topology and our combined nuclear and mitochondrial data allow us to strongly reject the Sibley-Ahlquist topology and a topology based on morphological data that unites Old and New World quail. Natural history museums are unique spaces for interdisciplinary research and educational innovation. Through extensive exhibits and public programming and by hosting rich communities of amateurs, students, and researchers at all stages of their careers, they can provide a place-based window to focus on integration of science and discovery, as well as a locus for community engagement. At the same time, like a synthesis radio telescope, when joined together through emerging digital resources, the global community of museums (the ‘Global Museum’) is more than the sum of its parts, allowing insights and answers to diverse biological, environmental, and societal questions at the global scale, across eons of time, and spanning vast diversity across the Tree of Life. We argue that, whereas natural history collections and museums began with a focus on describing the diversity and peculiarities of species on Earth, they are now increasingly leveraged in new ways that significantly expand their impact and relevance. These new directions include the possibility to ask new, often interdisciplinary questions in basic and applied science, such as in biomimetic design, and by contributing to solutions to climate change, global health and food security challenges. As institutions, they have long been incubators for cutting-edge research in biology while simultaneously providing core infrastructure for research on present and future societal needs. Here we explore how the intersection between pressing issues in environmental and human health and rapid technological innovation have reinforced the relevance of museum collections. We do this by providing examples as food for thought for both the broader academic community and museum scientists on the evolving role of museums. We also identify challenges to the realization of the full potential of natural history collections and the Global Museum to science and society and discuss the critical need to grow these collections. We then focus on mapping and modelling of museum data (including place-based approaches and discovery), and explore the main projects, platforms and databases enabling this growth. Finally, we aim to improve relevant protocols for the long-term storage of specimens and tissues, ensuring proper connection with tomorrow’s technologies and hence further increasing the relevance of natural history museums.

Society in general is distant from scientific culture, it is required to bring scientific knowledge closer to the population. In this context, an effective and attractive way for scientific dissemination is the establishment of natural history museums, which are institutions of knowledge, displaying the past. Natural history museums have the natural world as their object of study; and their collections may contain the most diverse types of materials (local and/or from various parts of the world), such as zoological, botanical, geological, archaeological, among others. Scientific collections are the largest and most important source of authoritative biodiversity data, contributing to studies of biodiversity composition, evolutionary (morphological and genetic), biogeographical, phenological, as well as geological. The materials present in these collections may serve for temporal comparison, being useful to produce predictive models. Likewise, they have a fundamental role in safeguarding type specimens, i.e. the first organisms identified to describe and name a new species. In addition, there is the component available to visitors in general, in order to raise public awareness on the preservation of the local fauna and flora and of other places in the world. In this way, the museums serve both the academic-scientific public and visitors who come to these sites for recreational purposes. It is intended to promote, in Vila Nova de Cerveira, the Natural History Museum of the Iberian Peninsula - NatMIP (“Museu de História Natural da Península Ibérica”), which intends to collect materials for scientific purposes, mainly Iberian.

Abstract This paper will explore the significance of the expeditions under- taken by Wanda Hanke (1893-1958) in South America, of the networks she established in the region, as well as of her contributions to ethnological studies, in particular her compilation of extensive data and collections. Through Hanke's experience, it is possible to elucidate aspects of the history of ethnology and that of the history of museums in Brazil, as well as to emphasize the status of female participation in these areas. Wanda Hanke spent 25 years of her life studying the indigenous groups of Argentina, Bolivia, Brazil and Paraguay and collecting ethnological objects for natural history museums. Trained in medicine and philosophy, she began to dedicate herself to ethnological studies in her forties, and she travelled alone, an uncommon characteristic among female scientists in the 1940s, in Brazil.
