Steinernema kandii n. sp. (Rhabditida: Steinernematidae), a new entomopathogenic nematode from northern Benin

Summary Two nematode isolates from the genus Steinernema were collected in northern Benin. Morphological, morphometric, molecular and cross-hybridisation studies placed these nematodes into a new species, Steinernema kandii n. sp., within the bicornutum-group. Phylogenetic analyses based on both ITS and D2-D3 regions of 28S rDNA revealed that S. kandii n. sp. is different from all known Steinernema species and sister to S. abbasi (97.3-97.6% ITS nucleotide similarity) and S. bifurcatum (98.3-98.4% D2-D3 similarity). Steinernema kandii n. sp. can be separated from other members of the bicornutum-group by the greater infective juvenile (IJ) max. body diam. of 35 (27-48) μm (type isolate). It differs from S. abbasi by the greater IJ body length 707 (632-833) μm (type isolate), EP distance 55 (52-60) μm (type isolate), spicule length 67 (57-75) μm (type isolate) and the occurrence of one pair of genital papillae at the cloacal aperture.

Steinernema beitlechemi n. sp. is a new species of entomopathogenic nematode isolated from soil samples collected in Bethlehem, Free State province, South Africa. The infective juveniles of S. beitlechemi n. sp. are characterised by a body length of 696 (606-768) μm, position of nerve ring of 96 (86-105) μm from the head and lateral fields with six ridges (i.e., seven lines) in mid-body. First-generation males of S. beitlechemi n. sp. can be distinguished by a spicule length of 93 (88-100) μm and 12 pairs of genital papillae. First-generation females have a moderately protruding vulva and a rounded tail tip bearing one spine-like projection. The new species is further characterised by sequences of the ITS and partial 28S regions of the ribosomal DNA. Phylogenetic analyses show S. beitlechemi n. sp. as a member of a strongly supported monophyletic clade with three other African species: S. sacchari, S. cameroonense and S. nyetense.

Abstract Steinernema ashiuense sp. n. was collected by a Galleria baiting method from grassland along the riverbank of the Yura river, near the Ashiu Research Forest Station of Kyoto University. The new species is characterised by an infective juvenile body length of 768 (720-800) μm, lateral field with five equally developed ridges (i.e., six lines or incisures), head smooth lacking horn-like structures, excretory pore located at level of middle of pharynx, hyaline portion short (ca one-third of tail length). Males have 50-65 μm long, broad, slightly yellowish spicules; spicule length / spicule width = 4 (3.5-4.4), and two subventral and one subdorsal pair of genital papillae in the postcloacal region. The analysis of ITS-rDNA sequence placed S. ashiuense sp. n. in the 'feltiae-kraussei-oregonense' group in the clade containing S. robustispiculum and S. monticolum. It can be distinguished from these species by morphological characters of the infective juveniles and males.

Summary A new species of entomopathogenic nematode, Steinernema riojaense n. sp., was isolated during a survey in La Rioja province of Spain. Morphological and morphometric traits place the new species in the glaseri-group, which is characterised by having the largest infective juvenile (IJ) with an average body length in excess of 1000 μm. The IJ of S. riojaense n. sp. possess a body length of 1043 (869-1172) μm, the lateral fields are formed by eight equally spaced and developed ridges (nine incisures) in the mid-body region, giving the formula 2, 6, 8, 4, the excretory pore is located posterior to the mid-pharynx region (D% = 58), and the hyaline region occupies ca half of the tail length. The male spicules are moderately curved with a sharp tip and are golden-brown in colour with the manubrium elongate and having a length to width ratio of 1.7:1. The first-generation male tail is non-mucronate, whereas a small papilla-like mucron is developed in the second generation. There are 11 pairs of genital papillae plus a single precloacal papilla. The first-generation female possesses a moderately protruding vulva and a slightly protruding postanal swelling. The second-generation female possesses a slightly protruding vulva and a postanal swelling. The new species is further characterised by sequences of the internal transcribed spacer (ITS) and partial 28S regions (D2-D3) of the ribosomal DNA (rDNA). Phylogenetic analyses confirm that S. riojaense n. sp. belongs to the glaseri-group and forms a monophyletic subclade with other European species.

Summary A new species of entomopathogenic nematode, Steinernema borjomiense n. sp., was isolated from the body of the host insect, Oryctes nasicornis (Coleoptera: Scarabaeidae), in Georgia, in the territory of Borjomi-Kharagauli. Morphological characters indicate that the new species is closely related to species of the feltiae-group. The infective juveniles are characterised by the following morphological characters: body length of 879 (777-989) μm, distance between the head and excretory pore = 72 (62-80) μm, pharynx length = 132 (122-142) μm, tail length = 70 (60-80) μm, ratio a = 26.3 (23.0-29.3), H% = 45 (40-51), D% = 54 (47-59), E% = 102 (95-115), and lateral fields consisting of seven ridges (eight incisures) at mid-body. Steinernema borjomiense n. sp. was molecularly characterised by sequencing three ribosomal regions (the ITS, the D2-D3 expansion domains and the 18S rRNA gene) and the mitochondrial COI gene. Phylogenetic analyses revealed that S. borjomiense n. sp. differs from all other known species of Steinernema and is a member of the monticolum-group.

Abstract During a survey for entomopathogenic nematodes in citrus orchards throughout South Africa, a new species of Steinernema was isolated from a citrus orchard on Rietkloof farm, near the town of Piketberg in the Western Cape Province, South Africa. The nematode was isolated from soil using the Galleria-baiting technique. Steinernema citrae n. sp. is characterised by the following morphological characters: third-stage infective juvenile with a body length of 754 (623-849) μm, distance from head to excretory pore of 56 (49-64) μm, tail length of 71 (63-81) μm, and ratio E value of 110 (85-132). The lateral pattern for the new species is 2, 7, 8, 6, 4, 2 and is not typical for the genus. Steinernema citrae n. sp. is closely related the feltiae-group. The body length of the IJ is close to that of S. texanum and S. weiseri, though it differs in body diam., the length of the pharynx and E%. The male of S. citrae n. sp. differs from S. feltiae in the length and shape of the spicule and body diam. Steinernema citrae n. sp. differs from all species in the feltiae-group in the morphology of the vulva, as it has a single flapped, low, epiptygma. It also differs from the most closely related species, S. feltiae, as there is no interbreeding between the two species. In addition, the new nematode differs from other species of the feltiae-group by characteristics of the ITS and D2D3 regions of its rDNA.

During a survey of soil nematodes in Iran, a new species of Ditylenchus Filipjev, 1936 was discovered. Ditylenchus sarvarae sp. n. is characterised by its body length (1.0–1.4 mm), lateral field with seven incisures and without areolation, long postuterine sac (1.8–2.8 times the corresponding body diameter) and conical female tail with pointed tip (68–89 μm long, c = 13.7–18.2, c' = 4.2–5.1). Male specimens have conical tails (64–70 μm long, c = 14.5–16.7, c' = 4.0–4.5), spicules (22–26 μm long) and gubernaculum (8–10 μm long). Measurements and illustrations are provided for this new species. A molecular study of the 28S rDNA region of D. sarvarae sp. n. demonstrates that the Iranian species belongs to a separate group compared with the other molecularly characterized species of genus Ditylenchus.

AbstractA new species of Nippostrongylinae (Nematoda: Heligmonellidae), Stilestrongylus rolandoi n. sp., is described from specimens collected from the small intestine of the rodent Euryoryzomys russatus in the Atlantic Forest (Santo Amaro da Imperatriz, Santa Catarina state, southern Brazil). The genus Stilestrongylus includes 23 species, which parasitize rodents occurring in the Neotropical region. Stilestrongylus aureus (Durette-Desset & Sutton, 1985) from Argentina, S. azarai (Durette-Desset & Sutton, 1985) from Argentina, S. flavescens (Sutton & Durette-Desset, 1991) from Uruguay, S. franciscanus (Digiani & Durette-Desset, 2002) from Argentina, S. gracielae (Digiani & Durette-Desset, 2006) from Argentina, and S. oryzomysi (Sutton & Durette-Desset, 1991) from Argentina are closely related to Stilestrongylus rolandoi n. sp., all having caudal bursa patterns of types 1–4 in one of the lobes. Stilestrongylus rolandoi n. sp. is distinguished from the aforementioned species by its ray 6 being short in relation to rays 4 and 5, which are long and robust, and by having caudal bursa patterns of types 1–4 in both lobes. The new species has 27 ridges in the mid-body in males, and 24 in females, and has one of the highest ratios of spicule length to body length (21−33%) in this genus.

Rotylenchus castilloi n. sp., a new bisexual species is described and illustrated based on morphological, morphometric and molecular data. The new species is characterised by having a hemispherical, continuous lip region with an irregular corncob-like appearance under SEM, very long stylet (62–68 µm), vulva located at 49.7–62.2% of body length from anterior end, with a protruding double epiptygma, a rounded to convex-conoid (rarely bi-lobed) tail with 8–12 annuli and specific sequences of D2-D3 segments of 28S and ITS1-rRNA genes. Differences between the new species and four other species of the genus (R. mesorobustus, R. cazorlaensis, R. magnus and R. jaeni) are discussed. Morphologically, the new species can be separated from these species mostly by its body length, lip region characters, stylet length and location of phasmid. Phylogenetic analyses using 721 bp partial sequences of D2-D3 expansion segments of the 28S and 590 bp ITS1-rRNA genes revealed the new species forming a clade with two isolates of R. eximius and two isolates of R. unisexus, two morphologically unrelated species.

Abstract Steinernema hermaphroditum n. sp., a new entomopathogenic nematode isolated from soil samples in the Moluccan islands, Indonesia, is described. Morphological observations as well as biological evidence (cross-hybridisation studies) indicate the distinctness of S. hermaphroditum n. sp. from other Steinernema spp. This new species is characterised by the presence of hermaphrodites in the first adult generation. Key morphological diagnostic characters include: a digitate tail with a mucro and a glandular spermatheca filled with sperm in the first generation hermaphrodite; the value of D%; the morphology of the male spicules and gubernaculum and the number and arrangement of the genital papillae; the values of D%, E% and the pattern of the lateral field of the third-stage infective juvenile. Additionally, molecular evidence obtained from ITS rDNA RFLP profiles, 28S rDNA sequence analyses, and phylogenetic reconstruction provide further evidence to establish this nematode as a new species.

Steinernema nguyenin. sp. was recovered by baiting from beneath anOlea africanatree in South Africa. The combination of morphological and molecular features suggests thatS. nguyenin. sp. is a member of thefeltiae-kraussei-oregonensegroup, clustering with members of this group in Clade III. The new species is morphologically characterised by the infective juvenile body length of 737 (673-796) μm and the number of ridges in the infective juvenile lateral field is 2, 8, 2. The male of the first generation can be recognised by the spicule length of 66 (58-75) μm and a gubernaculum length of 43 (30-55) μm. The first generation female can be recognised by the vulval lips only slightly protruding and the presence of low, double-flapped epiptygmata. Analysis of the ITS and D2-D3 regions of the ribosomal DNA confirms thatS. nguyenin. sp. differs from all other knownSteinernemaspecies.
