Description of Bursaphelenchus microcarpae n. sp. isolated from Cryphalus abbreviatus emerged from Ficus microcarpa, with a report of B. carpini re-isolation

Summary Two Bursaphelenchus spp. belonging to the eggersi group (subgroup 3) were isolated from bark beetles (Cryphalus spp.). One species, isolated from Cryphalus sp. cf. carpini that emerged from a log of an unidentified broad-leaved tree (considered to be Carpinus sp.) collected in Sugadaira, Nagano, Japan, was identified molecularly and typologically as B. carpini. The other species, isolated from C. abbreviatus that emerged from a dead log of Ficus microcarpa collected in Okinawa, Japan, was differentiated molecularly from other eggersi group species. The new species is characterised by its three-lined lateral field, seven genital papillae with the posteriormost pair forming ‘glandpapillae’, short and stout male spicule with a strongly dorsally recurved and pointed condylus, small and rectangular male bursal flap with a variable terminus, female vulva with a ‘side flap’, broadly rounded female tail tip and molecular sequences of the ribosomal RNA region and partial mtCOI gene. The species is described and illustrated herein as B. microcarpae n. sp.

Summary Aphelenchoides roulingae n. sp. is described and illustrated. This new species was isolated from Xylocopa collaris sauteri collected at Fushan, Yilan county, Taiwan. Typologically, the new species is characterised by a three-lined lateral field, conical female tail with various and not pointed termini, male spicule with well-developed and slightly dorsally truncate condylus, blunt triangular-shaped rostrum and unevenly ventrally curved dorsal contour of the spicule blade (calomus-lamina complex). The combination of typological characters of the new species is unique. Aphelenchoides roulingae n. sp. shares the three-lined lateral field and female tail shape with ‘A. helicus’, which was previously transferred to Robustodorus from Aphelenchoides, but can be differentiated from that species by its longer and more slender female tail, narrower female tail tip, and longer female stylet. In addition, the spicule morphology of A. roulingae n. sp. is unique and not similar to that of any other Aphelenchoides species. Phylogenetically, the new species forms a well-supported clade with the other Xylocopa bee-associated species, A. xylocopae, but can be typologically and molecularly distinguished from the species. In addition to the taxonomic description, some typological characters of the genus Aphelenchoides are discussed.

AbstractBursaphelenchus parvispicularis n. sp. is described and figured. The new species was isolated from the bark of a dead oak tree, Quercus mongolica var. grosseserrata, and reared on Botrytis cinerea. Specimens were collected from a 2-week-old culture on Botrytis cinerea. The new species has a body length of 894 (711-1012) μm in the female and 764 (600-870) μm in the male, a ratio of 33.4 (29.7-37.1) in the female and 35.6 (31.4-45.3) in the male, and c and c′ ratios of 16.7 (14.8-19.0) and 4.4 (3.9-5.1), respectively, in the female and 24.3 (20.0-28.4) and 2.5 (2.2-2.8), respectively, in the male. The stylet is 14-16 μm long in the female and 13-15 μm in the male; there are three lines in the lateral field; the spicules are small (13 (13-15) μm long) and broad; there are seven (2 + 1 + 2 + 2) male caudal papillae; the 'bursa' varies from roundish trapezoid to rectangular with a concave terminus, and the female tail is tapered with clearly annulated dorsal surface near the tail tip and a rounded terminus. Based upon morphology the new species belongs to the B. hofmanni-group sensu Braasch. However, the new species is distinguished from the other B. hofmanni-group species by the size and form of the spicule, female tail shape, and morphometric values. Based on the molecular phylogenetic analysis of the DNA base sequences of ribosomal DNA and mtCOI gene, the new species was close to two B. hofmanni-group species (B. paracorneolus and B. hofmanni) and to B. hylobianum. This result is in accordance with their morphological similarity.

Abstract Two species of aphelench, Bursaphelenchus rufipennis n. sp. and Ektaphelenchus obtusus, were isolated from the 'nematangia', cocoon-like structures found at the base of the hind wings of Dendroctonus rufipennis. The nematangia contained adult females of E. obtusus and the dauer juveniles of B. rufipennis n. sp. Only B. rufipennis n. sp. could be cultured on Monilinia fructicola on LGPDA (lactic acid-treated, glycerol-supplemented, potato dextrose agar). The new species of Bursaphelenchus is described and figured and some additional morphological characters are ascribed to E. obtusus, E. josephi, E. sandiaensis, E. smaelus (= E. prolobos) and E. terebranus after examination of type and/or voucher specimens. Bursaphelenchus rufipennis n. sp. has an adult body length of ca 500-1000 μm, medium a ratios (ca 25-38 for females and ca 30-40 for males), b ratios of ca 8-13 (female) and 7-11 (male), c ratios of ca 15-22 (female and male), c′ ratios of ca 3-4 (female) and ca 2-3 (male), and is characterised by three incisures in the lateral field, mitten-shaped spicules and a conical female tail that curves ventrally and possesses a variable tail tip. The new species is morphologically closest to B. corneolus, B. curvicaudatus, B. gerberae, B. paracorneolus and B. talonus. Morphological examination of type and/or voucher specimens of five Ektaphelenchus species revealed coarse transverse body annulation and three pairs of male caudal papillae (except for the two species where males are not described). Clear typological differences were observed among these five Ektaphelenchus species in the structure of the lip region, presence/absence of stylet knobs and male spicule morphology. Although these characters have not been consistently documented in the past, they may be diagnostic for species in the genus. Molecular phylogenetic analysis based on SSU and D2/D3 LSU sequences revealed that B. rufipennis n. sp. was closest to B. paracorneolus and that E. obtusus was closest to species of Ektaphelenchoides and a Cryptaphelenchus sp.

During a survey of soil nematodes in Iran, a new species of Ditylenchus Filipjev, 1936 was discovered. Ditylenchus sarvarae sp. n. is characterised by its body length (1.0–1.4 mm), lateral field with seven incisures and without areolation, long postuterine sac (1.8–2.8 times the corresponding body diameter) and conical female tail with pointed tip (68–89 μm long, c = 13.7–18.2, c' = 4.2–5.1). Male specimens have conical tails (64–70 μm long, c = 14.5–16.7, c' = 4.0–4.5), spicules (22–26 μm long) and gubernaculum (8–10 μm long). Measurements and illustrations are provided for this new species. A molecular study of the 28S rDNA region of D. sarvarae sp. n. demonstrates that the Iranian species belongs to a separate group compared with the other molecularly characterized species of genus Ditylenchus.

Abstract Bursaphelenchus eproctatus n. sp. is described and figured from a dead Japanese black pine, Pinus thunbergii. The new species has a body length of 639 (580-714) μm in the female and 545 (550-595) μm in the male, ratio a = 35.9 (32.6-41.9) in the female and 33.8 (31.5-35.1) in the male, and c and c′ ratios = 14.2 (13.3-15.0) and 2.7 (2.3-3.0), respectively, in the male. A functional rectum and anus are absent, the intestine ending as a blind sac in the female posterior region. The female stylet = 18.8-21.8 μm long and the male stylet = 14.9-19.8 μm, the median bulb is elongate-oval and there are three lines in the lateral field. The spicules are of medium size, 19.8 (18.8-20.8) μm long, and arcuate with a terminal cucullus; there are two pairs of male caudal papillae and the female tail is tapered with a rounded terminus and a clearly annulated dorsal surface near the tail tip. Based upon spicule morphology the new species belongs to the B. piniperdae-group sensu Ryss and is close to B. hunanensis, B. minutus and B. lini. However, the new species is distinguished from these three species by a number of morphological features and morphometric values.

Summary Three parasitaphelenchid species, Parasitaphelenchus frontalis n. sp., P. costati n. sp. and Bursaphelenchus hirsutae n. sp., are described. Parasitaphelenchus frontalis n. sp. was isolated from the body cavity of Scolytus frontalis emerging from dead logs of Zelkova serrata collected from Kanagawa, Japan, and characterised by its four-lined lateral field in the adults and dome-shaped lip region of the parasitic juveniles. Whereas the two first-mentioned species were isolated from Alniphagus costatus, B. hirsutae n. sp. emerged from dead logs of Alnus hirsuta collected from Nagano, Japan. Parasitaphelenchus costati n. sp. and B. hirsutae n. sp. were recovered from the body cavity and the underside of the elytra of their host/carrier beetle, respectively. Parasitaphelenchus costati n. sp. is characterised by its three-lined lateral field in adults and the presence of a ventrally-directed hook on the lip region of the parasitic juvenile. Bursaphelenchus hirsutae n. sp. belongs to the eggersi group of the genus and is characterised by its three-lined lateral field, dorsally truncate condylus and pointed rostrum of males, and long, smoothly tapering and strongly ventrally recurved, female tail. Molecular characterisation is provided for all three new species and their phylogenys discussed.

A new species of the maerid amphipod, Elasmopus nkjaf, from Miyako Island, Ryukyu Islands, Japan, is named and described. Additionally, nucleotide sequences of nuclear 28S ribosomal RNA and histone H3 as well as mitochondrial cytochrome c oxidase subunit I and 16S ribosomal RNA from its holotype and paratype were determined. Elasmopus nkjaf sp. nov. belongs to the rapax-group, and distinguished from the other rapax-group species by long setae on ventral margin of epimeral plate 3, gland cone of peduncular article 2 of antenna 2 not reaching to end of peduncular article 3, propodus of gnathopod 1 facially setose, length of propodus of male gnathopod 2 shorter than 3 times as long as length of carpus, medial face of propodus of male gnathopod 2 shallowly hollowed, posterior margin of epimeral plate 3 without denticles. A key to the species of Elasmopus species occurring Japanese and adjacent waters is provided.

Abstract Bursaphelenchus chengi sp. n. is described and illustrated. Dauer juveniles were isolated from imported wood packaging materials from Taiwan to Nanjing Port, China. Bursaphelenchus chengi sp. n. was reared and maintained on Petri dish cultures of the fungus Botrytis cinerea. The new species is characterised by the medium body size in both sexes, the presence of only two incisures in the lateral field and the robust and strongly curved spicules. The spicule lamina is angular distally, the rostrum digitate and the condylus rounded. The tail is arcuate with a pointed terminus. The bursa is usually truncate with the posterior margin indented in some specimens or rounded with a fine axial point. Females have a small vulval flap formed by a short extension of the cuticle of the anterior lip, and a conical tail that gradually tapers to an almost straight or slightly recurved, pointed or rounded terminus. Because of the presence of two lateral lines, similar spicule shape, tapering female tail and the presence of a small vulval flap, B. chengi sp. n. should be grouped in the abietinus-group sensu Braasch. together with B. abietinus, B. antoniae, B. hellenicus, B. hylobianum and B. rainulfi. ITS-RFLP profiles support the proposal of the new species, and phylogenetic analysis of the 28S rDNA D2/D3 domain sequence places it close to B. antoniae and other species of the abietinus-group.

Summary Ditylenchus acantholimonis n. sp. is described based on morphological, morphometric and molecular characters. It was isolated from the rhizosphere soil of Acantholimon sp. in Golestan province, Iran, and is mainly characterised by having four lines in the lateral field, a pyriform to bottle-shaped offset pharyngeal bulb, post-vulval uterine sac 36.6-56.1% of the vulva to anus distance long, and a subcylindrical to conical tail with widely rounded tip. It is further characterised by short to medium-sized females, 480-617 μm long, with a fine stylet having small rounded knobs, V = 80.8-83.6, c = 11.0-13.8, c′ = 3.3-4.6, and males with 16.0-17.0 μm long spicules. The new species was morphologically compared with six species having four lines in their lateral field, rounded tail tip and comparable morphometric data namely: D. dipsacoideus, D. emus, D. exilis, D. paraparvus, D. sturhani, and D. solani. It was also compared with two species, D. ferepolitor and D. angustus, forming a maximally supported clade in the 18S tree. The phylogenetic analyses using the maximal number of Anguinidae and several Sphaerularioidea genera based upon partial 18S and 28S rDNA D2-D3 sequences revealed that Ditylenchus is polyphyletic. In the 18S tree, the new species formed a clade with D. ferepolitor (KJ636374) and D. angustus (AJ966483); in the 28S tree it formed a poorly supported clade with D. phyllobios (KT192618) and Ditylenchus sp. (MG865719).

Summary Laimaphelenchus spiflatus n. sp. isolated from declining Chinese pine, Pinus tabuliformis, is described and illustrated. The new species can be characterised by its relatively long body size of 1150 ± 108 (976-1437) μm for females and 1092 ± 78.6 (905-1235) μm for males, lateral field with four lines, females with a long vulval flap, and tail conoid, slightly ventrally curved and ending in a stalk having 8-12 projections when observed with SEM. Males are common, with 27.3 (23.4-28.8) μm long spicules having blunt (not well-developed) condylus and rostrum and truncate simple distal tip, and four caudal papillae. By having a vulval flap, four lateral lines and tail end with a stalk (without four tubercles), the new species comes close to L. preissii, L. simlaensis, and L. unituberculus, but can be differentiated from them by morphological characters and morphometric data. In phylogenetic analyses using near full length 18S and D2-D3 expansion segments of 28S rDNA, Laimaphelenchus spiflatus n. sp. appeared as an independent lineage separated from the other Laimaphelenchus spp. that are currently sequenced for their aforementioned genomic regions.
