Morphological characteristics and molecular data reveal two new species of Dentipellis from China

Two new species of Dentipellis are described from China based on morphological characteristics and molecular data. Dentipellis longiuscula is characterized by annual, resupinate hydnoid basidiomata with moderately long spines, rough basidiospores (5–6 × 3–3.6 µm) and absence of gloeoplerous hyphae and gloeocystidia. Dentipellis tropicalis is characterized by hydnoid basidiomata with numerous gloeocystidia presenting in hymenium, and minutely rough basidiospores (5–5.5 × 2.5–3.2 µm). Phylogenetic analysis based on the internal transcribed spacer (ITS) and nuclear large subunit ribosomal (nLSU) RNA gene regions confirmed the affinity of the new species in Dentipellis and indicated their relationships with other species in the genus.

During survey on rock-inhabiting fungi from karst habitats in Guizhou, China, two new species in Cyphellophora were discovered and identified. Phylogenetic analysis based on combined sequences of the nuclear large subunit (nucLSU) and internal transcribed spacer (ITS) region of ribosomal DNA revealed that the tested isolates, clustered into two clades that well affiliated in the genus of Cyphellophora. Morphological characteristics were also supported the estabolishment of the new species. Herewith Cyphellophora botryose sp. nov. and Cyphellophora guizhouensis sp. nov. were described and their differences from closely related species were discussed.

A new polypore, Ceriporia albomellea, collected from tropical China, is described and illustrated based on morphological characteristics and molecular evidence. It is characterized by thin, resupinate basidiome with a white subiculum, cottony margin, white to cinnamon-buff pores, clavate cystidia and oblong-ellipsoid basidiospores measured as 3.1–3.8 × 1.7–2 µm. Phylogenetic analysis based on the internal transcribed spacer (ITS) regions and nuclear large subunit (nLSU) ribosomal RNA gene regions supported C. albomellea as a distinctive species belonging to Ceriporia.

A new species of Polyporales, named as Skeletocutis yunnanensis, was collected on angiosperm wood in northern Yunnan Province, southwestern China. It is described based on morphological characteristics and molecular evidence. The species belongs to the Skeletocutis subincarnata complex, but differs morphologically from all known species of the genus by white, cream to buff pores surface, angular pores mostly 5–6 per mm with entire mouths, a dimitic hyphal structure both in trama and subiculum, generative hyphae in whole basidiocarps covered by fine crystals, skeletal hyphae unchanged in KOH, not agglutinated, allantoid basidiospores measured as 3.5–4.5 × 1.0–1.2 µm, and growth on angiosperm wood. Phylogenetic analysis based on the internal transcribed spacer (ITS) regions and nuclear large subunit (nLSU) ribosomal RNA gene regions indicated that the new species grouped with Skeletocutis and nested in the tyromyces clade.

Two fresh collections of Discosia were made from dead leaves of Fagus sylvatica in Italy. As these collections could not be cultured, the fruiting bodies were directly used for sequencing using a Forensic DNA Extraction Kit. Based on analyses of the concatenated internal transcribed spacer regions of the nrDNA operon (ITS) and large subunit rDNA (LSU) gene sequences, as well as morphological characters, the fresh collections are introduced as two new species, namely D. italica and D. fagi. Phylogenetically, these two species are distinct from all other Discosia species. Morphologically, D. italica is somewhat similar with D. fagi, but can be distinguished using dimension of conidiomata and conidiogenous cells. Descriptions and illustrations of the new taxa are provided herein.

Two new fungal species, Phaeosaccardinula dalbergiae and Phaeosaccardinula yunnanensis collected from Yunnan Province, China are described based on morphology and molecular data. They are compared with related taxa of Phaeosaccardinula. Detailed description, illustrations and notes are provided for each species. Molecular analysis based on the phylogenetic tree of combined internal transcribed spacer and large subunit nuclear ribosomal DNA sequences show that the two species form distinct clusters within the genus Phaeosaccardinula.

Gyroporus species with cyanescent oxidation reactions were investigated, based on morphology and phylogenetic analysis of DNA sequences from the nuclear ribosomal large subunit (nrLSU), the nuclear ribosomal internal transcribed spacer (ITS) and the mitochondrial adenosine triphosphate ATP synthase subunit 6 (atp6). Three species, including two new species, namely G. alpinus and G. flavocyanescens and one previously-described species, namely G. brunneofloccosus, are revealed from China. Collections formerly reported from China as “G. cyanescens” are either G. alpinus or G. flavocyanescens. The new species are documented and illustrated in detail, while the concept of G. brunneofloccosus is refined with additional recently-collected materials. Additionally, the cyanescent species G. pseudomicrosporus, previously described from China, is shown to be a member of the genus Gyrodon, based on re-examination of the type specimen. A key to the cyanescent Gyroporus species from China is provided.

Phoma odoratissimi sp. nov. on Viburnum odoratissimum and Syringa oblate, and Phoma segeticola sp. nov. on Cirsium segetum from China are introduced and described, employing a polyphasic approach characterising morphological characteristics, host association and phylogeny. Both species are the first records of Phoma species on their respective hosts. Multi-locus phylogenetic tree was inferred using combined sequences of the internal transcribed spacer regions 1 & 2 and 5.8S nrDNA (ITS), and partial large subunit 28S nrDNA region (LSU), β-tubulin (TUB) region and RNA polymerase II (RPB2) region. The two new species clustered in two separate and distinct lineages, and are distinct from their allied species.

Species of Clavaria (Clavariaceae, Agaricales) collected from China were studied using morphological and molecular methods. Two species, C. griseolilacina and C. sinensis were described as new species; both possess branched and pale purple-tinged fruit bodies. However, the former possess a few branched, while the latter is profusely branched. The phylogenetic analysis based on the internal transcribed spacer (ITS) sequences confirmed the distinctions between these two new species and their close relationships with C. rubicundula and C. fumosa in the genus. An evolutionary tendency in genus Clavaria was discussed. Additionally, morphological features of the two new species were described in detail and were illustrated with line drawings and photographs. Holotypes of the new species are deposited in the Mycological Herbarium of Hunan Normal University (MHHNU). The ITS sequences generated in this study were deposited in GenBank. A key is provided for the known Clavaria species in China.

A new species, Amanita glarea, is described and illustrated from Pakistan. Molecular phylogenetic data derived from internal transcribed spacer (ITS) and nuclear ribosomal large subunit (LSU) regions along with morphological characters make it distinct from other currently known Amanita species. Amanita glarea is characterized by a grayish brown, slightly umbonate, pileus having universal veil remnants and striate margins, a long ringless stipe with a saccate volva bearing yellowish brown patches on the verrucose surface, inamyloid globose to subglobose basidiospores, and a filamentous pileipellis with some inflated round to elongated cells. Phylogenetic analysis of nuclear ribosomal ITS and LSU nucleotide sequences resolve A. glarea with other taxa in Amanita section Vaginatae.

Inocybe griseorubida sp. nov. is described from Kerala State, India. A comprehensive description, photographs, and comparisons with phenetically similar species are provided. The nuclear ribosomal internal transcribed spacer region (ITS), a portion of the nuclear ribosomal large subunit (nLSU) and a portion of the nuclear second-largest subunit of RNA polymerase II (rpb2) gene of this species were sequenced and analyzed. Phylogenetic analysis of rpb2 sequences confirmed both the novelty of the species and its placement within the Pseudosperma clade.
