CHROMATOGRAPHY-MASS SPECTROMETRY OF TOLUENE EXTRACT OF YARROW (ACHILLED MILLEFOLIUM L., ASTERACEAE FAMILY) (the message II)

The aim of the study is to study in detail the chemical composition of organic matter of common yarrow in order to establish the main biochemical processes responsible for the formation of the composition of the latter. Materials and methods of research. The chemical composition of toluene extract of yarrow was studied by chromatography-mass spectrometry under the following conditions. GC-2010 gas chromatograph connected to a GCMs-TQ-8030 triple quadrupole mass spectrometer running GCMS Solution 4.11 software. The results of studying the chemical composition of toluene extract-a product of sequential exhaustive extraction of yarrow by chromatography-mass spectrometry, which allowed identifying 129 individual compounds for which the quantitative content was determined, mass spectra and structural formulas were obtained, and the structure-group composition of the extract was calculated. Results and discussion. The basis of the extract is determined by hydrocarbons, esters, sterols and carboxylic acids, which account for: 43.59; 15.47; 15.33 and 7.59 (wt.% of the extract), respectively, the content of ketones, alcohols, aldehydes and organosilicon compounds-3.77; 2.77; 1.68 and 6.49 (wt.% of the extract). The presence of phenols and glycosides has not been established; furan and PYRAN fragments are part of the structures of individual alcohols and ketones. Based on the features of the chemical composition, it can be argued that the pharmacological effect of toluene extract of yarrow is determined precisely by the content of hydrocarbons, with the dominance of alkenes, alkynes, arenes and cycloalkanes; sterols of the following types: Betulin, Lupeol, γ-Sitosterol and Sitostenon, Campesterol, 24-Noroleana-3.12-dien; carboxylic acids containing up to three double and triple bonds in the hydrocarbon chain, as well as esters mainly formed by Oxalic and Benzeneacetic acid. Undoubtedly, a certain contribution to the direction of the pharmacological action of this extract is made by organosilicon compounds, the proportion of which is 6.49 (wt.% of the extract).

The purpose of the study is to detail the information about the peculiarities of the chemical composition and structural organization of different groups of compounds of organic matter, yarrow, revealing the influence of nature of the extractant on the qualitative composition and quantitative content of extractable compounds in the obtained chloroform extract, and identifying new pharmacological actions of the latter, the extension of the range of compounds already known in the literature on herbal medicine. Materials and methods of research. The chemical composition of the chloroform extract was studied by chromatography-mass spectrometry under the following conditions. The chemical composition of the ethanol extract of yarrow was used by chromatography-mass spectrometry under the following conditions. GC-2010 gas chromatograph connected to a triple quadrupole mass spectrometer GCMS-TQ-8030 running GCMS Solution 4.11 software. Identification and quantitative determination of the content of compounds were performed under the following chromatography conditions: sample input with flow division (1:10), ZB-5MS column (30m × 0.25 mm × 0.25 mm), injector temperature 280 °C, carrier gas-helium, gas velocity through the column 29 ml/min. Conclusion. For the first time, the chemical composition of chloroform extract, a product of sequential exhaustive extraction of yarrow by chromatography —  mass spectrometry, was studied, which allows identifying 94 individual compounds for which mass spectra and structural formulas were obtained, the quantitative content was determined, and the structural and group composition of the extract was calculated. The basis of the composition of the chloroform extract is determined by hydrocarbons enriched with arenes, alkenes and alkynes, esters formed mainly by oxalic and benzoic acids, polyunsaturated fatty carboxylic acids, steroid compounds, alcohols, nitrogen-containing structures that include individual fragments of natural alkaloids, as well as a variety of organosilicon compounds that determine the main directions of pharmacological action of various yarrow preparations, including chloroform extract.

The aim of the study was to perform column adsorption liquid chromatography in order to obtain ethanol eluate, after preliminary sequential elution of ethanol extract with solvents in the order of their polarity: n–hexane, toluene, chloroform, acetone; to study in detail the chemical composition of this eluate by chromatography-mass spectrometry and X-ray fluorescence spectroscopy, with the intensification of compounds determining the composition of eluate, obtaining their masses -spectra, structural formulas, determination of the nature of trace elements. Materials and methods of research. A detailed study of the chemical composition of acetone eluate of ethanol extract of walnuts+ leaves was carried out using column adsorption liquid chromatography, chromato-mass spectrometry and X-ray fluorescence analysis. The results and their discussion. The features of the chemical composition of ethanol eluate of the product of column adsorption liquid chromatography of ethanol extract were studied, the qualitative composition and quantitative content of compounds identified in the first one were established by chromatography-mass spectrometry. Mass spectra and structural formulas were obtained for 31 individual eluate compounds. Ethanol eluate is characterized by the following indicators of structural and group composition (mass. % of eluate): esters of phthalic acid - 52.14, silicon and sulfur-containing compounds, respectively - 20.19 and 6.43, hydrocarbons - 15.38, glycosides - 2.21, alcohols - 1.92, carboxylic acids - 1.07, sterols - 0.89. It is assumed that the pharmacological effect of ethanol eluate is mainly determined by a set of free carboxylic acids, as well as phthalic acid, formed as a result of biochemical and hydrological transesterification of its esters containing compounds, alcohols, glycosides and sterols. Organosilicon compounds during hydrolysis give silica in the size of nanoparticles, which play the role of carriers of the above compounds to various organs of a living organism, as well as drainage of blood vessels from the products of cell metabolism, for example, cholesterol, various fats, etc. A comparative characteristic of the chemical composition of all ethanol extract eluates was carried out, the effectiveness of column adsorption liquid chromatography was shown.

Conifers are well represented in Vietnam where a new pine species has been recently discovered in Son La province: Pinus cernua, synonym P. armandii ssp. xuannhaensis. The compositions of needle, cone, and branch oils have been investigated by gas chromatography (retention index), gas chromatography-mass spectrometry, and 13C nuclear magnetic resonance. Myrcene (47.0%) was the main component of needle oil, followed by β-pinene (28.4%) and α-pinene (12.5%). Branch oil also contained myrcene (32.8%), α-pinene (17.9%), β-pinene (9.8%), and a high content of limonene (20.0%). Finally, cone oil displayed α-pinene (44.1%) beside myrcene (11.5%), β-pinene (8.1%), and limonene (5.8%).
