The present study was carried out to evaluate the genetic polymorphism in Ovine MTNR1A of two tropical breeds ie., highly prolific NARI Suwarna sheep and Mandya sheep from southern peninsular region of India with respect to differential fertility. Fifty animals from each breed were randomly selected and subjected to RsaI-RFLP analysis of exon-2 of Ovine MTNR1A. The study revealed monomorphic patterns in all the animals for both the breeds. PCR amplicon of 824 bp fragments with four cut sites at 53 bp, 320 bp, 343 bp and 754 bp, resulted in 411 bp, 267 bp, 70 bp, 53 bp and 23 bp. Only presence of CC genotypes at rs406779174 SNP of Ovine MTNR1A in both the breeds supports non-seasonality in breeding of these animals and no association of SNP with increased fecundity of NARI Suwarna sheep over Mandya sheep.

Abstract. This study was carried out to detect polymorphisms in the melatonin receptor 1A (MTNR1A) and arylalkylamine N-acetyltransferase (AA-NAT) genes and their association with reproductive traits. Blood samples of 126 animals from three Egyptian sheep breeds were collected. DNA was extracted and subjected to PCR restriction fragment length polymorphism (RFLP) analysis using the RsaI and SmaI enzymes. Two alleles (C and T) and three genotypes (CC, CT and TT) for MTNR1A and for AA-NAT (A and G; GG, GA and AA) were detected. The alleles C and A and the genotypes CT and GA showed the highest frequencies for the MTNR1A and AA-NAT genes, respectively. Association analysis of the MTNR1A single nucleotide polymorphism (SNP) with ewe reproductive traits revealed significant associations in the Ossimi and Rahmani breeds with age at first lambing, and the C allele seemed to be the favorable allele. The results for the AA-NAT SNP demonstrated significant correlations in Ossimi with age at first lambing and litter size and in Rahmani with lambing interval; the G allele seemed to be the desirable allele. In the first conception season, ewes carrying CT exhibited a significantly lower age of first lambing in the unfavorable season. Additionally, GG ewes exhibited a significantly lower age of first lambing in the early favorable season, followed by the unfavorable season. To the best of our knowledge, this is the first study of these associations in Egyptian sheep breeds. In conclusion, the polymorphisms revealed in this study could be used as genetic markers to improve reproductive efficiency during the unfavorable season, and the obtained desirable genotypes could be considered in new genetic selection schemes.

Alcaligenes sp. JG3 is a local strain bacterium from Indonesia, isolated from cultivated corn field of Central Java. This bacterium is able to produce lipase with fairly high activity. In order to do lipase gene sequence characterization, two sets of primer pair were used in this study (primer Fjg3 5’- ATGACCGAGCTGACTGTAG-3’, Rjg3 5’-TCAGGAGGGGTAAATCCAC-3’ and internal primer Fi 5’-TGACCCATGACCAGGCGGAA-3’ and Ri 5’-TTCGCCTGGTCATGGGTCA-3’). The complete lipase JG3 gene sequence consists of 1081 bp from start codon ATG to the stop codon of TGA. Lipase JG3 had high similarity to another lipase from genus Alcaligenaceae which was up to 90%. However, the 3D protein visualization analysis indicated that this lipase JG3 also has the characteristic of ABC transporter protein.

Abstract. The objective of the present study was to determine the polymorphism in the prolactine receptor (PRLR) gene in Chios, White Karaman and Awassi, which are native sheep breeds in Turkey. By means of PRLR gene sequence homology between sheep and humans, two primer pairs were designed for polymerase chain reaction (PCR) amplification within intron 1 and exon 10 of the PRLR gene in sheep. A total of 160 amplicons (99 for intron 1 and 61 for exon 10) were subjected to DNA sequence analysis. For intron 1, 6 different haplotypes were determined. For exon 10, 7 different haplotypes were obtained. Some variations determined for exon 10 (g.14A>T p.Q14L; g.160G>A p.D160N; g.166G>A p.E166K; g.167A>T p.E167V; g.176A>T p.H176L; g.206G>A p.S206N; g.208G>A p.G208R) led to changes in the amino acids, but no amino acid changes were determined in g.2A>T, g.81A>G, g.138A>G, g.186C>T, g.207T>C. It was noted in particular that White Karaman and Awassi were similar to each other in both PRLR exon 10 and intron 1 haplotypes, whereas the Chios breed had a different variation.
