Occurrences and characterization of endophytic Gluconacetobacter isolated from sugarcane tissues of North Eastern region of India

Sugarcane plants including roots were collected from 21 sugarcane growing locations of Golaghat, Dimapur and Jorhat districts of North Eastern region for isolation of endophytic Gluconacetobacter. Based on the nitrogenase activity, only eight isolates were selected for further characterization of Gluconacetobacter. The nitrogenase activity of the isolates ranged in between 12.4 to 72.4 n mole C2H4 h-1 culture-1. The characteristic yellowish surface pellicles were formed on inoculating the selected isolates into semisolid LGI medium and acetic LGI medium. On enumeration in LGI plates, the frequencies of well recognized Gluconacetobacter like colonies ranged in between 0.75 and 7.68 x 105 (cfu g-1 fresh tissue). Colony morphology of the isolates was irregular flat, irregular raised or circular raised yellow colonies of 1-5 mm in diameter. Light brown to brown pigmentation was observed on potato agar plates and light brown pigmentation with clearing of plates was observed on GYC plates. All the isolates exhibited gram negative and shape of the cells were rods. Reactions to extracellular and intracellular enzymes as well as to the various carbon sources by the isolates explained the diversity among them. The IAR profiles of the eight isolates against twelve antibiotics exhibited wide variation in their response to various antibiotics.

Nitrogenase activity was analysed after supplementation of various carbon and nitrogen sources in the growth medium by Acetylene Reduction Assay methods in selected 9 isolates from 74 diazotrophic isolates.  Enhancement in nitrogenase activity was recorded many fold by the addition of different organic carbon sources in which maltose and pyruvate showed better result than others. In case of supplementation of nitrogen sources, reduction of nitrogenase activity was observed.  Nitrogenase activity increased from 22.7 to 72.7% in various strains when they are tested in anaerobic condition, Amplification of fragment of 390 bp showed that nitrogenase activity due to presence of nifH gene.  Sequences were submitted to NCBI GeneBank and the accession number of nifH sequence of ECI-10A (FJ032023), AF-4B (FJ032024), AF-4C (FJ032022) and BN-2A (FJ032021) has been obtained. Nitrogenase activity has been found associated with roots of sweet potato (Ipomoea batatas (L.) Lam). The objective of this investigation was to characterize N2-fixing isolates of the genus Azospirillum found in association with roots of different sweet potato cultivars. Five cultivars ('Carver', 'Jewel', 'Rojo Blanco', 'Centennial', 'Southern Queen') were grown in greenhouse pots on Norfolk sandy loam at pH 6.5 for periods of 1 to 3 months. Enrichment cultures of fibrous and storage root samples were grown on N-free semisolid malate medium at 35 °C. Sixteen bacterial isolates with taxonomic characteristics similar to those of Azospirillum were isolated from the five cultivars. Nitrogenase activities of single colonies grown in semisolid media for 30 h ranged from 32 to 472 nmol C2 H4∙culture−1∙h−1. The isolates were characterized by their acidification of carbon sources, ability to reduce nitrate, and morphology.

Carbon plays a crucial role in determining the ecosystem balance and slight changes in its concentration in the atmosphere can have significant impacts. The launch of JAXA’s GOSAT (Greenhouse gases Observing SATellite) in 2009 has started a new era of high accuracy CO2 concentration and flux measurements from space borne sensors. This paper reports the spatial and temporal variability of terrestrial biospheric carbon fluxes over the agro-climatic zones of India derived using GOSAT data for the period June 2009 to October 2011. The country averaged biospheric carbon flux varied from &minus;0.47 (October) to 0.37 (April) gC m<sup>&minus;2</sup> day<sup>&minus;1</sup>. Maximum variability in fluxes was observed for the North-Eastern region (&minus;2.18 to +1.38 gC m<sup>&minus;2</sup> day<sup>&minus;1</sup>) whereas the dry region of Rajasthan showed extremely low values (&minus;0.1 to +0.1 gC m<sup>&minus;2</sup> day<sup>&minus;1</sup>). The temporal variation in flux values was compared to averaged NDVI for each zone and indicated that growing season corresponds to more sequestration of carbon from the atmosphere. We compared GOSAT derived biospheric flux with Carbon Tracker (CT) data and observed that the two values show good agreement for all months except June and July. This study provides new estimates of biospheric carbon flux using satellite data driven models to better understand the carbon dynamics associated with terrestrial biosphere over India.

Aim. To validate the suitability of commercial API 20E test-system (bioMerieux) for the identifi cation and characterization of facultative gram-negative phytopathogenic bacterial isolates. Methods. Conventional mi- crobiological methods, API 20E test-system (bioMerieux) according to the manufacturer’s instructions. Re- sults. The identifi cation results for Erwinia amylovora, Pectobacterium carotovorum and Pantoea agglome- rans isolates were derived from the conventional and API 20E test systems, which, were in line with the literature data for these species. The API 20E test-system showed high suitability for P. agglomerans isolates identifi cation. Although not all the species of facultatively anaerobic phytopathogenic bacteria may be identi- fi ed using API 20E test-system, its application will surely allow obtaining reliable data about their physiologi- cal and biochemical properties, valuable for identifi cation of bacteria, in the course of 24 h. Conclusions. The results of tests, obtained for investigated species while using API 20E test-system, and those of conventional microbiological methods coincided. The application of API 20E test-system (bioMerieux) ensures fast obtain- ing of important data, which may be used to identify phytopathogenic bacteria of Erwinia, Pectobacterium, Pantoea genera.
