AbstractGentiana dinarica Beck, rare and endangered species of Balkan Dinaric alps, was in vitro propagated (micropropagated) from axillary buds of plants collected at Mt. Tara, Serbia. G. dinarica preferred MS to WPM medium, with optimal shoot multiplication on MS medium with 3% sucrose, 1.0 mg l−1 BA and 0.1 mg l−1 NAA. Rooting was not clearly separated from shoot multiplication since BA did not completely inhibit root initiation. Spontaneous rooting on plant growth regulator-free medium occurred in some 30% of shoot explants. Rooting was stimulated mostly by decreased mineral salt nutrition and a medium with 0.5 MS salts, 2% sucrose and 0.5–1.0 mg l−1 IBA was considered to be optimal for rooting. Rooted plantlets were successfully acclimated and further cultured in peat-based substrate.

An experiment was conducted at the Plant Biotechnology Laboratory, Department of Horticulture, Patuakhali Science and Technology University during the period from August 2016 to April 2017 to standardize the protocol for in vitro propagation of banana. The experiment was laid out in completely randomized design with four replications. Three to four months aged field grown rhizome attached shoots were used as explants and cultured on MS medium with different concentrations and combinations of BAP (6-benzylamino purine), BAP + KIN (Kinetin) + NAA (Naphthalene Acetic Acid) and IBA (Indole-3- Butyric Acid) + IAA (Indole-3- Acetic Acid) to observe their efficacy on single shoot initiation, shoot multiplication and root formation respectively. Minimum number of days required for shoot initiation (9.07) with highest shoot initiation percentage (91.14) and the longest shoot (2.23 cm) was found in MS medium supplemented with 5.0 mg/L BAP. On the other hand, highest shoot multiplication percentage (80.99) with maximum number of shoots per explant (4.47), the highest length of shoots (4.17 cm) and maximum number of leaves (4.04)was observed in MS medium supplemented with 4.0 mg/L BAP + 2.0 mg/L KIN + 2.0 mg/L NAA. In case of root regeneration, the best results on days required for root initiation (9.00), the highest root initiation percentage (85.05), maximum number of roots per plantlet (5.83) and the highest length of roots (4.17 cm) was obtained in MS medium supplemented with1.5 mg/L IBA + 0.5 mg/L IAA. This study investigated in vitro production of diploid (AS2) and tetraploid (AS4) cytotypes of snowdrop anemone. The effect of plant growth regulators (PGRs) on in vitro shoot multiplication and rooting was investigated. The effect of activated charcoal (AC) on root induction was also studied. Ploidy level affected growth characteristics during multiplication and rooting. Shoot induction in AS4 was higher on medium supplemented with cytokinin (3.2–3.6), while the AS2 clone formed the most shoots on PGR-free medium (3.6). The highest rooting percentage was achieved on PGR-free medium in both genotypes (AS2 clone, 100% and AS4 clone, 93.3%). The addition of AC to the PGR media largely increased root induction and root length. Rooted plantlets were successfully acclimatised in the greenhouse with 100% survival. Thus, the described micropropagation protocol represents a rapid and effective in vitro propagation method for utilisation in horticulture and conservation programmes of snowdrop anemone.

The purpose of our research was to establish a protocol for the in vitro culture of Viburnum treleasei, a rare and endangered taxon with high ornamental potential endemic to the Azores islands. The surface sterilization of the explants was better achieved with a pretreatment of 0.1% (w/v) Benomyl for 2 h followed by 0.2% (w/v) HgCl2 for 10 min with agitation. Shoot tips were the most efficient explants for shoot development and single-node segments for proliferation. Woody plant medium (WPM) was adequate for all micropropagation stages. For culture establishment and shoot development, a hormone-free medium was adequate, whereas a 1.1 μM N6-benzyl adenine medium supplement was more efficient for shoot multiplication. Elongation and rooting could be carried out on a 1.3 μM 1-naphthaleneacetic acid-supplemented medium. Acclimatization of in vitro-produced plantlets was achieved after 1 month with a success rate of 50%. This in vitro propagation procedure will be useful for the conservation of Viburnum treleasei through production of morphologically true-to-type plants, allowing the recovery of depleted natural populations. The present experiment was conducted to optimize protocols for in vitro propagation of banana (Musa sp.) cv. ‘Patakpura’ (AAB), supplemented with different growth regulators. Shoot tips obtained from sword suckers were cultured aseptically on MS medium supplemented with different concentrations of cytokinins like 6-Benzylaminopurine (BAP) and Kinetin (KN) for multiplication of shootsand auxins such as indole acetic acid (IAA) and naphthalene acetic acid (NAA) for induction of roots. The best result from the initial culture was obtained from MS medium supplimented with 4 mg/l BAP + 0.5 mg/l IAA. The highest shoot fresh weight, shoot length and number of shoots per explant were recorded from MS medium supplemented with 4 mg/l BAP + 0.5 mg/l IAA + 0.25 mg/l NAA. Therefore, the MS medium supplemented with 4 mg/l BAP + 0.5 mg/l IAA + 0.25 mg/l NAA was found to be most effective and productive combination for shoot multiplication and proliferation of the culture in vitro. Shoot explants from actively growing, greenhouse-maintained plants of Baptisia `Purple Smoke' were cultured in vitro for shoot initiation on Murashige and Skoog (MS) basal medium containing vitamins and supplemented with 30 g·L–1 sucrose, 8.87 μm BA, and 4.14 μm K-IBA. All subsequent media were supplemented with 2.47 mm NaH2PO4 to enhance shoot growth. Single-node explants were subcultured for shoot multiplication on MS medium with either no plant growth regulator or with 2.22, 4.44, 8.87, 17.74, or 35.48 mm BA in combination with 0.0 or 4.14 μm K-IBA. Explants produced a maximum of 4.1 shoots on the medium with 2.22 μm BA. Shoots rooted on all concentrations of K-IBA (2.07, 4.14, 10.36, or 20.72 μm) and K-NAA (2.23, 4.46, 11.15, or 22.29 μm) tested. Maximum rooting was 100% on MS medium with 11.15 μm K-NAA; however, this treatment induced copious stem callusing. Rooted shoots were greenhouse-acclimatized for 2.5 weeks. Overall survival was 86%. For optimal rooting and subsequent acclimatization, treatment with 2.23 μm K-NAA is recommended; this resulted in 83% rooting and 87% acclimatization. Chemical names used: N6 benzyladenine (BA); potassium salt of indole-3-butyric acid (K-IBA); potassium salt of 1-naphthalene acetic acid (K-NAA).

An experiment was conducted for standardization of in vitro propagation technique of Bacopa monnieri (L.), a medicinal herb of India. Healthy leaf segments of the herb were used as explants with basic Murashige and Skoog (MS) medium containing various combinations of different growth regulators for callus, shoot and root initiation. The best callus induction percentage (95.47%) was observed on MS + 0.5 mg/L NAA and 2.0 mg/L BAP (T3). The maximum number of shoots (8), shoot length (9.30 cm) and shoot induction percentage (90.48%) was achieved on MS + 3.0 mg/L BAP and 1.0 mg/L Kn (ST4). The maximum number of roots (8) and root length (7) was observed on MS + 1.5 mg/L IAA (RT5). The rooted micro shoots were successfully hardened and acclimatized in green house and subsequently established in soil with survival rate of 90%.

Gerbera plant Gerbera jamesonii is classified according to the flower colors to four strains: white, yellow, pink and purple. Capitulum and scape explants were tested on MS medium in half or full salts strength, supplemented with different combinations of plant growth regulators cytokinins kintin (Kin) and benzel adinine (BA), auxin indolacitic acid (IAA). Results revealed that the capitulum showed better response to shoot formation 64.13% whereas the scape did not show response. Yellow flowers showed higher response in shoot formation 37.5% than other strains. growth regulators combination BA and IAA (3.0 + 0.1) mg/L respectively showed better response for shoot multiplication. Auxin IBA (0.5) mg/ L gave better rooting percentage 60% than other auxins IAA and NAA all concentrations. The acclimatization of the gerbera was 78.59%.

Seeds of four tomato hybrids Ginan, Nirtia, Speedy and Rola were cultured on aseptic germination medium after surface sterilization with NaOCl. Shoot tips 1cm length were dissected and grown on MS medium supplemented with Benzyl adenine (BA) and Indole acetic acid (IAA) for multiplication. Callus was induced on hypocotyls cultured on MS medium supplemented with BA under light intensity of 1000 lux and 25 0C. Results showed that NaOCl at 4% completely disinfected seeds and resulted in pathogen free seedlings. Results also showed that a concentration of 4 mg/l of BA was superior in increasing callus fresh and dry weights. Shoot multiplication was achieved on the same medium supplemented with 2 and 0.8 mg/l of BA and IAA respectively.

Micropropagation (shoot tip and nodal cuttings culture) was used for the rapid propagation of the non-invasive, decorative, native plants of maiden pink (Dianthus deltoides L.) in order to preserve their genetic diversity. In vitro culture was successfully established on Murashige and Skoog medium (MS) using seeds as the initial material. In the shoot multiplication phase, the explants were cultured on MS medium supplemented with different concentrations of 6-benzylaminopurine (BAP) and naphthaleneacetic acid (NAA). The highest multiplication rate was achieved on a medium containing 0.1 mgL-1 of BAP and 0.1 mgL-1 of NAA. The rooting was successful on a hormone-free medium (100%), and the highest percentage of microplant acclimatization (97%) was recorded in a 4: 1 mixture of peat and sand.
