Abstract Background: SYP71, the plant-specific Qc-SNARE protein, is reported to regulate vesicle trafficking. SYP71 is localized on the ER, endosome, plasma membrane and cell plate, suggesting its multiple functions. Lotus SYP71 is essential for symbiotic nitrogen fixation in nodules. AtSYP71, GmSYP71 and OsSYP71 are implicated in plant resistance to pathogenesis. To date, SYP71 regulatory role on plant development remain unclear.Results: AtSYP71-knockout mutant atsyp71-4 was lethal at early development stage. Early development of AtSYP71-knockdown mutant atsyp71-2 was delayed, and stress response was also affected. Confocal images revealed that protein secretion was blocked in atsyp71-2. Transcriptomic analysis indicated that metabolism, response to environmental stimuli pathways and apoplast components were influenced in atsyp71-2. Moreover, the contents of lignin, cellulose and flavonoids as well as cell wall structures were also altered.Conclusion: Our findings suggested that AtSYP71 is essential for plant development. AtSYP71 probably regulates plant development, metabolism and environmental adaptation by affecting cell wall homeostasis via mediating secretion of materials and regulators required for cell wall biosynthesis and dynamics.

Cynara cardunculus L. is a species native to the Mediterranean basin that comprises 2 crops, globe artichoke (var. scolymus L.) and cultivated cardoon (var. altilis DC), as well as wild cardoon (var. sylvestris (Lamk) Fiori). Globe artichoke represents an important component of the South European agricultural economy but is also cultivated in North Africa, the Near East, South America, the United States, and China. Breeding activities and molecular marker studies have been, to date, extremely limited. Better knowledge of the genome of the species might be gained by developing a range of molecular markers. Here, we report on the development of 14 microsatellites (simple sequence repeats (SSRs)) through a novel approach that we have defined as the microsatellite amplified library (MAL). The approach represents a combination of amplified fragment length polymorphism and a primer extension based enriched library, is rapid, and requires no hybridization enrichment steps. The technique provided a ~40-fold increase in the efficiency of SSR identification compared with conventional library procedures. The developed SSRs were applied for genotyping 36 accessions of C. cardunculus, including a core of 27 varietal types of globe artichoke, 3 accessions of cultivated cardoon, and 6 Sicilian accessions of wild cardoon. Principal coordinates analysis made it possible to differentiate both cultivated and wild forms from each other.Key words: globe artichoke, wild and cultivated cardoon, molecular markers, AFLP, MAL (microsatellite amplified library).

Abstract In contrast to ripening of climacteric fruits knowledge, non-climacteric fruits ripening still poorly understood. In this sense, strawberry (Fragaria x ananassa L. Duch) is a high commercial crop and has an expressive world production that has been used as a model study. Thus, the aim of this study was to evaluate the effect on physicochemical and phytochemical characteristics of strawberry Cv. Camiño Real, in four maturation stages (green, white, pink and red) after the ethylene (Etephon®) and brassinosteroid (epi-brassinolide) exogenous spray. The evaluations were carried out at application time (field assay) and after 24h (post-harvest assay). Brassinosteroid application tends to reduce phenolic compounds content mainly in the red stage, whereas ethylene treatment tends to increase supposing that while brassinosteroids reduce phenolic compounds acting to fruit conservation, by the reduction of oxidative stress, increasing antioxidant activity, the ethylene act in fruit ripening leading to senescence. However, for vitamin C content while brassinosteroid treatment increases ascorbic acid in postharvest for the white fruit, the ethylene treatment do a dramatically increased in vitamin C content to red fruits, showing that although ethylene and brassinosteroids can affect the vitamin C content, the response can change according to the development stage spray, suggesting that more studies are necessary to understand the hormonal interaction for ascorbic acid production during strawberry ripening.
