Leaf explants were cultured to evaluate the effect of different auxins and cytokinins and its concentrations; MS salts on micropropagation of the promising biodiesel Jatropha curcas plant under Egyptian conditions. Results showed that shoot initiated on 0.5 mg/l BA + 0.25 mg/l IBA. Multiplication and elongation were found to be the best using 0.5 mg/l BA in combination with 0.05 mg/l IBA. The multiple shoots were cultured on MS or half strength of MS supplemented with different concentrations of IAA and IBA for rooting phase. Half strength of MS containing 1.0 mg/l IAA was the best for rooting of micropropagated shoots. The rooted plantlets were acclimated in sand : peat?moss mixture (1 : 1) successfully.Plant Tissue Cult. & Biotech. 26(1): 85-96, 2016 (June)

Lulo is a species of great importance to the fruticulture of Colombia, but has significant phytosanitary problems that require an aggressive breeding program oriented toward the production of genotypes with tolerance to phytopathogens. These programs need to establish highly efficient mass plant propagation protocols, such as somatic embryogenesis. This study focused on research on the somatic embryogenesis of lulo using kinetin, naphthalene acetic acid-NAA (Plant Growth Regulators, PGRs), and different sucrose concentrations in a MS medium. Two lulo varieties, Solanum quitoense var. septentrionale and S. quitoense var. quitoense, and two explant types (hypocotyl and cotyledon) were used, incubated in dark conditions at 25±2°C. The highest production percentage of the embryos was obtained when 50 mM of NAA were added to the medium with sucrose (50.0 and 263.1 mM) for the two explant types used. In lulo with spines, the highest percentage of embryonic structures (50%) was observed with cotyledonary leaf explants and 50 mM of NAA ; while in the spineless lulo, the embryonic structures were observed in the same type of explant with 50 mM of NAA + 263.1 mM of sucrose (32%).

In vitro: regeneration of Chenopodium quinoa Willd. was achieved from cotyledonary nodes explants. In this study, used 6-Benzylaminopurine (BAP) and α-Naphthalene Acetic Acid (NAA) of plant growth regulators with different concentrations individually as well as in combination and used different concentrations of sugar (sucrose) with different concentrations. For was rooting, used half strength (½MS), full-strength MS and ½ MS supplemented with 0.2 mg/l of NAA. The results mentioned, explant responding (%) to multiplication was about 73% for all BAP treatments compared with control and average numbers of shoot increased with increased BAP concentration except 5 mg/l of BAP. The highest explant responding (%) was in media supplemented BAP without NAA compared other treatments noted that the media with combination of BAP and NAA gives formation of callus in bases of the plantlets. Also, the result inducted the combinations between (BAP–NAA) was highly significantly (P≤ 0.001) and less effective on number of shoots where the highest number of shoot was 3.40 in media with 3 mg/l BAP compared other treatments. The highest of explant responding 93.33% was in media supplement with 10 g/l sucrose and (10 g/l sucrose + 3 mg/l BAP), but sucrose level for good greening and developed shoots (4 shoots) was in medium supplement with 10 g/l sucrose. The shoots rooted well on half-strength MS medium with 60% percentage of root. The rooted shoots were acclimatized and transferred to green house to follow their development.

A leaching frame was constructed to detect residual plant growth regulators in media. The table was 0.9 × 1.8 m and designed to hold 40 10-cm diameter by 30-cm PVC cylinders. Each cylinder was cut lengthwise in half and resealed with duct tape. Rooted cuttings of `Freedom' poinsettias were planted into each cylinder using two media combinations: 2 vermiculite: 2 peat moss: 1 pine bark and 2 vermiculite: 1 peat moss: 2 pine bark (by volume). Four growth regulator treatments were applied to the medium two weeks after transplanting: control, 0.25 mg paclobutrazol, 0.25 mg uniconazole, and 0.125 mg paclobutrazol applied as spike. After plant growth was recorded, the cylinders were removed and sliced lengthwise. Snapdragon plugs were then transplanted into the medium along the length of the cylinder to determine if any residual paclobutrazol remained. Paclobutrazol and uniconazole reduced stem length. The presence of pine bark in the media reduced the effect of the plant growth regulators.

<p class="IsiAbstrakIndo"><em>Justicia gendarussa </em>Burm.f., a medicinal plant, is Acanthaceae that has many functions. Furthermore, the compounds in gendarussa must be produced in high quantity and quality by applying callus culture method. Accordingly, it is important to study the effects of plant growth regulators (2,4-D, IBA, and BAP) on callus induction of gendarussa leaves. This research design utilized a factorial design with two factors (2,4-D and IBA: 0.5, 1, 1.5 mg/L and BAP: 0.5, 1, 1.5, 2 mg/L). The experiment consisted of 24 treatments, each of which was repeated 3 times. Observation was carried out in 6 weeks. Data on the time of callus formation, percentage of explants formed callus, and callus morphology were analyzed descriptively,while data on fresh and dry weight were analyzed by Two-Way ANOVA (<span>α</span> = 0.5). Interestingly, the results showed that various concentration of plant growth regulators (2,4-D, IBA, and BAP) affected callus induction from leaf explants of gendarussa. We concluded that the most optimal treatment combination of concentration of plant growth regulators in inducing callus from leaf explants of gendarussa is 1.5 mg/L 2,4-D and 2 mg/L BAP with a relatively long period of callus formation at the earliest, i.e. on day 5, 2.247 g of fresh weight, 0.108 gof dry weight, white callus translucent, and friable. <span lang="EN-GB">Moreover, t</span>he optimum treatment will be used to produce secondary metabolite and seed s<span lang="EN-GB">y</span>nthetic by cell suspension culture.</p>

Kohlrabi (Brassica oleracea var. gongylodes), with its edible stem tuber formed at the base of the plant stem, presents a valuable source of nutrients. The potential effects of plant growth regulators (PGRs), as well as various concentrations of different sugars on the in vitro development of kohlrabi were studied. Ten-day-old kohlrabi seedlings were cultivated in vitro for 5 weeks at 18?2?C on half-strength MS media containing different concentrations of carbon source such as sucrose, fructose, glucose, xylose and mannitol, combined with or without specific plant growth regulators (N6-benzyladenine (BA), gibberellic acid (GA3), 2,3,5-triiodobenzoic acid (TIBA)). Results showed no tuber formation in all treatments, but growth and development of treated kohlrabi seedlings was significantly affected in a distinctive manner, with a variety of morphological traits being altered in comparison to matching controls.

Three experiments were conducted to evaluate the effect of different potting media and horticultural practices on the infectivity of Steinernema carpocapsae (Weiser). Bioassays were conducted with S. carpocapsae added to potting media containing Galleria mellonella (L.) larvae. Potting media tested were aged pine bark, new pine bark, peat moss, aged cow manure, and a peat/vermiculite soilless medium (Pro-Gro 200). The horticultural practices tested were potting media pH level, fertilization, salt level, and application of plant growth regulators. The plant growth regulators ancymidol, paclobutrazol, chlormequat, and daminozide were each applied as a medium drench. The LC50s of the nematodes against G. mellonella, in potting media with different horticultural practices, were all between 0.50 and 2.06 with no significant difference in infectivity. Nematodes could be used for insect control with no negative effects under these growing conditions.
