AbstractFrom a screen of pupal lethal lines of Drosophila melanogaster we identified a mutant strain that displayed a reproducible reduction in the larval response to light. Moreover, this mutant strain showed defects in the development of the adult visual system and failure to undergo behavioral changes characteristic of the wandering stage. The foraging third instar larvae remained in the food substrate for a prolonged period and died at or just before pupariation. Using a new assay for individual larval photobehavior we determined that the lack of response to light in these mutants was due to a primary deficit in locomotion. The mutation responsible for these phenotypes was mapped to the lethal complementation group l(2)34Dc, which we renamed tamas (translated from Sanskrit as “dark inertia”). Sequencing of mutant alleles demonstrated that tamas codes for the mitochondrial DNA polymerase catalytic subunit (DNApol-γ125).

Königinnen von 12 Taxa der Untergattung Bombus sensu stricto (Bombus affinis, B. albocinctus, B. cryptarum, B. lucorum, B. magnus, B. moderatus, B. occidentalis, B. patagiatus, B. sporadicus, B. terrestris, B. terricola and B. tunicatus) wurden im Frühjahr gefangen, um künstliche Kolonien zu züchten. Zusätzlich wurden Männchen von B. franklini und B. lucorum/China gefangen. Mitochondriale Cytochrome Oxidase Untereinheit I (COI) von 53 Proben wurde sequenziert (Teilsequenzen der Länge 1257 bp). GenBank Sequenzen der Ostasiatischen Arten B. hypocrita, B. ignitus und B. lucorum/China wurden einbezogen. Der Unterschied zwischen den Arten beträgt 30 – 92 Basen-Substitutionen und die Tamura-Nei genetische Distanz 0.030-0.103, während der Unterschied innerhalb der Arten nur 1 – 3 Basensubstitution beträgt und die Tamura-Nei genetische Distanz nur 0.001-0.003. Dreizehn Taxa mit Artrang sowie eine neue Art B. minshanicola Bischoff 1936 (= B. terrestris ssp. minshanicola = B. lucorum/China) wurden nachgewiesen. In der Topologie des phylogenetischen Stammbaums gibt es 4 Cluster: ein Artenpaar B. sporadicus – B. ignitus und ein Artenpaar B. terricola – B. occidentalis, ein Cluster für B. magnus, B. patagiatus and B. cryptarum mit den Subspecies B. cryptarum albocinctus und B. cryptarum moderatus sowie ein Cluster für B. hypocrita, B. lucorum, B. minshanicola, B. franklini and B. affinis. Die Arten B. terrestris mit den Subspecies B. terrestris canariensis und B. terrestris sassaricus sowie B. tunicatus sind einzeln abgetrennt. Da sich im Alignment der COI Sequenzen keine Lücken finden, können die einzelnen Nucleotid-Positionen als homolog betrachtet werden. Jede Art besitzt eine Reihe einzigartiger (‘privater’) Positionen, die als diagnostische Merkmale benutzt werden können um die Art zu definieren und zu identifizieren.

AbstractThe large-clawed scorpion, Scorpio maurus, is a medically important scorpion and yet nothing is known regarding genetic diversity within this species. As a preliminary analysis we determined variation within the cytochrome oxidase 1 (CO1) mitochondrial gene from specimens from Morocco. High levels of genetic diversity were found that presented some geographical coherence. Of the two identified subspecies from Morocco, S. maurus birulai and S. maurus fuliginosus, the latter included genetically distinct lineages (8.0% uncorrected sequence divergence), indicating a detailed morphological and molecular revision is needed for this species.

The phylogeny of Rhyacophilidae was explored with 28S ribosomal RNA (rRNA) and Cytochrome Oxidase Subunit I (COI) mitochondrial DNA (mtDNA). Eighty one rhyacophilids were included in the analysis. We found that although Rhyacophilidae was recovered as monophyletic, intrafamilial relationships are not well-resolved using this dataset. Bootstrap support was poor for intrageneric relationships and additional data will be required to present a more robust hypothesis. The recovered phylogeny places Fansipangana as the sister taxon of the rest of Rhyacophilidae. We found that Himalopsyche was nested inside the genus Rhyacophila with the verrula group sister to Himalopsyche and remaining Rhyacophila. These results and possible relationships should be tested with a more extensive data set.

Undescribed females representing four morphological types were found in a collection of adult Diamesa from about 5000 m altitude in Rongbuk, Tibet. Short DNA sequences of cytochrome oxidase subunit 2 were used to associate two single males in the material with conspecific females. Diamesa solhoyi n.sp. and Diamesa aculeata n.sp. are described. The complete type material and additional specimens have been deposited in the Insect Collection at the Institute of Zoology, Academia Sinica, Beijing (IZAS). The sequences are deposited in Genbank with accession numbers AM051227–AM051233.
