SummaryPoltava chickens are native to Ukraine, with a remarkable history, genetics and economic traits. They include three varieties: Clay, Cuckoo and Black. The Poltava Clay variety exemplifies what can be achieved by thorough selection in transforming a local, low production chicken population into an established breed with high performance characteristics and selected line structure. Two other varieties, the Cuckoo and Black, are extinct but plans for their restoration are underway.

Opium poppy (Papaver somniferum L.) is an important medicinal plant produces more than 80 alkaloids obtained from the capsules and straw of the plant. The estimate of combining ability gives an indication of the genetic behaviour of the parental material. It is therefore, desirable to select the parents for hybridization on the basis of their per se performance and combining ability effects. The F1, F2 and their reciprocals of an eight parent diallel cross in opium poppy were studied for combining ability of seven economic traits. The mean sum of squares due to GCA, SCA and reciprocals were significant for all the traits. The magnitude of GCA variances were invariably higher than those of SCA and thus indicating the preponderance of non-additive genetic variances, which was further affirmed by the measure of average degree of dominance i.e. ?(?2s/? 2g).  Among the parents VN35I for plant height, Sanchita and VG26 for capsules per plant, VN23, VN35I and Vivek for capsule index, VG26 and Sanchita for seed and straw yield per plant, and VN35I and VG20 for morphine content were found good general combiners. Earliness being a desired trait, parent VG20 having significant negative GCA estimates coupled with per se performance can be considered as good general combiner for early flowering. Inclusion of good general combiners in a multiple crossing program or an inter-mating population involving all possible crosses among them subjected to bi-parental mating may be expected to offer maximum promise in breeding for economic traits.DOI: http://dx.doi.org/10.3329/bjpbg.v24i2.17000

The main goal of the current study was cloning and expression of the human insulin gene in Pichia pastoris expression system, using genetic engineering techniques and its treatment application. Total RNA was purified from fresh normal human pancreatic tissue. RNA of good quality was chosen to obtain a first single strand cDNA. Human preproinsulin gene was amplified from cDNA strand, by using two sets of specific primers contain EcoR1 and Notl restriction sites. The amplified preproinsulin gene fragment was double digested with EcoRI and Not 1 restriction enzymes, then inserted into pPIC9K expression vector. The new pPIC9K-hpi constructive expression vector was transformed by the heat-shock method into the E.coli DH5α competent cells. pPic9k –hpi, which was propagated in the positive transformant E. coli cells, was isolated from cells and then linearised by restriction enzyme SalI, then transformed into Pichia pastoris GS115 using electroporation method. Genomic DNA of His+ transformants cell was extracted and used as a template for PCR analysis. The results showed, that the pPic9k – hpi was successfully integrated into the P. pastoris genome, for selected His+ transformants clones on the anticipated band at 330 bp, which is corresponded to the theoretical molecular size of the human insulin gene. To follow the insulin expression in transformans, Tricine–SDS gel electrophoresis and Western blot analysis were conducted. The results showed a successful expression of recombinant protein was detected by the presence of a single major band with about (5.8 KDa) on the gel. These bands correspond well with the size of human insulin with the theoretical molecular weight (5.8 KDa).
