This paper addresses the determination of aldehyde group (% of dialdehyde units) in dialdehyde cellulose (DACs) oxidized from bacterial cellulose (BC). With 4 determination methods, the oxidation degrees of DACs were studied: Automatic potential titration method through Schiff base reaction, Acid-base titration in Cannizzaro reaction, Metaperiodate consumption determination, and Measurement of amino nitrogen content in DAC derivatives. By comparing determination results of these methods, we found out the proper way suitable for this metaperiodate oxidation system of BC to measure the dialdehyde unit content of the DACs.

AbstrakTahu merupakan makanan yang digemari oleh masyarakat.Tahu mempunyai daya tahan sekitar 1 - 2 hari sehingga pedagang sering menambahkan formalin sebagai pengawet. Formalin merupakan bahan pengawet yang dilarang oleh pemerintah yang penggunaannya masih terdapat secara luas di masyarakat dan bila dilihat dari tekstur tahu yang dijual di pasar kota Padang, dicurigai tahu memiliki kandungan formalin.Tujuan penelitian ini adalah untuk mengetahui perbedaan kadar formalin pada tahu yang dijual di pasar pusat kota dengan pinggiran kota Padang. Penelitian ini dilakukan di laboratorium Balai Riset dan Standardisasi Industri Padang.Jenis penelitian ini adalah analitik yang telah dilaksanakan pada bulan Juni-September 2013. Jumlah sampel adalah sebanyak 36 buah yang terdiri dari 18 sampel tahu yang berasal dari pasar pusat kota dan 18 sampel tahu yang berasal dari pasar pinggiran kota Padang. Uji kualitatif formalin pada tahu dilakukan dengan metode asam kromatropat dan uji kuantitatif formalin menggunakan metode titrasi asam basa. Analisis data dilakukan secara bivariat dengan menggunakan uji t. Hasil penelitian didapatkan kadar formalin pada tahu di pasar pusat kota Padang dari 18 sampel yang diperiksa terdapat 17 sampel yang positif formalin dengan kadar paling tinggi adalah 3.65%. Kadar formalin pada tahu di pasar pinggiran kota Padang dari 18 sampel yang diperiksa terdapat 17 sampel yang positif formalin dengan kadar paling tinggi adalah 2.73%. Rata-rata kadar formalin pada pasar pusat kota adalah 1.08% dan pasar pinggiran kota adalah 0.67%.Kata Kunci: kadar formalin, tahu, pasar pusat kota Padang, pasar pinggiran kota PadangAbstractTofu is a favorite food among the community. Tofu has resistance 1 - 2 days so that merchant often add formalin as a preservative. Formalin is a preservative which is banned by the government that there is still widespread use in the community and the texture of tofu sold in the market is suspected for having formaldehyde content. The purpose of this study was to difference the levels of formaldehyde in tofu sold in downtown market and suburban market of Padang. This research was conducted in the laboratory Industry Research and Standardization Padang. This research is an analytic that has been conducted in June-September 2013. The number of samples of this study are 36 units consisting of 18 pieces of tofu samples derived from downtown market and 18 pieces of tofu samples derived from suburban market of Padang. Qualitative test performed with chromotropic acid method and quantitative formalin test using acid-base titration method. Bivariate data were analyzed using t test. The results showed that the levels of formaldehyde found in tofu sold in downtown market of Padang is from 18 samples tested there were 17 positive samples with the highest levels of formaldehyde was 3.65%. Levels of formaldehyde found in tofu sold in suburban market of Padang is from 18 samples tested there were 17 positive samples with the highest levels of formaldehyde was 2.73%. One of the uses of acid-base indicators is to show the end point of the titration, so the accurate determination of acidity constant and pH range of indicators needs to be done. This study aims to determine the acidity constant (pKa) of mangosteen pericarp extract and its accuracy as an indicator of acid-base titration. Determination of pKa was done by a simple potentiometric titration method. The titration data were plotted in three graphs, i.e., pH, ΔpH/ΔV (the (the first derivative), and Δ2pH/ΔV2 (the second derivative) versus titrant volume to determine the equivalence point of the titration. The accuracy test was carried out by comparing the volume of oxalic acid used to titrate NaOH solution using the indicator of mangosteen pericarp extract and phenolphthalein indicator. The result showed that the equivalence point was found on the titrant volume of 8.6 mL and a measured pH of 9.84. so the pKa value of mangosteen pericarp extract was 7.20, and the pH range was 6.20 to 8.20. the average volume of oxalic acid used to titrate 5 mL of NaOH using phenolphthalein as the indicator was 5.2 mL while the titration used mangosteen pericarp extract was 5.23 mL. The accuracy of mangosteen pericarp extract was 99.42%. By the result, it can be concluded that potentiometric titration can be used as a simple way to determine the acidity constant of mangosteen pericarp extract. Moreover, the mangosteen pericarp extract can be used as an alternative acid-base titration indicator to substitute the common acid-base titration in the laboratory.
