The aim of this article is to quantify the loads exerted by heavy-duty vehicles when crossing over buried pipeline. This problem arises in connection to the question pertaining to the use of protective sleeves (casings) applied to gas pipelines in regions with increased demands on pipeline operation safety. An experiment was conducted on a test pipe section made from L360NE pipeline steel equipped with strain gauges along the pipe perimeter, measuring strains in the axial and circumferential directions. Strain measurements were taken after back-filling the pipe trench, then during vehicle crossings over the empty pipe, and again after pressurizing the test pipe with air. Strain-based hoop stresses at the surface of the empty test pipe were found to exceed 30 MPa after back-filling the trench and increased to more than 40 MPa during the vehicle crossings. Similarly, axial stresses reached extremes of around 17 MPa in compression and 12 MPa in tension. Applying internal air pressure to the test pipe resulted in a reduced net effect on both the hoop and axial stresses.

This study explored the order of acquisition of various types of syntactic-movement and embedding structures in Hebrew, using a sentence-repetition task, in which 60 children aged 2;2-3;10 repeated 80 sentences (with a total of 4800 sentences), and an analysis of the spontaneous speech of 61 children aged 1;6-6;1 (27,696 clauses). The sentence repetition task revealed a set order of acquisition of the various types of syntactic movement: A-movement is acquired first, then A-bar-movement, and finally movement of the verb to C. The analysis of spontaneous speech revealed the same order: A-movement of the object of unaccusative verbs to subject position appears first, together with simple SV sentences; then, wh-questions appear, then relative clauses and topicalization, which appear together with embedding of finite clauses, and lastly, V-to-C movement. Previous studies have shown that Hebrew speakers under age six have difficulty comprehending and producing sentences with A-bar-movement in which a lexically-restricted object crosses over a lexically-restricted subject. And indeed, whereas children produced A-bar structures very early (wh-questions from age 1;6, relative-clauses and topicalization from age 2;6), until age 5;8 these structures never included a lexical DP crossing over another lexical DP. Both tasks indicated that the order of structure acquisition is fixed, creating Guttman scales between structures, but different children acquire the same structure at very different ages. It seems that whereas the syntactic path and the stages of structure acquisition along it are constant between children, each child walks this path in their own pace.

AbstractThe synaptonemal complex (SC) is a supramolecular protein scaffold that mediates chromosome synapsis and facilitates crossing over during meiosis. In mammals, SC proteins are generally assumed to have no other function. Here, we show that SC protein TEX12 also localises to centrosomes during meiosis independently of chromosome synapsis. In somatic cells, ectopically expressed TEX12 similarly localises to centrosomes, where it is associated with centrosome amplification, a pathology correlated with cancer development. Indeed, TEX12 is identified as a cancer-testis antigen and proliferation of some cancer cells is TEX12-dependent. Moreover, somatic expression of TEX12 is aberrantly activated via retinoic acid signalling, which is commonly disregulated in cancer. Structure-function analysis reveals that phosphorylation of TEX12 on tyrosine 48 is important for centrosome amplification but not for recruitment of TEX12 to centrosomes. We conclude that TEX12 normally localises to meiotic centrosomes, but its misexpression in somatic cells can contribute to pathological amplification and dysfunction of centrosomes in cancers.

Abstract Bridge weight-in-motion (BWIM) system is a method, that provides to identify axle weights. It is a non-destructive method, which allows not only to identify the axle weight, but it can show current shape of the structure, so it has a great potential. There are various methods to do measurements for this system. Mostly, accelerometers or strain gauge are used. Signal noise has significant effects to the results. It could be resonance of the bridge, wind, defect at the support system, defect at the roadway, etc. It is necessary to filter all this effects, to get clear data. There are many ways to do the filtering. Digital filters allow it. Sometimes, this type of filtering could remove important data about the crossing of the vehicle. It could generate inaccuracy of the whole system and create major errors to identified vehicles. It is necessary to find the optimal way, to keep important data and remove all dynamic noise. This paper will investigate the previously mentioned problems. Measurements will be accomplished on a small-scale model of the bridge. Vehicle will be crossing over the bridge, while the bridge will be awakened to the first vibration shape and other frequencies, that will have a great impact to the measurements.

<p>This thesis explores the dynamic and oftentimes puzzling relationship between singer, song, and listener in the music of contemporary singer-songwriters. I argue that this relationship is simultaneously understood through and confused by the use of indeterminate pronouns in song lyrics. The crossing over of language from speech to song leads to a multiplicity of “readings” of music and artists, where the “I” of the song’s narrator is elided with the “I” of the composer and the “I” of the performer. In this research I highlight the importance of listener experience in the ways these “I”s are contextualized and examined, shedding light on the dynamic and varied ways listeners are already engaging with this music. I also explore how these understandings ultimately impact listeners’ broader perceptions of the often-gendered values of emotional honesty and truthfulness of singer-songwriters.  This research brings together original ethnographic research and theoretical explorations of musical and literary authorship and interpretation, focusing on the creative work of three singer-songwriters – Amanda Palmer, Hera Hjartardóttir, and Katie Morton. By drawing on these diverse research areas and musical repertoires, I propose an interdisciplinary model that highlights the active role listeners play in the creation (or discovery) of musical meaning. My hope is that this thesis opens up a space for future discussions to take place that examine the many layers of live performance that impact and inform interpretations of narrative, while also giving a voice to the listeners and fans whose engagement forms a crucial element of these singer-songwriter performances.</p>

Plant breeding relies on the meiotic recombination or crossing over to generate the new combinations of the alleles along and among the chromosomes. However, crossing over is constrained in the crops such as barley by a combination of the low frequency and biased distribution. In this study, we attempted to identify the genes that limit the recombination by performing a suppressor screen for the restoration of fertility to the semi-fertile barley mutant desynaptic10 (des10), carrying a mutation in the barley ortholog of MutL-Homolog 3 (HvMLH3), a member of the MutL-homolog (MLH) family of DNA mismatch repair genes. des10 mutants exhibit reduced recombination and fewer chiasmata, resulting in the loss of obligate crossovers (COs) leading to chromosome mis-segregation. We identified several candidate suppressor lines and confirmed their restored fertility in an Hvmlh3 background in the subsequent generations. We focus on one of the candidate suppressor lines, SuppLine2099, which showed the most complete restoration of fertility. We characterized this line by using a target-sequence enrichment and sequencing (TENSEQ) capture array representing barley orthologs of 46 meiotic genes. We found that SuppLine2099 contained a C/T change in the anti-CO gene RecQ-like helicase 4 (RECQL4) resulting in the substitution of a non-polar glycine to a polar aspartic acid (G700D) amino acid in the conserved helicase domain. Single nucleotide polymorphism (SNP) genotyping of F3 populations revealed a significant increase in the recombination frequency in lines with Hvrecql4 in the Hvmlh3 background that was associated with the restoration of fertility. The genotyping also indicated that there was nearly double the recombination levels in homozygous Hvrecql4 lines compared to the wild type (WT). However, we did not observe any significant change in the distribution of CO events. Our results confirm the anti-CO role of RECQL4 in a large genome cereal and establish the possibility of testing the utility of increasing recombination in the context of traditional crop improvement.
