Abstract The impact of the brominated flame-retardant mixture, DE-71, on gonadal steroidogenesis during sexual differentiation in Silurana tropicalis was examined. A partial lifecycle study exposing S. tropicalis to varying concentrations of DE-71 (0.0, 0.65, 1.3, 2.5, and 5.0 μg/l [nominal]) was conducted from early gastrula-stage embryo to 150 days postmetamorphosis (dpm). Exposure of S. tropicalis to DE-71 induced liver necrosis and induced abnormal ovary development characterized by previtellogenic oocyte necrosis and arrested development of vitellogenic oocytes in females in a concentration-dependent manner. Decreased mean plasma dihydrotestosterone (DHT) and T, gonad T, and increased mean plasma E2 levels were found in 150 dpm DE-71-treated male S. tropicalis compared to controls. Plasma E2 levels in females were not significantly altered compared to control S. tropicalis, although lower plasma and gonad T were detected. Mean gonadal CYP 19 aromatase activity in both male and female S. tropicalis exposed to DE-71 was not appreciably affected. Decreased mean male 5α-reductase and CYP17 activities in both male and females were observed compared to control frogs. Overall, these studies suggested that PBDE exposure induced liver necrosis and abnormal ovary development; and reduced circulating and gonadal androgens resulting in a phenotypic skew in sex ratio toward the female sex in S. tropicalis.

ABSTRACT Macrobrachium amazonicum (Heller, 1862) is the most important species for aquaculture native to South America. This study evaluates the phenotypic plasticity of females of Macrobrachium amazonicum with respect to the gonadal development, and determines the spawning type. Our study consisted of macro and microscopic analysis of the reproductive tract. Ovaries exhibited the following reproductive cells in developmental sequence: oogonia (OO) - mean diameter and standard deviation of 25.4 ± 6.5 μm; previtellogenic oocyte (PVO) - 61.7 ± 10.7 μm, vitellogenic oocyte (VO) - 113.9 ± 24.5 μm; and mature oocyte (MO) - 308.7 ± 56.3 μm. Ovaries increased in volume due to vitellogenesis and changing of basophilic to acidophilic composition. Follicular cells, atresic oocytes and postovulatory follicles were also analyzed. By combining macro and microscopic analysis, the ovaries of M. amazonicum were classified into six stages: Immature, In maturation, Mature, Spawned in maturation, Spawned and Resting. The ovarian development of M. amazonicum followed a standard pattern even among different populations. Considering our results and the evidence in literature, we conclude that spawning for this species is parceled or multiple, that is, synchronous in more than two groups (clutches of oocytes). Such observations provide basis for further studies addressing sustainable management strategies for species conservation and contribute to elucidate the biology of their specimens.

Previtellogenic ovarian fragments from eel,Anguilla australis, were culturedin vitroin a chemically defined medium containing steroids and/or peptide hormones for 18 days in order to investigate their involvement in control of early oocyte growth. 11-Ketotestosterone (11-KT), but not estradiol-17β, induced a significant 10–20% increase in diameters of previtellogenic oocytes and oocyte nuclei in a dose-dependent manner. Effects were greatest for 100 nM 11-KT, a dose that is within the physiological range seen in very early vitellogenic eels in the wild. The effect was not accompanied by obvious ultrastructural changes in the oocytes other than an apparent increase in nuclear size. Similarly, treatment with recombinant human IGF-I resulted in increased oocyte diameters, whereas no such effect was seen after treatment with heterologous insulin, GH, leptin, or human chorionic gonadotropin. Interestingly, lipid supplementation also resulted in an increase in oocyte diameter, and greater radioactivity in ovarian explants following incubation with14C-triglycerides and 11-KT, but not FSH, suggesting that the androgen may play a role in lipid accumulation into the oocyte. Our results implicate hormones from both the reproductive and the metabolic axes in control of previtellogenic oocyte growth in a teleost fish.
