Over the past decade, social policy in Ireland has taken an increasingly ‘workfarist turn’. This has proceeded through benefit cuts, tighter eligibility criteria for payments, and claimant activation via penalty rates for breaching new conduct conditions. However, key to understanding the post-crisis reconfiguration of welfare is not just the increasingly workfarist content of social policy but also how the delivery of public employment services has been reorganised through processes of marketisation and tightening performance management of delivery organisations and the staff who work within them. Positioning these governance reforms as processes of ‘double activation’, and drawing on survey and interview research with frontline staff working for agencies contracted by government to deliver activation, this study explores how frontline staff experience performance management as a disciplinary regime: the degree to which frontline workers are subject to management control and performance management in their jobs, what forms this takes, and how it shapes their field of action and choice. In so doing, the study draws attention to the ways in which the governance of caseworkers and the governance of claimants are inter-related, and the degree to which performance management regimes influence frontline practices to motivate the enforcement of workfarist policy practices.

The Lewis acid-catalyzed Friedel-Crafts alkylation of an aromatic ring with an alkyl halide is extensively used in organic synthesis. However, its biological counterpart was not reported until the elucidation of the cylindrocyclophane biosynthetic pathway in Cylindrospermum licheniforme ATCC 29412 by Balskus and co-workers. CylK is the key enzyme to catalyze the formation of the cylindrocyclophane scaffold through the Friedel-Crafts alkylation reactions with regioselectivity and stereospecificity. Further research demonstrates that CylK can accept other resorcinol rings and secondary alkyl halides as substrates. To date, the crystal structure of CylK has not been disclosed and the catalytic mechanism remains obscure. Herein we report the crystal structures of CylK in its apo form and its complexes with the analogues of its substrate and reaction intermediate. Combining the crystal structures, free energy simulations and the mutagenesis experiments, we proposed a concerted double-activation mechanism, which could explain the regioselectivity and stereospecificity. This work provides a foundation for engineering CylK as a biocatalyst to expand its substrate scope and applications in organic synthesis.

Understanding the dynamical hallmarks of network motifs is one of the fundamental aspects of systems biology. Positive feedback loops constituting one or two nodes, self-activation, toggle switch, and double activation loops, are commonly observed motifs in regulatory networks underlying cell-fate decision systems. Their individual dynamics are well-studied; they are capable of exhibiting bistability. However, studies across various biological systems suggest that such positive feedback loops are interconnected with one another, and design principles of coupled bistable motifs remain unclear. We wanted to ask what happens to bistability or multistability traits and the phenotypic space (collection of phenotypes exhibited by a system) due to the couplings. In this study, we explore a set of such interactions using discrete and continuous simulation methods. Our results suggest that couplings that do not connect the bistable switches in a way that contradicts the connections within individual bistable switches lead to a steady state space that is strictly a subset of the set of possible combinations of steady states of bistable switches. Furthermore, adding direct and indirect self-activations to these coupled networks can increase the frequency of multistability. Thus, our observations reveal specific dynamical traits exhibited by various coupled bistable motifs.

AbstractThe aim of this study was to analyse the high density EEG during movement execution guided by visual attention to reveal the detailed topographic distributions of delta and theta oscillations. Twenty right-handed young subjects performed a finger tapping task, paced by a continuously transited repeating visual stimuli. Baseline corrected power of scalp current density transformed EEG was statistically assessed with cluster-based permutation testing. Delta and theta activities revealed differences in their spatial properties at the time of finger tapping execution. Theta synchronization showed a contralateral double activation in the parietal and fronto-central regions, while delta activity appeared in the central contralateral channels. Differences in the spatiotemporal topography between delta and theta activity in the course of movement execution were identified on high density EEG.

Abstract Since the financial crisis, Ireland’s welfare state has been reorientated around a regulatory, ‘work-first’ activation model. Claimants now face penalty rates for non-compliance with activation requirements that have been significantly extended since 2009. Alongside these formal policy reforms, the organisations delivering Public Employment Services, and the modes by which they are commissioned, have also been reconfigured through a series of New Public Management style governance reforms, including, most notably, the creation of a quasi-market for employment services (JobPath) in 2015. This article addresses the intersection between activation and quasi-marketisation, positioning the latter as a form of ‘double activation’ that reshapes not only how but also what policies are enacted at the street level. It unpacks their shared logics and mutual commitment to governing agents at a distance through a behavioural public policy orientation, and reflects on the extent to which marketisation is capable of producing lower-cost but more responsive employment services.
